Cultivation of plant tissues in a contained environment, such as plant tissue culture in vitro , could effectively address safety issues associated with transgenic plants as well as strict manufacturing standards. However, unfavourable in vitro conditions can often lead to elevated stress levels and suppressed growth of in vitro tissues urging the need to improve plant adaptation to in vitro conditions. As the beneficial effect of endophytic bacteria on the growth of in vitro cultures has been described previously, the aim of the present study was to assess the effect of growth-promoting endophytic bacteria isolates of the Bacillus cereus group on the growth of transgenic tobacco shoots in vitro and the accumulation of recombinant protein (RP) construct comprising a green fluorescent protein and a collagen mimicking peptide conjugate. The research revealed that transgenic tobacco shoots had reduced growth and increased oxidative injury levels compared with the non-transgenic parental line. The inoculated bacteria isolates colonised plant tissues and were sustained at densities of 0.6–9.0 × 10 5 CFU g -1 of the shoot fresh weight for at least several passages. Enhanced, 18% to 30% higher, biomass accumulation compared to the uninoculated transgenic tobacco shoots, and 14% to 25% lower levels of the oxidative lipid injury marker malondialdehyde (MDA) were detected for three-week-old shoots co-cultivated with isolates of Bacillus mycoides Nt.10.1, B. toyonensis Nt.18 and B. wiedmannii Nt.3.2. B. wiedmannii isolate Nt.14.2 reduced oxidative stress injury symptoms but had no effect on shoot growth. Co-cultivation with endophytic isolates had no effect on the expression of the recombinant protein. {"title":"Short-term effect of heavy precipitation on nutrient leaching from arable sandy loam soil amended with fertiliser and biochar","authors":"Tatiana V. Abramova, Natalya P. Buchkina","doi":"10.13080/z-a.2022.109.002","DOIUrl":"https://doi.org/10.13080/z-a.2022.109.002","url":null,"abstract":"The purpose of this study was to assess the effect of biochar application on the leaching of the main nutrients from the topsoil of the arable sandy loam soil (Albic Luvisol) of North-Western Russia after application of mineral fertiliser in the conditions of heavy precipitation. A short-term laboratory experiment included 6 treatments: (1) without fertiliser and biochar (control), (2–3) soil + biochar (10 and 20 t ha-1), (4) soil + fertiliser, and (5–6) soil + biochar (10 and 20 t ha-1) + fertiliser. During the experiment, the soil in the cylinders was watered three times with high rates (345 cm3) of distilled water, corresponding to the maximum amount of daily precipitation (44 mm) observed in the natural conditions of the area. The leachate was collected (on the 1st, 2nd and 5th day of the experiment) and tested for pHH2O, available nitrogen (N-NO3 −, N-NH4 +), phosphorus (P2O5) and potassium (K2O). At the end of the experiment, separate soil samples were formed of the upper 8-cm and lower 4-cm soil layers in the cylinders. The pHH2O values, concentrations of N-NO3 −, N-NH4 +, P2O5 and K2O were determined in the samples. The results showed that the effect of biochar on the movement and accumulation of the main available nutrients in the soil after fertiliser application and under the effect of heavy precipitation was more pronounced at the higher biochar application rate of 20 t ha-1. Biochar application reduced the acidity of the leachate and the soil not only in the layer, where the biochar had been applied, but also in the lower layer affected by the leachate. Available NNH4 +, P2O5, and K2O were retained in the soil through biochar application for a short period of time, while N-NO3 − leaching was not affected. This study was aimed to evaluate the effect of mammalian sex hormones (progesterone and β-estradiol), abscisic acid (ABA), and salicylic acid (SA) on plant growth, chlorophyll, malondialdehyde (MDA), hydrogen peroxide (H2O2) and the activities of anti-oxidative and nitrate assimilation enzymes in common bean (Phaseolus vulgaris L.) seedlings under cold stress. For this purpose, cold stress was conducted to 11-day-old seedlings in a climate chamber at 9/5°C (day/night), but for control, the temperature was set at 25/20°C (day/night). Progesterone (Pro) 10-9 M, βoestradiol (ES) 10-6 M, ABA 10-4 M, and SA 10-4 M were applied to 11-day-old seedlings using a handheld sprayer. The results of this experiment showed that cold stress significantly decreased plant growth, and it was followed by an increase in MDA and H2O2 content in common bean seedlings. However, the exogenous application of Pro, ES, ABA, and SA mitigated the adverse effect of cold stress in common bean seedlings. In this respect, cold stress decreased the activities of nitrate reductase (NR), nitrite reductase (NiR), and glutamine synthetase (GS) by 41.03%, 35.20%, and 47.54%, respectively, compared to control. However, under cold stress, Pro, ES, ABA, and SA treatments significantly increased NR, NiR, and GS activities by 41.87%, 14.80%, 33.87%, and 64.82 % for NR, 3.4%, 30.10%, 12.75%, and 33.12% for NiR, and 83.00%, 50.10%, 83.14%, and 9.34% for GS, respectively, compared to the plants treated with cold stress. Changes in plant diversity reflect the environmental conditions and practices of grasslands management. The aim of this study was to investigate the impact of abandonment on the grassland community and soil seed bank under the conditions of a hilly relief. The experiment was carried out at the Vėžaičiai Branch of the Lithuanian Research Centre for Agriculture and Forestry on the hilly topography of Žemaičiai Highland. The study analyses long-term monitoring data of a soil erosion experiment set up on the slopes of 9–11° steepness. The soil of the southern exposition slope was a slightly eroded Eutric Retisol. In 1993, to protect the hill from erosion, a mixture of five grasses consisting of 20% timothy grass (Phleum pratense L.), 20% red fescue (Festuca rubra L.), 20% meadow grass (Poa pratensis L.), 20% white clover (Trifolium repens L.), and 20% common bird’s-foot trefoil (Lotus corniculatus L.) was sown in different parts of the hill: the summit, the midslope, and the footslope. The grassland had not been fertilised and used. After 27 years, 56 vascular plant species were identified. In the summit, midslope, and footslope parts of the hill, the relative abundance (P%) of the sown species was 17.2%, 23.9%, and 27.2%, respectively. A good growth of Festuca rubra did not depend on abandonment. In all parts of the hill, P% of other sown species significantly decreased. The lowvalue plants (Elytrigia repens L., Cirsium arvense L., and Equisetum arvense L.) spread in grasslands. The soil seed bank of abandoned grasslands was composed of the arable weeds: Chenopodium album L. was the dominant species in the soil seed bank of the summit of the hill (0–15 cm depth), while Stellaria media (L.)
