{"title":"A new group of mitogenic lectins from invertebrate sources.","authors":"K Schumacher, G Uhlenbruck, B P Chatterjee, G Steinhausen, P Vaith, A van Mil","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In the haemolymph of the Tridacnid bivalve clams anti-galactans occur which do not have only glycosubstance precipitating and cell agglutinating properties, but also show mitogenic activity with respect to the blast transformation of human peripheral lymphocytes. This new property can be inhibited in a specific way by galactans and is destroyed by the degradation of the mitogenic lectin into subunits by polymer bound pronase. Quite a number of invertebrate haemolymph samples and snail albumin gland extracts, containing agglutinin activity, proved to have no mitogenic potency. The only exception was found in an extract of the snail albumin gland of Ampullaria canaliculata which contained a strong mitogen of the lectin type.</p>","PeriodicalId":23935,"journal":{"name":"Zeitschrift fur Immunitatsforschung. Mice immunized with various doses of LPS developed anti-LPS antibodies. Antibody activity was due to 19S and 7S immunoglobulins. Extracts rich in RNA from spleen cells of immune mice were able to transfer specific anti-LPS response to normal recipients. {"title":"Antibody dependent cellular cytotoxicity (ADCC) against human erythrocytes, mediated by blood group alloantibodies: a model for the role of antigen density in target cell lysis.","authors":"H Northoff, A Kluge, K Resch","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Antibody dependent cellular cytotoxicity (ADCC) of human mononuclear cells against human erythrocytes could be obtained with anti-A and anti-D sera. The degree of lysis varied considerably depending on the antigen system and on the experimental conditions. Anti-D mediated in contrast to anti-A mediated ADCC turned out to be very sensitive to conditions which interfere with target cell lysis: for most of the anti-D sera, removal of unbound IgG was found to be crucial to detect their ability to mediate ADCC. Pretreatment of the target cells with various enzymes dramatically improved specific lysis and left spontaneous release and spontaneous cytotoxicity essentially unaffected. In the case of neuraminidase treatment it could be shown that the effect was independent from the exposure of additional binding sites. When enzyme treatment and removal of excess IgG were applied in combination, as little as 10(4) antigenic determinants proved to be sufficient to induce specific lysis.</p>","PeriodicalId":23935,"journal":{"name":"Zeitschrift fur Immunitatsforschung. Besides the increase of the antigens A3, A11 and B7 and the decrease of A2, the large number of homozygous antigens at the A or/and at the B locus was found to be the most interesting result. The paper describes attempts to localize the site of Tg-AAb production in various organs of these animals using the immunofluorescence method.
