However, necrosis frequently occurs when the flap's aspect ratio exceeds 2:1. Promoting angiogenesis and regulating inflammation are essential for treating ischemia-reperfusion injury in random flaps. In this study, calcium-doped silica nanoparticles loaded with deferoxamine (CD) were created through physical adsorption by rapidly mixing biodegradable calcium-doped silica nanoparticles (CS) with deferoxamine (DFO). A gelatin methacryloyl (GM) hydrogel nanofiber scaffold containing 1% (w/v) CD (GM/CD-1) was subsequently produced using electrospinning technology. The GM/CD-1 scaffold showed excellent biocompatibility and significantly promoted flap regeneration in mice, achieving a 96.17 ± 3.17% flap survival rate at 14 days. Additionally, it effectively stimulated hair follicle growth and exhibited an inflammatory-modulating effect. We conducted a systematic search of the PubMed, Embase, Web of Science, EBSCO, Cochrane, ClinicalKey, Ovid Medline, Sinomed, CNKI, and WANFANG databases for studies assessing the efficacy of low-level laser therapy for diabetic foot ulcers, with a cutoff date of 15 September 2024. Two researchers independently selected relevant studies and extracted data, utilising RevMan 5.4 software to perform a meta-analysis. A total of 11 studies involving 657 participants with grade I-II diabetic foot ulcers were included. In patients with grade I-II diabetic foot ulcers, low-level laser therapy significantly increased the complete healing rate (RR = 4.97, 95% CI: 2.65-9.32, p < 0.00001), the wound shrinkage area (MD = 7.76, 95% CI: 6.51-9.0, p <0.00001), and the wound bacterial clearance rate (RR = 3.72, 95% CI: 2.16-6.39, p <0.00001) compared to the control group, while also reducing the infection rate (RR = 0.11, 95% CI: 0.01-0.8, p <0.03). Low-level laser therapy is an effective adjuvant therapy that accelerates wound healing in patients with grade I-II diabetic foot ulcers. While the crucial relationship between keloid formation and the vascular system has been highlighted, conflicting findings have been reported regarding the characterisation of keloid vasculature. Here, we successfully characterised the detailed three-dimensional (3D) structure of vasculature in keloid tissues using tissue clearing methods combined with 3D imaging. First, we compared two optical tissue clearing methods, the clear, unobstructed brain imaging cocktails and computational analysis and immunolabelling-enabled 3D imaging of solvent-cleared organ protocols, and found the latter to provide greater transparency of keloid scars. We then conducted a detailed 3D vascular analysis using light sheet and confocal fluorescence microscopy. In normal skin, capillary loops and the superficial vascular plexus were located in the papillary layer and at the boundary between the papillary and reticular layers, respectively. However, the density of these vessels was higher in keloid scars than in normal skin. The reticular layer of normal skin exhibited fewer blood vessels. In contrast, keloid scars exhibited significantly thickened dermal reticular layers, with the upper reticular layer showing significantly greater vascularisation. The lower reticular layer of keloid scars also exhibited vertically aligned blood vessels, although their density was lower than in the upper reticular layer. These results indicate that excessive vascularisation is predominantly induced in the papillary and upper reticular layers of keloid scars, which might contribute to keloid pathogenesis. A key element of prevention is the assessment of PI risk. The study aimed to investigate whether guidance documents relevant to the United States (US) advocated for specific risk assessment recommendations. We conducted a systematic review of guidance documents published between 2010 and 2024. Embase, Medline, Cinahl, and four key organisational websites were systematically searched to retrieve relevant articles. Two independent reviewers screened the articles for inclusion. One reviewer extracted the data, and a second reviewer checked all extracted data. Three reviewers assessed the guidance documents quality using the Appraisal of Guidelines for Research & Evaluation (AGREE II) tool. A narrative synthesis was used to describe and summarise findings. Six clinical practice guidelines (CPGs) and eight other best practice recommendations were included. The median scores of most AGREE II domains were higher for CPGs compared to other best practice recommendations. Risk assessment was consistently positioned as a critical first step in the prevention of PIs, emphasising its role in identifying at-risk individuals and informing targeted interventions. Although risk assessment was presented as a crucial step in PI risk prevention, there was no clear and unanimous recommendation for a specific risk assessment strategy across all guidance documents, either for the general population or for specific subgroups of patients in US healthcare settings. Our group has employed artificial dermis grafting for full-thickness temporal defects to overcome the limitations of traditional reconstructive methods, such as local flaps and skin grafts. This study evaluates the changes in colour matching and the degree of scar contraction following artificial dermis grafting and explores its potential for reconstructing such defects. This retrospective study included 25 patients who underwent artificial dermis grafting after skin cancer excision in the temporal region. Colour differences between the scar and surrounding skin were quantified using dE2000 scores. Scar contraction was assessed by measuring scar areas. These two parameters were evaluated intraoperatively, immediately after wound healing, and at 3, 6, and 12 months post-healing. The dE2000 scores immediately after healing and at 3, 6, and 12 months were 15.4 ± 7.4, 14.9 ± 6.8, 10.4 ± 4.6, and 6.3 ± 2.0, respectively (p < 0.01). According to reference values, the colour mismatch was rated as \"fair\" until 6 months post-healing and as \"very good\" at 12 months. The amounts of scar contraction immediately after healing and at 3, 6, and 12 months were 55.3 ± 10.5%, 65.6 ± 8.6%, 32.5 ± 15.9%, and 20.6 ± 14.8%, respectively (p < 0.01). These findings indicate that artificial dermis grafting for full-thickness temporal defects initially leads to significant colour mismatch and scar contraction. However, both parameters improve over time, achieving favourable outcomes within 12 months. Understanding the sociodemographic factors influencing length and cost of hospital stay is crucial for addressing healthcare disparities. This study investigated the associations between sociodemographic factors and hospital stay outcomes among patients with pressure injuries in the United States from 2009 to 2019. Using the National Inpatient Sample (NIS), we analysed data from 1,252,729 patients with pressure injuries. We performed survey-weighted multivariable linear regression to assess the impact of sociodemographic factors on hospital stay duration and costs. Minority racial groups, lower-income patients, and those with non-Medicare insurance experienced longer hospital stays and higher costs. Older age groups (≥30 years) had shorter stays and lower costs. Significant disparities exist in hospital outcomes for patients with pressure injuries. Considering the unmet need for safe and effective therapeutic approaches for wound healing, research on discovering new bioactive materials that support all stages of wound healing is gaining importance. In this study, the wound-healing activity of postbiotics obtained from Limosilactobacillus reuteri EIR/Spx-2, isolated from the gut microbiota of long-lived blind mole rats (Nannospalax xanthodon), was investigated. Our results demonstrated that postbiotics exhibited a strong inhibitory effect against important skin pathogens, eliminated their biofilm formation, and downregulated the expression of genes involved in their quorum-sensing regulatory mechanisms. Furthermore, treatment with postbiotics resulted in a significant increase (23.82% ± 2.11%) in L929 fibroblast cell proliferation. Additionally, postbiotics applied on scratched fibroblast monolayer significantly accelerated the re-epithelialization by 66.78% ± 3.74%. The treatment also increased the mRNA expression and protein levels of COL1A1 in the early healing phase. Moreover, the intracellular ROS levels of L929 cells suppressed by H<sub>2</sub>O<sub>2</sub> were significantly reduced, which could be attributed to the content of flavonoids (4.8 mg/g) and phenolic compounds (7.12 mg/g) in postbiotics, as well as their DPPH scavenging activity. After treatment with postbiotics, the mRNA levels of IL-6 (5.77-fold) and TNF-α (1.76-fold) and the amount of NO (79.25% ± 3.18%) were significantly decreased in LPS-induced murine macrophages.
