Methods: A total of 140 patients with type 2 diabetic peripheral neuropathy, who were treated at the Zhanjiang Central People's Hospital between February 2022 and February 2023, were selected as subjects. They were randomly divided into two groups using a computer- generated random number table. The control group (n = 70) received epalrestat treatment, while the study group (n = 70) was treated with a combination of epalrestat and Sulodexide11. Efficacy, nerve conduction velocity, levels of inflammatory factors and oxidative stress markers were compared between groups. Results: After treatment, total effective rate in study group was 97.14 %, which was higher than that of control group (85.71 %; p < 0.05). Sensory nerve conduction velocity (SCV) and motor nerve conduction velocity (MCV) of the common peroneal nerve and median nerve in study group were significantly faster than those in control group (p < 0.05). Levels of serum high-sensitivity C-reactive protein (hs-CRP), interleukin-6 (IL-6), procalcitonin (PCT) and tumor necrosis factor-α (TNF-α) in study group were significantly lower than those in control group (p < 0.05). Total antioxidant capacity (T-AOC) and superoxide dismutase (SOD) levels in study group were higher than those in control group, while malondialdehyde (MDA) level was lower than control group (p < 0.05). Conclusions: Epalrestat combined with Sulodexide in treatment of diabetic peripheral neuropathy in type 2 diabetes improves efficacy, accelerates nerve conduction velocity, reduces inflammatory factor levels and decreases oxidative stress response. Further studies are necessary to accurately verify the combination therapy value using epalrestat and Sulodexide in treating peripheral neuropathy associated with type 2 diabetes.","PeriodicalId":23347,"journal":{"name":"Tropical Journal of Pharmaceutical Research","volume":"23 12","pages":"2017-2023"},"PeriodicalIF":0.0,"publicationDate":"2025-01-17","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ajol.info/index.php/tjpr/article/download/286170/269610","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147919156","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods: Maestro software was utilized to design new HDAC inhibitors. The organic synthesis of compounds VIa-VIc was started with the Williamson reaction between benzylic halide derivatives and methyl 4-hydroxybenzoate to form ethers IIIa-IIIb. The resultant ethers were subjected to ester hydrolysis, followed by an amide reaction with 1,2,4-thiadiazol-5-amine to produce the final compound VIa-VIc. The structures of synthesized compounds were characterized using NMR and FTIR spectroscopic techniques. Anti-proliferative activity on colon cancer cells (HRT) was evaluated using MTT assay. Results: Docking study revealed that compounds VIa-VIc had in silico binding affinity for HDAC enzymes, while MTT assay showed that the IC50 values of VIa and VIc (1.00 and 1.44 µM, respectively) were comparable to IC50 of 3.00 µM for the reference compound, vorinostat used in this study. Conclusion: New potential HDAC inhibitors with a thiadiazole moiety as a possible zinc-binding group have been successfully designed, synthesized and characterized. Results from preliminary cytotoxicity evaluation were highly promising. These findings may be useful for developing novel therapeutic agents.","PeriodicalId":23347,"journal":{"name":"Tropical Journal of Pharmaceutical Research","volume":"23 7","pages":"1069-1076"},"PeriodicalIF":0.0,"publicationDate":"2024-09-06","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ajol.info/index.php/tjpr/article/download/278006/262250","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147890902","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods: Differential expression of miR143HG between NSCLC tissues and healthy counterparts was identified through bioinformatic analysis. Subsequently, this expression difference in A549 and BEAS2B cells was validated using quantitative polymerase chain reaction (qPCR). Overexpression of miR143HG in A549 cells was achieved through liposome-mediated transfection, while cell proliferation as well as apoptosis levels were assessed using cell counting kit-8 (CCK-8) assay, flow cytometry, and protein blotting. Expression of miR-504 (predicted as a target of miR143HG) was determined. Furthermore, A549 cells that overexpress both miR143HG and miR-504 were generated for comparison with cells overexpressing only miR143HG. Results: Compared to BEAS-2B cells, A549 cells showed significantly reduced miR143HG and elevated miR-504 levels (p < 0.05). In A549 cells with miR143HG overexpression, cell proliferation and miR-504 expression significantly reduced while pro-apoptotic proteins (BAX, p53) significantly increased (p < 0.05). Simultaneous overexpression of miR143HG and miR-504 in A549 cells counteracted the effect of miR143HG alone, thus enhancing proliferation and diminishing pro-apoptotic protein expression, similar to control. Conclusion: Downregulation of MiR143HG occurs in human lung adenocarcinoma cells. Upregulation of MiR143HG impedes A549 cell proliferation, promotes apoptosis through pro-apoptotic protein modulation and suppresses miR-504. Co-overexpression of miR143HG and miR-504 reverses these effects, resembling control conditions. Thus, miR143HG exerts its anti-cancer effect in A549 cells by modulating miR-504 and activating p53 pathway.","PeriodicalId":23347,"journal":{"name":"Tropical Journal of Pharmaceutical Research","volume":"23 8","pages":"1231-1237"},"PeriodicalIF":0.0,"publicationDate":"2024-09-06","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ajol.info/index.php/tjpr/article/download/278023/262267","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147887048","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods: Forty male Sprague-Dawley (SD) rats were assigned to four groups: control, sham, UUO and cordyceps-treated groups (10 rats per group). The UUO and cordyceps groups underwent surgery to induce unilateral ureteral obstruction. The cordyceps group received intravenous cordycepin (10 mg/kg) daily for 14 days, while the control and UUO groups received normal saline. Histopathological examination, assessment of fibrosis markers (α-SMA, collagen III) and autophagy markers (Atg5, LC3II/I) were conducted. Results: The UUO group exhibited significant tubular damage and interstitial fibrosis, with elevated serum levels of pro-inflammatory and oxidative markers (p < 0.05). Cordycepin treatment attenuated these pathological changes, evidenced by reduced fibrosis, inflammation and oxidative stress. Enhanced autophagic activity was observed in the cordycepin group, suggesting a potential mechanism of its renoprotective effect. Conclusion: Cordyceps is effective in inhibiting renal tubulointerstitial fibrosis, potentially through the activation of autophagy and reduction of inflammation and oxidative stress. Future studies should focus on unraveling the specific molecular mechanisms of cordycepin's action and assessing its applicability in CKD treatment.","PeriodicalId":23347,"journal":{"name":"Tropical Journal of Pharmaceutical Research","volume":"23 3","pages":"529-535"},"PeriodicalIF":0.0,"publicationDate":"2024-04-14","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ajol.info/index.php/tjpr/article/download/268385/253252","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147880021","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
