Hori","doi":"10.3191/thermalmed.37.141","DOIUrl":"https://doi.org/10.3191/thermalmed.37.141","url":null,"abstract":"ラズマ活性酢酸リンゲル液との併用 田中 宏昌・堀 勝 名古屋大学低温プラズマ科学研究センター 近年,低温プラズマによるがん治療研究が盛 んに行われるようになり,その中でもプラズマを 照射した溶液(プラズマ活性溶液と呼ばれる) による抗腫瘍効果が注目を集めている.2012年 に プ ラ ズ マ 活 性 培 養 液(Plasma-Activated Medium, PAM)と称されるプラズマ照射した細胞 培養液がアストロサイトの正常細胞に対しては影 響しないが,脳腫瘍培養細胞に対して選択的に細 胞毒性を示すが報告されて以来,様々な溶液や 様々ながん細胞に対してプラズマ活性溶液の抗腫 瘍効果が報告されてきた.2016年にはプラズマ 活性乳酸リンゲル液やプラズマ活性酢酸リンゲル 液による抗腫瘍効果が報告され,それらの溶液 内の成分,作用機序,動物実験など,プラズマ活 性溶液の臨床応用に向けた研究が広く進められて いる. ここではハイパーサーミアに加えてプラズマ活 性酢酸リンゲル液(Plasma-activated Ringer’s acetated solution, PAA)を併用し,相乗的に抗腫 瘍効果を高めることを証明した研究内容について 紹介する.A549ヒト非小細胞肺癌細胞株を用い て,ハイパーサーミア(42 °C)及び PAAの相乗 効果が調べられた.ハイパーサーミア単独やPAA 単独に比べ,ハイパーサーミアとPAAの併用は相 乗的に A549細胞に対して細胞毒性を示すことが 示された.更に,ハイパーサーミアと PAAの併用 によりA549細胞の細胞内Caの増加が確認され たため,Transient Receptor potential(TRP)チャ ネルの 1種である TRPM2(TRP metastatin 2)の 機能が調べられた.TRPM2の阻害剤(2-aminoethoxydiphenyl borate, 2APB)や siTRPM2による遺 伝子のノックダウンにより,ハイパーサーミアと PAAによる細胞内 Caの増加と抗腫瘍効果が抑 制されたことから,ハイパーサーミアとPAAの併 用による抗腫瘍効果には TRPM2が関与している ことが証明された.さらに,A549細胞における ハイパーサーミアとPAAの併用により,細胞内活 性酸素種の増加,DNAの断片化,poly(ADPribose)polymerase-1(PARP-1)の活性化の促進, アポトーシスの誘導が示された.以上より,PAA 処理とハイパーサーミアの併用により細胞内活性 酸素種が増大し,その結果 DNAの断片化と PARP-1の活性化が促進される.そのことによっ て TRPM2が活性化されて,細胞内 Ca2+がさら に増加することで細胞死が誘導されるというメカ ニズムが提唱された.つまりハイパーサーミアは, がん細胞の PAAに対する感受性を相乗的に増強 する.正常細胞として用いられた線維芽細胞にお いては,ハイパーサーミアとPAAの併用による細 胞毒性は認められなかった. 最近,ハイパーサーミアと低温プラズマの併用 による抗腫瘍効果の増大が U937リンパ腫細胞株 で報告されており,ハイパーサーミアと低温プ ラズマ/プラズマ活性溶液との併用療法は,有望 な治療法として期待される.","PeriodicalId":23299,"journal":{"name":"Thermal Medicine","volume":"15 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2021-12-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"75608814","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
