The cooked sweet potato was fermented according to the factorial design with the following factors: the time (21.51h-38.48h) and the amount of Lactobacillus (1.58*106-5*106UFC). Responses were represented by production yield, pH, and titrable acidity. Characterization of the exopolysaccharides was then done by determination of total sugars, solubility index and viscosity. The results show that the pH of unfermented cooked potatoes was between 6.23 to 6.63. The pH of fermented potatoes varies from 3.96 to 4.06. The lactic acid content was found from 7.75 to 9.9% for the fermented samples. The production yields are 1.90% for the samples fermented for 21.5 hours with 3.5*106UFC of bacteria and 5.62% for those fermented for 30 hours with the same volume of inoculum. The average viscosity of the products was 4mPas regardless of the fermentation time. Chemical characterization indicates glucose contents of 68.21 and 94.01% in fermented potato for 24h and 21h respectively. The solubility index gives values of 70.3 ± 0.16 and 88.11 ± 0.23 for fermentation times of 21h and 24h respectively. Results of this work indicated that cooked sweet potatoes ferment for 21h was a promising substrate for production of exopolysaccharides.","PeriodicalId":220919,"journal":{"name":"Biotechnology and Bioprocessing","volume":"1 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2021-09-03","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"130390507","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This definition has expanded to include the specific interaction between a target and a ligand. The use of affinity chromatography has reached a zenith with the explosion of Mab therapeutics and the use of Protein-A chromatography for antibody capture. Now, affinity chromatography has moved to non Mab proteins. This can result in the same economic advantages as Mab, by enabling the standardization of process development and manufacturing processes in flexible multiproduct production sites. The output is improved product throughput, higher target recoveries, and potentially less expensive drugs. These advantages are available to the developed world but how do we make this technology available to the developing world?","PeriodicalId":220919,"journal":{"name":"Biotechnology and Bioprocessing","volume":"70 ","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2021-09-03","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"120874481","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The positive effects of the plant on health have been described frequently from the ancient Roman period until the middle ages. It is used for the treatment of bronchial infections, colds and cough. It is also traditionally used to relieve gastraintestinal symptoms such as dyspepsia, gas, indigestion, bloating. In the treatment of excessive sweating in menopause. It has been shown to be effective. Its effectiveness is used as a mouthwash in the irritation of the oral mucosa as well as in pharyngitis, tonsillitis, gingivitis, glossitis infections. These teas used in treatment are prepared as infusion, decoction and cold maceration. S. officinalis plant grown in July Organic Farm in Akyokuş region in Konya province was used. The analyzes were carried out in the Western Mediterranean Agricultural Research Institute Directorate Medical and Aromatic Plants Center Laboratory in 2017. Approximately 20 g of dried plant material prepared for analysis. The sample is weighed. The weighed sample is placed in the glass clevenger flask. Approximately 10 times (200 ml) of pure water is added to the sample. It is subjected to hydrodistilation process for about 2 hours. Then the reading of the essential oil sample, which accumulates in the graduated part and creates a phase difference with water, is taken and the result is recorded in ml. Then, based on the weight, the amount of essential oil is calculated as a percentage. In our study, α-tujone 24.48%, camphor 16.45.1.8% cineol 12.38%, while viridiflorol was found as 6.5%. We think that the differences may be due to differences in geographical regions and precipitation. Ventilatory oil ratios have been shown in course studies that ontogenetic factors can also be affected by many factors.","PeriodicalId":220919,"journal":{"name":"Biotechnology and Bioprocessing","volume":" 47","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2021-08-13","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"120829914","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The initial CRISPR/Cas9 technologies, though emerged an improved version of genome editing in competition with TALENS and ZFNs, was nevertheless not free from technical and off-target effects. Technological improvements overtime start addressing issues with original CRISPR/Cas9 technology. The major areas of improvement targeted nucleases and delivery methods. Overtime the nuclease like Cas9 had some modifications like FokI-dCas9, Truncated guide RNAs (tru-gRNAs), Paired Cas9 nickase, Cpf1, Cas6 with Csm/Csr complex and chemically treated Cas9. In terms of delivery methods the improvements came along after almost all methods including viral methods like Recombinant Adeno Associated Viruses (rAAV), Lentivirus (LV), and bacteriophages. The review summarizes various non-viral gene delivery modes including physical methods like electroporation and chemical methods like nano particles, cell-derived membrane vesicles (CMVs) with upgraded developments. The review also compares various modes of delivering CRISPR gene editing machinery.","PeriodicalId":220919,"journal":{"name":"Biotechnology and Bioprocessing","volume":"121 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2021-06-24","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"115092416","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Among different soil samples analysed, Puttad (Ptd) recorded significantly superior actinobacterial population on starch casein aga, Kenknight & Munaier’s agar and actinomycetes isolation agar. Actinobacterial colonies could not be detected in any of the flooded soil samples on any medium, even at a dilution of 10-1, except in Meppadi soil, which recorded a low population of 0.3x101cfu g-1 soil. Starch casein agar is best media to isolate actinobacteria from soil samples compared to other two media. The cultural, morphological and biochemical characterization of thirty-five isolates was carried out. Further the isolates were evaluated for their plant growth promoting traits such as IAA production, nitrogen fixation, P, K and Zn solubilization. The isolates Ptd-A and Amb-C were found to be significantly superior to all other isolates, with IAA production of 15.9 g ml-1 and 15.38 g ml-1 respectively. The four isolates viz. Ptd-A, Ptd-E, Ptd-B and Ptr-A recorded significantly superior nitrogen fixation and the phosphate solubilized was significantly higher in Ptd-E, Ptd-D, Ptr-E, Ptd-A and Ptr-A, as compared to other isolates. All isolates were negative to K and Zn solubilization. Based on in vitro evaluations, three isolates were shortlisted (Ptd-A, Ptd-E and Ptr-A) and subjected to in vivo evaluation for growth promotion in black pepper (variety Panniyur 1). Rooted plants of black pepper were raised in sterile potting mixture. Bioinoculants applied at the time of planting and 45 days after planting. The PGPR Mix-1 and Organic Package of Practices Recommendations (2017) were used for comparison with the microbial inoculants along with control. In the in-planta experiment, biometric characters were recorded at monthly intervals, up to five months. The actinobacterial treatment, T1: Ptd-E, T2: Ptd-A and T3: Ptr-A showed significant increase in shoot length, number of leaves and internode length throughout the growth period from planting to five MAP. Significantly higher root growth was observed in treatment T2: Ptd-A, with significantly higher root volume, fresh and root weight. The potential actinobacteria were identified Ptd-A and Ptr-A as Streptomyces sp. and Ptd-A as Actinobacteria bacterium using 16S r RNA gene sequencing.","PeriodicalId":220919,"journal":{"name":"Biotechnology and Bioprocessing","volume":"220 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2021-06-24","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"126114423","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Accumulation of aluminum has been associated with a variety of pathologies such as anemia, osteodystrophy, joint diseases, muscular weakness, and Alzheimer’s diseases Fenugreek extracts have been shown to be neutralizing of free radicals and enhancing antioxidant status. Objectives: The present study aimed to evaluate the ameliorative effects of fenugreek seeds against hematotoxicity induced by Aluminum chloride in male rabbits. Materials and Methods: This study included twenty-four adult male rabbits, which were divided into 4 groups, 6 rabbits for each. Group I (control group): Animals were provided with tap water and fed with a normal diet for 30 days. Group II (Fenugreek seeds powder group): Fenugreek seeds powder was given to rabbits in food at a dose of 10 g per kilogram of diet weight/kg of body weight/day for 30 Days. Group III (Aluminum chloride (ALCl3) group): Rabbits were treated orally with 150 mg/kg BW of AlCl3/day for 30 consecutive days. Group IV (Aluminum chloride/fenugreek co-administered group): Fenugreek seeds flour was added at a rate of 10 g per kilogram of diet weight, and rabbits were treated orally with 150 mg/kg BW of AlCl3/day for 30 consecutive days. At the end of the experiment and 24 hours after the last dose, all animals were anesthetized with ether and blood samples were collected by heart puncture. Results: The results of the study showed that the treatment of male rabbits with aluminum chloride resulted in a significant decrease (P<0.01) in RBCs count, hemoglobin concentration, hematocrit value, MCV, MCH, and MCHC as compared to the control group. While there was a significant increase (P<0.01) in WBCs count, lymphocytes, and monocytes percentages and a significant decrease in granulocyte percentage when compared with the control group. Co-administration of fenugreek seeds powder and AlCl3 significantly improved all haematological parameters. Conclusion: The results showed that the administration of rabbits with aluminum chloride caused a hematotoxicity, and co-administration of fenugreek seeds powder with AlCl3 alleviate the hematotoxicity induced by AlCl3. The use of fenugreek seeds powder by humans can be considered beneficial in the alleviation of hematotoxicity. It is recommended that humans exposed to AlCl3 should be advised to take fenugreek seeds powder as a rich source of antioxidants to prevent hematotoxicity induced by AlCl3. Further studies are necessary to elucidate the exact mechanism of the anti-hematotoxic effect of Fenugreek seeds powder and the potential usefulness of fenugreek seeds powder as a protective agent against AlCl3 induced hematotoxicity in clinical trials.","PeriodicalId":220919,"journal":{"name":"Biotechnology and Bioprocessing","volume":"51 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2021-05-28","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"124017836","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
