Meštrovića Split Croatia Fisheries","doi":"10.24193/subbchem.2020.1.10","DOIUrl":"https://doi.org/10.24193/subbchem.2020.1.10","url":null,"abstract":"Fish otoliths pose increasing interest due to their potential of rendering information about environmental changes, underlined in their non-linear time-dependent biogenic crystal growth. Otoliths are acellular, metabolically inert and continuously grow in a complex process which still needs to be understood. In the present work confocal Raman microspectroscopy (CRM) and imaging is employed to investigate the growth pattern in otoliths from Sparus aurata of Mediterranean provenance. CRM signal acquired from otolith sagittal section is exploited to associate it with the periodic growth increments denoted as rings. Raman signal collected from the core to the margins with micrometer spatial resolution invariably revealed characteristic signal of aragonite. Relative intensity variability was observed particularly for the lattice modes, indicating changes in crystalline orientation relative to incident laser. Bands associated with organic fraction were absent in the 90-1840 cm-1 spectral range. Daily growth rings were further studied using the Raman mapping of main aragonite bands intensities at 1083, 704 and the lattice modes in the 100-350 cm-1 range. The spectral intensity pattern closely follows the daily growth pattern. The resulted reagentless amperometric biosensors were inserted in a flow injection setup and used as electrochemical detectors for the biogenic amines (BA) detection. Both biosensors were operated at low applied potential (-50 mV vs. Ag/AgCl, KCl0.1M) where electrochemical interferences are minimal. The quantification of ten BA (tyramine, putrescine, cadaverine, histamine, cystamine, phenylethylamine, agmatine, tryptamine, spermine, and spermidine) either individual or in mixture (after a preliminary separation by using cation exchange chromatography) was reported. G/(Os-RP)-HRP-GPAO biosensor detected all ten BA, while G/(Os-RP)-HRP-OPAO biosensor detected only spermine and spermidine. Finally, a simple and low-cost method for free and acetylated polyamines determination in human urine samples, by using the highly selective G/(Os-RP)-HRP-OPAO biosensor, was proposed. The sensitivity of the two methods (LODs HPLC-NDA-FL in the range 27-73 μg/L; LODs HPLC-DBS-TLS in the range 3.4-11 μg/L) was higher than that of the official method for biogenic amines in wines, OIV-MA-AS315-18 (60-77 μg/L). For its best performances, the HPLC-DBS-TLS technique was applied to the analysis of putrescine, cadaverine, histamine and tyramine in two white wine samples. Additionally, exploiting the Berthelot reaction, the TLS fast screening of biogenic amines in wines, following the release of ammonia by transglutaminase, was also proposed. The HLB values of non-ionic surfactants and their mixtures were determined by three different methods: theoretical HLB based on the chemical structure and the given HLB of surfactants, the Greenwald method and an improved, titrimetric method. Our aim was to investigate if the new method to determine the water number and HLB value is suitable for this type of commercial surfactants and their mixtures. Distinct molecular descriptors were used. Dimensionality of data, using PCA, correlation plots and clustering, was evaluated. Analyzing data dimensionality allowed model optimization. Docking studies and PCA were used in order to expand data dimensionality. Optimization of protein expression and purification conditions yielded 15 mg pure soluble protein from one liter of E. coli culture. Enzymatic activity measurements of the ammonia elimination reaction from different natural aromatic amino acids proved the protein to be a phenylalanine ammonia-lyase.
