The pressure in unsealed-chamber is ambient-pressure and sealed-chamber is a specific pressure defined during bonding processes. Two types of sensors were characterized to verify the process scheme. One is a 32 kHz resonator and the other is a Pirani gauge. Characterizations on these two sensors prove that the sensors are functional in different pressure conditions in both unsealed chambers and 10mbar sealed chambers. The rheological testing demonstrated tunable mechanical properties of composite hydrogel based on UV crosslinking time. The leakage testing verified longer stability time of gold nanoparticle-based X-ray attenuating media. One limitation, however, is that retrieving synthesized nanoparticles from microdroplets typically requires laborious and time-consuming “off-chip” procedures (e.g., droplet collection, centrifugication and nanoparticle resuspension). To bypass these issues, here we introduce a continuous flow microfluidic system to enable the rapid and autonomous droplet-based generation and retrieval of nanoparticles. Specifically, we utilize a micropost array railing technique in order to passively: (i) generate microdroplets in which nanoparticles are synthesized, (ii) guide the particle-containing droplets into an oil-phase wash solution (i.e., to remove surfactant), and (iii) “lyse” the microdroplets to release the nanoparticles into the water flow. The unique aspect of the sensor is the integration of electroplated palladium (Pd) working and auxiliary electrodes on copper (Cu) substrate with a new Cu/CuCl2 reference electrode, which reduces cost and simplifies fabrication. The sensor was fully characterized and was found to exhibit a linear response in borate buffer with limit of detection (LOD) of 330nM (18.6ppb). The morphology, structure and chemical composition of the film were examined by SEM and XRD. Test sensor employing the ZrO2-WO3 mixed oxide film as active layer was fabricated and utilized for detecting H2 and CO gases of various concentrations (100-1000 ppm) at temperatures up to 300°C. The sensor was fast and highly responsive to hydrogen while showing much weaker and slower response to carbon oxide. Due to surface tension, the liquid filled the cantilever's gap while it did not leak through the microscale gap. Evaluation experiments showed that the combination of liquid bridge and cantilever enables the hydrophone to have high sensitivity. The device is capable of both isolating analyte from samples and dispensing purified samples onto a probe plate for a following MS analysis. Aptamers selectively captured and released target molecules for isocratic elution. An integrated nanodrop generator produced small sample spots (300 μm diameter), significantly minimizing spatial intensity variations. Repeated sample deposition on the same spot (50 times) induced on-spot analyte concentration improving the detection sensitivity. Thanks to these properties, they have been integrated into different micro- and nanoscale transducers, resulting in a new type of biosensors, known as `aptasensors', used in a truly new generation of diagnostic tools.
