The directions are controlled by electrical charges of cargo molecules partially labeled on the minus ends of MTs. We designed MTs labeled with streptavidin (SA), nanogold (NG), or dsDNA molecules, and evaluated their curvatures in an electrical field. The curvature of DNA-MT was larger than those of SA-MT or NG-MT because of DNA's large negative charge. Recent advances in epigenetic studies highlight the importance of DNA methylation in the biology of cancer. The epigenetic signature of promoter regions in the genes ADAMTS1 and BNC1 has been shown to have strong correlation with the early stages of pancreatic carcinogenesis [1]. Most analytical techniques for studying methylation require pre-processing of sample DNA via bisulfite conversion. However, repetitive addition and removal of liquid reagents using conventional tubes and pipettes present a technical hurdle for microliter-scale reagents, as well as a high risk for contamination and human error. Previously, we reported a magnetically actuated platform for integrated DNA extraction and amplification from crude biological samples [2]. Data evaluation was performed using multivariate statistics, here by Linear Discriminant Analysis. It was found that both temperature cycling and gate bias cycling are suitable for quantification of different concentrations of carbon monoxide. However, combination of both approaches enhances the quality of the separation. Novel force and displacement sensors and actuators have been applied to study mechanisms of cell adhesion and the development and response of pluripotent cells in varied mechanical environments, to assay the function of hear cells, and to probe the mechanics and biology of the sense of touch in C. elegans or the mechanosensation of hair cells. In the Stanford Microsystems Lab, we design and fabricate many custom tools and sensors to probe cellular dynamics. We are interested in the reliable manufacture and operation of these micromachined devices in harsh biological environments as well as calibration approaches for precise measurements of nanoscale mechanical behaviors. We use these tools to address questions in the areas of physiology, biology, stem cells, neuroscience and cardiology with an eye toward quantitative and fundamental biophysics. In particular, microcantilevers have enabled measurements in a variety of applications over the past decades, ranging from scanning probe microscopy to visualize topography or characterize materials properties, to chemical sensing. An electrowetting-based valve consisting of gold and zinc opened when an electrolyte solution reached the zinc part in a controlling flow channel and a mixed potential was applied to the gold valve electrode. Clock circuits that open the valves at predetermined times and logic circuits have been realized. A bubble micropump that moves a solution forward and backward was also constructed with three flow channels in which platinum black, zinc, and silver electrodes connected with each other. The volume sensor and the glucose sensor in the microfluidic system are characterized. The role of the capacitor is to eliminate the electric potential difference between the suspended beam and the drain electrode at the switch “opening” instant. In the case of a pure Au-Au contact, we have experimentally demonstrated that the lifetime of conventional MEMS switches with the suggested method was improved dramatically, by an order of magnitude, in the relatively high electric-field hot switching condition. We clearly observed material transfer at failure in the device without the capacitor, whereas stiction was the main failure mechanism in the device with the capacitor. Confinement is realized in a so-called phoxonic crystal, a specific version of a metamaterial. Metamaterials are engineered to achieve desired effects by incorporating structural elements of (sub-)wavelength sizes. The sensor platform is a solid plate featuring an array of holes arranged in a square lattice.
