{"title":"Uncovering the Role of Ribosomal Protein L8 in Milk Fat Synthesis Mechanisms in Yak Mammary Epithelial Cells","authors":"Nan Jiang","doi":"10.17582/journal.pjz/20230415100435","DOIUrl":"https://doi.org/10.17582/journal.pjz/20230415100435","url":null,"abstract":", RPL8, mTORC1, SREBP1 The fat content in yak milk is higher than that in dairy cows, but the molecular mechanisms responsible for milk fat synthesis in yaks are still unclear. This study examined the regulatory mechanism of milk fat synthesis in yak mammary epithelial cells (YMECs) by investigating the role of Ribosomal protein L8 (RPL8) in the mTORC1-SREBP1 signaling pathways. The results showed that over-expression or inhibition of RPL8 had a significant effect on triglyceride (TG) secretion, which also affected the sterol regulatory element-binding protein 1 (SREBP1) pathway. Similarly, the intervention of SREBP1 revealed that RPL8 promoted TG secretion through the SREBP1 pathway. Additionally, the study found that over-expression or inhibition of RPL8 regulated the signaling activity of the mammalian target of rapamycin complex 1 (mTORC1), which promoted the SREBP1 signaling pathway through mTORC1. Further examination by over-expressing or inhibiting SREBP1 or mTOR showed that mTOR promoted TG secretion through the mTORC1-SREBP1 signaling pathway. Moreover, YMECs treated with palmitic acid showed increased expression of RPL8, mTOR, and SREBP1, as well as increased TG secretion. Overall, the study’s findings provide insights into the underlying mechanisms of milk fat synthesis in yaks, and suggest that the RPL8 gene and mTORC1-SREBP1 signaling pathways could serve as potential genetic markers for milk fat synthesis in yak mammary glands.","PeriodicalId":19975,"journal":{"name":"Pakistan Journal of Zoology","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"136208718","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

A total of 112 patients with manic episodes from the Wuhan Wudong Hospital between July 2021 and December 2022 were assigned into a control group (CG) and a hypertension group (HG). The patients’ blood lithium concentrations, blood lithium C/D ratios, antihypertensive medication use, Young Mania Rating Scale (YMRS) scores, adverse reactions (ARs), and other parameters were monitored and compared. The average blood lithium concentration in the CG was 0.78±0.27 mmol/L, and the average blood lithium C/D ratio was 0.91±0.43 mmol·L -1 ·g -1 ·d, which were remarkably inferior to those (0.89±0.20 mmol/L and 0.97±0.46 mmol·L - 1 ·g -1 ·d) in the HG. Further subgroup analysis based on antihypertensive medication revealed a drastic increase in blood lithium C/D ratio in the calcium channel blocker (CCB) subgroup. After the treatment, the YMRS score was 7.84±0.82 in the CG and 9.12±0.89 in the HG. The total number of ARs in the HG was superior to the CG, and the occurrence of ARs in both groups was concentrated within 3-15 days, accounting for 48.6% of all ARs. Patients in the HG had higher blood lithium concentrations, slower lithium metabolism, and a higher risk of ARs versus the CG. It was concluded that further investigation is warranted to explore the underlying mechanisms of these effects.","PeriodicalId":19975,"journal":{"name":"Pakistan Journal of Zoology","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"136210037","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Post-fire silvicultural practices contributed to dramatically converting the structure of forests. Coverage of overstory, midstory, and ground vegetation, number of tree stems, woody seedlings, snags, and volume of coarse woody debris all had significant differences among study sites. We captured 456 individual small rodents during the dry and rainy seasons. The mean number of small rodents captured in primary forests was significantly higher than that in two secondary forests. In addition, there were more rodents in burned rice field stands than in burned rubber plantation stands. The value of overstory to understory vegetation coverage and the volume of coarse woody debris were strongly associated with small-mammal abundance in the PKKNPA. Thus, post-fire silvicultural practices should take the differences in small rodent abundance as well as forest structure into account. Long-term experiments may help illuminate the potential effects of management strategies after forest fires in the Lao PDR.","PeriodicalId":19975,"journal":{"name":"Pakistan Journal of Zoology","volume":null,"pages":null},"PeriodicalIF":0.6,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"67546175","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Both in-vitro and in-vivo studies were carried out to evaluate the antimicrobial activities and potency of this combination. A total of one hundred and eighty day-old chicks were randomly allotted to 6 groups i.e. G1 (negative control), G2 (positive control), G3 (standard antibiotic neomycin only), SN-1 (Serratiopeptidase @40g/L+Neomycin@10mg/L), SN-2 (Serratiopeptidase @50mg/L+ Neomycin @20mg/L), and SN-3 (Serratiopeptidase @ 60mg/L+ Neomycin @30mg/L). Each group consisted of thirty birds with 5 replicates (6 birds/ replicate). On day 21, fresh inocula of E. coli (1x10 9 cfu ) were orally administered to all the groups except negative control. The results revealed that birds in SN-3 group showed significantly larger zone of inhibition, increased feed intake, weight gain and improved feed conversion ratio compared to standard and control groups. Moreover, reduced (p<0.05) mortality and morbidity index was found in SN-3 group among all the treated groups. It was concluded, that use of serratiopeptidase combination with neomycin has better impact on broiler chickens to enhance the antimicrobial effect of Neomycin against E. coli infection and to enhance production parameters.","PeriodicalId":19975,"journal":{"name":"Pakistan Journal of Zoology","volume":null,"pages":null},"PeriodicalIF":0.6,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"67570310","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"生物学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
