In the present study, the anthelmintic activity of the extract of herbal complex (HC) composed of four herbs (Origanum vulgare, Cinnamomum verum, Rosmarinus officinalis and Capsicum annuum) was assessed against eggs and adult Haemonchus (H.) contortus. Egg hatch test (EHT) and adult motility assay (AMA) was performed to evaluate the anthelmintic potential of HC. The reference drug used in EHT was oxfendazole and for AMA two drugs i.e. oxfendazole (2.265 mg ml) and levamisole (3mg ml) were used. In EHT, LC50 values of HC and oxfendazole were 498 and 1.6 ppm, respectively. In AMA, 100% mortality of H. contortus was observed after 6 hr of treatment with HC (100 mg ml) where as two positive control groups could not kill all worms after this exposure time. These results indicated the anthelmintic potential of HC. ©2019 PVJ. The aim of this study was to characterize S. haemolyticus isolates from sick and healthy cats according to their antibiotic properties and biofilm formation. A total of 80 S. haemolyticus isolates from 36 healthy and 20 sick cats were investigated. All the isolates from the sick as well as from the healthy animals were multidrug resistant at the genetic level, whereas 82.5% showed phenotypic resistance. Almost all the isolates were methicillin-resistant S. haemolyticus (MRSH) at the genotypic and phenotypic levels of 98.75% and 96.25%, respectively. Moreover, the most frequently observed phenotypic resistances were those for erythromycin (87.5%) and penicillin (93.75%), and the isolates harboured genes involved in resistance to penicillin – blaZ (97.5%), aminoglycosides – aac(6’)Ie-aph(2”)Ia (92.5%), macrolide-lincosamide-streptogramin – ermB (98.75%) and ermC (72.5%), and tetracyclines – tet(K) (98.75%) and tet(M) (100%). The percentage of biofilmpositive strains in healthy and sick cats amounted to 92.68% and 97.44% for the microtiter plate test (P=0.4962) and 71.17% and 56.41% for the Congo red agar assay (P=0.5164), respectively. Statistically significant associations were observed when the owner’s occupation was connected with healthcare, both human or veterinary, there was previous hospitalization of the owner, or cats were kept with other animals in the household. MRSH strains isolated from companion animals were frequently multidrug resistant, although they remained susceptible to antibiotics used in mainly human medicine. ©2020 PVJ. Between 2012 and 2015, 55 samples from one layer flock from UAE, three breeder flocks from SAU and one broiler flock from SL were screened for MG using a pr- mgc 2 PCR; the positive samples were characterized by the gene-targeted sequencing (GTS) analysis. BLAST comparisons, followed by phylogenetic analysis, was carried out to compare the obtained sequences against published vaccine and field strain sequences. Fifty of the fifty-five (91%) examined samples, were MG positive by pr- mgc 2 PCR. The sequences detected in the samples from ts-11 vaccinated breeders from SAU showed 100% identity with the ts-11 vaccine sequences. The sequences detected in the samples from layers in UAE, were attributed to field strains, clustering together with Israeli MG strains. The sequences obtained from the samples from Sri Lanka broilers clustered separately from USA, Israeli, Australian, vaccine and reference strains. A potential route of MG field infection to the examined flocks may have been through the movement of birds, either directly (flock-to-flock transmission) or indirectly (through wild bird transmission). The study was conducted from July 2018 to January 2019. Blood samples were collected from male kids (n=26) at 10 days interval from 10 days of age to puberty for measurement of AMH, testosterone and E2 concentrations. The body weight of all kids was measured at 10 days of age and then on monthly basis till puberty. The results showed that the mean body weight at 10 d after birth was 7.04±0.86 kg, and significantly increased till the onset of puberty (36.4±4.49 kg). The minimal plasma AMH concentration was seen at 10 d, gradually increased to its peak level (P<0.05) at 40 d and 100 d and then declined steadily till puberty. The serum testosterone level increased non-significantly from day 10 to day 50 and then gradually increased to its peak level at 170 and 180d (P<0.05). The serum E2 concentration was low at 10 days of age (5.76±0.78 pg/ml), increased to its peak level at 60d (74.3±6.26 pg/ml; P<0.05 compared with 10d), then declined till puberty. The serum AMH at early pre-pubertal age (40, 50, 70 and 80d) was positively correlated with pubertal age around 180d (r=0.99, 0.95, 0.99 and 0.94), whereas testosterone concentration was negatively correlated with serum AMH level at 60d, while positively correlated at 150 and 180d. In conclusion, goat kids exhibit a characteristic serum AMH profile along with testosterone and E2 during pre-pubertal life, such that serum AMH at an early pre-pubertal age could be used as a biomarker for predicting initiation of sexual behavior and onset of puberty. ©2020 PVJ.
