In the process of ascites formation there is increased vascular permeability, decreased lymphatic resorption and gross change in the concentrations of functional proteins and metabolites. Liver cirrhosis is a chronic disorder in which ascites develop in advanced stages of the disease, where as in ovarian cancer, most of the cases develop ascites in the early stages after the onset of disease. Among malignant ascites, ovarian cancers are the most common primary tumors and sixth most common malignant neoplasm in women with highest mortality rate.1 Oxidative stress is caused by free radicals leading to oxidative destruction of poly unsaturated fatty acids constitutive of cellular membrane and give rise to lipid peroxides which are unstable and decompose to form reactive carbonyl compounds. Among them malondialdehyde (MDA) is the most abundant, and it readily combines with several functional groups on proteins and forms DNA-MDA adducts, which are highly mutagenic, cytotoxic, co-carcinogenic and acts as a tumor promoter.2–5 Oxidative stress induces cancer among the biological molecules and plays an important role in the pathogenesis of liver disease and other hepatic alterations.6 Intestinal permeability is increased in patients with cirrhosis and oxygen free radicals play an important role in gut epithelial damage. Hence measurement of MDA is widely used as an indicator of oxidative stress. The antioxidant enzyme superoxide dismutase (SOD) is widely distributed in the cells and present in higher levels in erythrocytes,7 and it plays an important role in scavenging and protects cells against superoxide radical by dismutation of the highly reactive superoxide anion to oxygen and less reactive oxygen species hydrogen peroxide.8 The low levels of SOD may be due to increased utilization of scavenger lipid peroxides. The imbalance between the free radicals and antioxidants results in oxidative stress. In case of Psoriasis there is an abnormal hyper proliferation in keratinocytes, arised due to the activation of T-cells which produces a rich amount of arachidonic acid which leads to generation of various pro inflammatory mediators like PGs, LTs, cytokines and adhesion molecules via MAPK/AP-1, EARK1/2 and protein kinase-C (PKCs) activation pathways.1 In order to treat Psoriasis there is a need for the incorporation of naturally occuring bioactives like, omega (ω)-3 fatty acids (i.e., EPA and DHA) in a dose dependent manner which results in the inhibition of various pro-inflammatory mediators. Metabolization of EPA and DHA leads to the dampening of inflammation and higher resolution of abnormalities in skin. In case of natural treatment the synthesis of ω-3 PUFA-derived lipid mediators, namely resolvins and protectins have been widely used alone or in combination with other drugs in the treatment. Despite of their meritorious visages, the use of these bioactives is associated with several hiccups like higher range of instability and vulnerable to degradation due to lipid per oxidation. This treatment is also resulted in poor and inconsistent bioavilability by oral and topical administration. The potential use of nanomedicines in the delivery of such bio-actives has gained wider attention owing to their promise in enhancing the characteristics of bioavailability, improved stability and better efficacy. HSP60 provides assistance in correct folding of a multitude of denatured proteins. The Group II eukaryotic chaperonin TCP-1 ring complex is (TRiC or CCT) important cytosolic chaperonin which plays important role in folding of essential subset of cytosolic proteins and is believed to be highly conserved among different eukaryotic species. In this study, an extensive in silico analysis has been performed ranging from sequence comparison among species to homology modeling of Bos taurus Cct5 protein and determination of Hsp60 interacting partners. The comparative analysis of the protein revealed certain significant variations in Bos taurus. The experimental protein structure for Cct5 in Bos taurus is unknown till date, therefore the putative protein structure was determined using homology modeling. The stereo chemical properties of protein were assessed by utilizing several scrutiny tools such as PROCHECK, VERIFY3D and RAMPAGE to ensure model accuracy. Proteins, the most important component of plasma, are considered the main executors of the physiological function of blood [1]. Posttranslational modifications (PTMs) of proteins can regulate protein functions by changing the physicochemical properties [2]. For example, phosphorylation or dephosphorylation may turn the kinase cascades in signaling on or off [3], and is often associated with tumorigenesis. So far, more than 500 types of PTMs or biological and chemical modifications have been identified and documented in the PSI-MOD [4], RESID [5], UniProt [6] and UniMod [7] databases. These translational modifications may determine the diversity of protein functions [8]. The molecular mechanisms of depression are poorly understood even though many studies have been attempted. Moreover, there are currently no distinctive biological markers to aid in the diagnosis and treatment of depression [2]. The diagnosis, evaluation of medication effects, and prediction of relapse are carried out completely by clinical judgments based on the phenomenology of the disease [1]. The first step requires identifying the molecular pathway for depression. Genomic approaches have attempted to find the underlying molecular pathways, but have not successfully identified the major genes for depression [3]. The nucleic DNA activity and regulation depend on the signals and the levels of the tRNA and rRNA which mtDNA produced in the cell. The foremost attention-grabbing issue, mtDNA has the ability to adapt with each individual cell by modifying the sequence by slightly the initiation and termination points, likewise, the direction of transcription 5’=>3’ or 3’=>5’.1 Mitochondria generate most of the cellular energy within the form of adenosine triphosphate ATP, regulate cellular oxidation-reduction state and integrate several of the signals for initiating necrobiosis. There raised an increase in pace to develop new technologies to diagnose the molecular signatures of complex diseases to initiate the therapy with respect to the principle of personalized medicine and hence the new era of molecular medicine laid the path for detection of disease, diagnosis of complex disorders and performing treatment with respect to the molecular profile of each and every individual [1-4]. This revolution had laid the foundation for the availability and application of new and novel biomarkers to predict the behavior of disease using advanced technologies to perform rapid diagnosis and detection. The evolutions of modern therapies to target cells were based on the principles of biocomputing. In order to address the molecular profiling and diagnostics, a major challenge was raised in characterization by histologic lesions in complex disorders because those lesions are heterogeneous at the cellular and molecular levels. In cancerous tumors, malignant cells are intermixed with blood vessels, stroma, and inflammatory cells [5-8]. Current technologies like Gene microarrays and Real-Time Polymerase Chain Reactions (RT-PCR) were not designed to handle the heterogeneic nature of cancer lesions and hence the development of nanotechnology provided a new opportunity to integrate morphological profile of diseased contion and molecular signatures and it is also used for correlating the observed cellular and molecular changes with respect to the behavior of pathological condition in complex disorders [9-11]. In particular, the application of bio conjugated quantum dots (QDs) [12-15] quantifies the presence of multiple biomarkers in intact tissue specimens and cancer cells and it allows us to conduct a comparative test between traditional histopathology V.S. molecular signatures of the same tissue [16-20]. Five cases of it were found among the Ibo/Igbos of South-eastern Nigeria on using the method of histopathology data pool that facilitates epidemiological analysis. These were then documented. Thereafter, it was theorized that, since the kidney was another seat of clear cell cancer, it was recognized as being researchable if the patients had been investigated from that direction!
