Mousa","doi":"10.21608/ejcp.2020.205590","DOIUrl":"https://doi.org/10.21608/ejcp.2020.205590","url":null,"abstract":"Biochemical and histological studies were made on roots of pepper (Capsicum annuum) infected with root-knot nematode and treated by certain chemical resistance inducers (salicylic acid (SA), indol acetic acid (IAA),ethylene (Eth), Jasmonic acid (JA), in addition to a biofertilizer named (Halex-2). These resistance inducers were tested at three concentrations 100, 200 and 300 μM for SA, IAA, and Eth and 15, 20 and 25 μM for JA while the biofertilizer was tested at 7g/L as biotic resistance inducers against root-knot nematode Meloidogyne incognita under greenhouse conditions at the Faculty of Agriculture Shebin El-Kom, Menoufia University in season 2016. All concentrations of the tested inducers significantly reduced root-galling and nematode population in soil as well as the number of egg masses and developmental stages per root system. The greatest inhibition effect on reproduction of nematode was recorded with Eth at 300μM, which did not significantly differ from that of a nematicide-treated plants. Also, all treatments significantly enhanced all vegetative plant growth characters compared with the infected plants. Biochemical analysis showed a significant increase in the activity of antioxidant enzymes (peroxidase, polyphenoloxidase and catalase), total and reducing sugars, and total phenols at the highest concentration of the tested inducers, especially, Eth compared with the nematode-infested plants. Histological studies indicated that less giant cells were observed in almost all treatments compared to the nematode-infected control plants. However, the highest concentrations of IAA, Eth (300μM) and Halex-2 treatments showed a good performance, with no giant cells found. Sixty days after nematode inoculation, all treatments (except SA and Halex-2) showed a poor formation of regular giant cells divided from cytoplasm and contained less number of nuclei compared to the infected plants. It seems that the tested inducers could be recommended to control M. incognita on pepper plants. Results indicated that, one month after applications, the highest decrease in the average numbers of plant parasitic nematodes was recorded with the treatments of Palestinian arum + Poultry manure (185.3 nematodes) and Neem + Poultry manure (200.3 nematodes), in comparison with the control treatment (390.7 nematode). After two months of applications, the highest decrease in the average numbers of nematodes was recorded with Neem + Poultry manure (90.3 nematodes) and Palestinian arum + farmyard manure (96.3 nematodes), in comparison with the control treatment (469.7 nematodes). Three months after applications, the highest decrease in nematodes was recorded with Palestinian arum + Poultry manure (68.3 nematodes) and Neem + Poultry manure (56.3 nematodes), in comparison with the control treatment (567.3 nematodes).The highest increase of tomato fruit yields was recorded with the nematicide Carbufuran , Neem + Poultry manure, and Neem + farmyard manure (50.68, 46.96, 45.36 %) . The current work was conducted to screen the efficacy of eight different Trichoderma isolates against two isolates of M. phaseolina under laboratory and greenhouse conditions. Dual culture technique results showed that the highest inhibitory rate of M. phaseolina isolates (M1 and M3) recorded with T. viride(2), whereas the least inhibitory effect recorded with T. harzianum under laboratory conditions. Greenhouse results showed that T. hamatum (1, 2) showed the highest percentage of survival plants infected with M. phaseolina isolates (1 and 3) while the lowest percentage of survival plants obtained with T. harzianum. The results revealed that all tested amino acids at concentration of 400 mg/ml reduced significantly both eggs hatching and juveniles’ viability compared to control (nematode treated with tap water) under in vitro conditions. Results revealed that chapped leaves of the four evaluated plants significantly, reduced all related nematode parameters i.e. number of galls, egg masses, females and developmental stages / root system, eggs / egg mass and number of juveniles / 250 g soil, in comparing with nematode alone. The highest significant reduction in galls, egg masses and J2 / 250 g soil (93, 92.8 and 89.6%, respectively) obtained with moringa at 4% of soil weight, followed by cabbage chapped leaves at 4% (90.8, 88.9 and 85.8%, respectively), whereas the lowest one observed with cassia chapped leaves at 1%. Generally, the effective dose was 4% regardless the plant species and the effective plant was moringa regardless the rate significantly enhanced all vegetative plant growth parameters i.e. fresh shoot and root weight (g), dry shoot weight (g), plant height and root length (g). Results showed also all treatments significant increase antioxidant enzymes activity i.e. peroxidase and phenoloxidase, total sugars, total phenols and proline concentration. Two magnetic levels were used to produce a high magnetic field, (MF1) = (20.8 mt. in lower and 1.3 mt. in center jars) and another low, (MF2 = (2.1 mt in lower and 1.21mt in center) compared with non exposed adults (control). The results demonstrated that the total eggs laid /female and hatchability in exposed adults were highly significantly affected compared with control. Mean numbers of eggs laid/female were 77.0 and 109.6, when adults were exposed to MF1 and MF2, respectively, compared with 175.0 eggs/ female in control. The percentages of eggs hatchability were 49.6 and 79%, in exposed adults to MF1 and MF2 respectively, compared with 95 % in control. Data reported that exposure of adults to selected magnetic fields; MF1 and MF2 reduced the mean number of eggs /female. Percentages were 56% and 48.8% when exposed to high and low power (MF1and MF2) respectively. The results indicated that longevity of exposed females and males were significantly affected compared with control. Longevity of adult males decreased to about half the time when exposed to high powers MF1, the longevity of exposed males to MF1 and MF2 were 6.6 and 9.6 days/ male, respectively but it was 13.3 days/male in control. In females, the longevity decreased after exposure to the higher MF, whereas prorogated after exposure to the lower magnetic levels, the longevities of exposed females to MF1 and MF2 were 12.8 and 16.4 days / female respectively, compared with 15.8 days / female in control. In additions, the effects of exposure to the magnetic fields MF1 and MF2 on some biochemical parameters were determined. Two application methods, spore suspension and powder of Pp EGY and Pp Jap used during the experiment. Results indicated that Pp EGY and Pp JAP showed a high reduction of root-knot nematode gall numbers compared to the control. Infected females of RKN with Pp EGY were markedly significant compared to Pp JAP. Plant growth was greatly influenced when both Pp EGY and Pp JAP were used. fresh shoot root and shoot dry weights were increased markedly compared to the control. Karnk) and faba bean ( Vicia faba , L cv. Balady) plants were collected from different districts in menoufia governorate, Egypt; showing vein chlorosis followed by green and/or yellow mosaic symptoms that may be due to virus infection. Diseased samples were subjected to identification of isolated virus based on symptomatology host range, serological tests (DAS- ELISA, Tissue blot immunoassay TBIA and electron microscopy). Histological studies of bean infected leaves by BYMV cleared that chloroplasts contained starch grain turned to spherical in shape and with lost of their envelopes totally or partially. Chlorophyll a, b and carotenoids as well as oxidative enzymes (peroxidase and polyphenol oxidase) were determined in viral infected leaves. Testing four strawberry cultivars for their susceptibility toRhizoctonia root rot disease and test the efficiency of (bio-control agents, soil mulch, antioxidant, chemical inducer and fungicides) to control the disease. The obtained data showed that all tested antagonistic and mulching soil significantly reduces root rot and increased fruit yield. The severity of infection was reduced when different antioxidant andchemical inducer were applied to the soil compared with the untreated soil. The combination between black mulch and fungicides was the best treatment compared to the other treatments. Integrated control significantly decreased the percentage of infected plants using (Black mulch+ Boscalid+ Trihoderma hamatum + Salisylic acid) followed by (Boscalid+ Black mulch) was the most effective treatment in controllingRhizoctonia root rot disease on cv. Five different organic solvents i.e., Petroleum Ether, Chloroform, Acetone, Ethyl Alcohol and Methyl Alcohol were used. Plant extracts screened for nematicidal activity against egg hatching and mortality of second-stage juveniles of M. incognita under laboratory conditions and on nematode infecting tomato plants under greenhouse conditions. Results showed significantly effect of all tested oleander extracts on egg hatching and larvae mortality under laboratory conditions. A great reduction in number of galls; eggmasses; females/root system of tomato plants and number of second stage juveniles (J2S)/250 g soil was recorded compared to the treated plants with nematode alone.
