Cestari","doi":"10.5132/EEC.2013.01.004","DOIUrl":"https://doi.org/10.5132/EEC.2013.01.004","url":null,"abstract":"The Comet Assay is a rapid and sensitive method for detection of breaks in the DNA strand into individual cells, commonly used at ecotoxicology. At this time, the method has been refined, but not yet fully standardized and variations on protocol are common. When environmental reviews are performed, the large number of samples required and the conditions of handling during transportation of samples to the laboratory are frequent problems. Until now, however, no truly effective method for samples preservation for this test was described. Therefore, we tested three stock solutions: fetal bovine serum (FBS), phosphate buffered saline (PBS) e Tris. Gill cells of Rhamdia quelen contaminated with insecticide Fipronil were maintained in these solutions, for 0, 24 and 48 hours to perform Comet Assay. It was found that fetal bovine serum was the solution that best preserved the DNA integrity of gill cells, followed by Tris and finally by PBS. In liver, a significant decrease of GST activity was observed at 0.031 mg L-1 (p<0.05), while, at higher concentrations, there were non-significant changes compared to controls. Neither the GR nor the CAT activity was different from control. Gills was the most sensitive organ, showing increased GST activity at 0.31 and 1.53 mg L-1 and GR at 3.06 mg L-1; while CAT activity was inhibited at 0.31, 3.06 and 6.13 mg L-1 (p<0.05). The brain GST activity was increased at 1.53 mg L-1 but inhibited at 3.06 and 6.13 mg L-1, meanwhile GR and CAT activities were inhibited at 1.2, 3.06 and 6.13 mg L-1 (p<0.05). Este composto pode ter uma liberacao lenta e gradativa quando complexado em nanocapsulas de goma/gelatina/quitosana, o que pode limitar sua toxicidade para o ambiente. No entanto, a genotoxicidade do [Cu(EDTA)]-2 ainda nao esta totalmente elucidada. Para avaliar o potencial mutagenico do [Cu(EDTA)]-2, analisamos a frequencia de micronucleos no sangue periferico de camundongos e peixes submetidos a [Cu(EDTA)]-2 livre e nanoencapsulado. A carcaca dos peixes foi avaliada por espectrometria de massa para quantificar o cobre tecidual. Em camundongos a frequencia de micronucleos nos animais expostos ao [Cu(EDTA)]-2 na forma livre foi significativamente maior em relacao aos animais submetidos a forma nanoencapsulada. Em peixes, somente o [Cu(EDTA)]-2 livre foi capaz de induzir aumento na frequencia de micronucleos. A analise espectrometrica demonstrou acumulo de ions cobre no figado nos grupos de peixes estudados, com diferenca significativa em relacao ao grupo controle. Nas brânquias dos peixes nao foram observadas diferencas na concentracao de cobre entre os grupos e ainda, o conteudo de cobre foi significativamente menor em relacao ao encontrado no figado. Pouco se conhece sobre a geoquimica dos sedimentos da regiao de estudo, tornando-se indispensavel para as acoes de monitoramento a determinacao de metais pesados no sedimento. Nesse contexto, foi desenvolvido o estudo dos metais pesados como Cu, Pb, Zn e Cr no sedimento do Saco do Laranjal, porcao estuarina da Laguna dos Patos. A amostragem percorreu seis pontos do local de estudo, nos periodos de inverno e primavera de 2009 e verao de 2010. As amostras foram extraidas em triplicata com agua regia a 50% e acido perclorico e analisadas atraves de espectrometria de absorcao atomica em chama. Neste estudo tambem foram avaliados a materia orgânica por metodo indireto atraves da perda de volateis e, granulometria. Os resultados demonstram baixos indices de contaminacao comparados com o Guia de Qualidade para Sedimentos Estuarinos Canadense, apresentando estreita relacao entre o aumento da materia orgânica no teor de finos (fracao < 63 μm) e a concentraca dos metais. Fontanetti","doi":"10.5132/JBSE.2012.02.003","DOIUrl":"https://doi.org/10.5132/JBSE.2012.02.003","url":null,"abstract":"The comet assay was applied to erythrocytes of fish Oreochromis niloticus (Perciformes, Cichlidae), exposed in the laboratory to water samples taken from the Paraiba do Sul (Brazil), a river that receives effluents from an oil refinery. Water samples were collected at three points: point 1 – upstream from the discharge of effluent from the oil refinery; point 2 – the dump location of the effluent from the oil refinery, and point 3 - downstream from the oil refinery dump. Artesian well water was used as the control. Samples were collected in May and August of 2004 (dry season) and in November of 2004 and January of 2005 (rainy season). The data obtained indicated the presence of genotoxic substances in points 2 and 3 in all the sampling months. We conclude that the final effluent from the oil refinery, even after treatment, can affect the quality of the Paraiba do Sul River. The aim of this work was to establish a standard system to monitor effluents by using juvenile Nile tilapia ( Oreochromis niloticus ) as a bioindicator of genotoxicity, utilizing micronuclei test and comet assay. For this, the fish were exposed during 24, 48, 72 and 240 hours (10 days), to water samples collected at two sites of the Itajai-Acu River: Ilhota and Blumenau, in the Santa Catarina State, Brazil. For positive control the herbicide 2,4-D (75 ppm) was utilized, and the negative control of each fish were the values obtained before the exposure to the river water, termed time zero (T 0 ). Water samples from both sites of the Itajai-Acu River showed significant genotoxic effects in erythrocytes of the exposed fishes. The comet assay was a more sensitive test to detect genotoxic damage in shorter exposure times (24 and 48 hours) than the micronuclei test. Machado","doi":"10.5132/JBSE.2012.02.006","DOIUrl":"https://doi.org/10.5132/JBSE.2012.02.006","url":null,"abstract":"The purpose of this study is to determine the concentrations of total mercury in muscle of Hoplias malabaricus collected in lakes of the North Fluminense and bought in the Municipal Market (MM) since these lakes have suffered from the use of mercury fungicides in sugar cane plantations. The concentrations of mercury were measured by equipment ICP/AES with enhancement of cold vapor. The concentrations of Hg in the lake Campelo (525.6 μg kg -1 ) exceeded the concentrations of other lakes and fish of MM. An inverse correlation was observed between the concentrations of Hg and the standard length, weight and age in samples of fish from the Taquarucu and Campelo lakes. Direct correlation between the concentrations of Hg and biological variables were observed for Feia lake, Cima lake and samples of MM. All the models of regression generated by CCD were highly significant, with R 2 between 77 and 88%, which is the percentual variability in the response that the model can account for. This is indicative of a satisfactory representation of the process models whose data can be used for simulations of response. The maximum shrinkage biovolume presented 28–69% reduction compared to controls. Results from this study suggest that the smaller algal cells amplify metal binding sites, leading to an increased bioaccumulation and a consequential increased capacity to accumulate chromium. Nevertheless, the absorption capacity decreases for more elevated chromium concentrations and for longer exposure. Os objetivos deste trabalho foram avaliar a toxicidade aguda e o risco ambiental e seus efeitos em leucocitos de mato grosso. Os peixes foram expostos a 2,0; 4,0; 6,0 e 8,0 mg L -1 da OTC e um grupo utilizado como controle. Os testes foram conduzidos em um delineamento inteiramente casualizado, com quatro repeticoes e ao final do experimento, foram coletadas amostras de sangue para contagem dos leucocitos. Para a classificacao do risco ambiental foi utilizada a CL 50 e a concentracao ambiental estimada (CAE) do antibiotico. A concentracao letal 50% (CL(I) 50 ;96h) estimada da OTC foi 5,49 mg L -1 com limite inferior de 5,08 mg L -1 e superior de 5,95 mg L -1 . A OTC causa moderado risco de intoxicacao ambiental com a menor CAE e alto risco de intoxicacao ambiental com a maior CAE. Nao ocorreu diferenca significativa na porcentagem de eosinofilos, celula granulocitica especial e basofilos, porem ocorreu linfocitopenia e monocitose em 4 mg L -1 . Assim, a OTC e moderadamente toxica e causa risco de intoxicacao ambiental para o mato grosso e pode causar estresse em 4,0 e imunossupressao em 6,0 mg L -1 . Rocha","doi":"10.5132/JBSE.2012.02.013","DOIUrl":"https://doi.org/10.5132/JBSE.2012.02.013","url":null,"abstract":"In this study, we measured the life history parameters of the tropical cladoceran Pseudosida ramosa at sublethal concentrations of sodium and potassium. The results showed that salinity affected its maximum body length at a concentration of 249 mg L -1 of sodium. There was also an increase in the number of days taken by females to reach maturity at a concentration of 11 mg L -1 of potassium, a reduction in the number of neonates produced by primipara at concentrations ranging from 25 to 249 mg L -1 of sodium, reductions in the survival at concentrations of 249 mg L -1 of sodium and 11 mg L -1 of potassium and reductions in the number of total neonates hatched at concentrations ranging from 41 to 249 mg L -1 of sodium and from 3.6 to 11 mg L -1 of potassium. The concentrations of the both salts that affected the reproduction of P. ramosa were lower than those that affected other endpoints.
