A 70% methanol extract of A. iwayomogi Kitam. was fractionated with a series of solvents in order of increasing polarity; these fractionated extracts showed high 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) scavenging activity in the ethyl acetate fractions and high 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity in both the ethyl acetate and chloroform fractions. Levels of total phenolics and flavonoids were highest in the ethyl acetate fraction extracts. Eight bacteria were then treated with the A. iwayomogi Kitam. extract fractions and most showed similar or lower levels of anti-bacterial activity when compared to the control group, except for Staphylococcus aureus. In addition, the anti-oxidant activities of the ethyl acetate fractions of the A. iwayomogi Kitam. extract were higher than those of the other fractions. To confirm the anti-inflammatory activity of the extract fractions, we applied them to RAW 264.7 cells using LPS, and the results indicate that an ethyl acetate fraction at 250 μg/ml concentration inhibits nitric oxide production. Lee, Minhee Baek, Joon-Ha Lee, In-woo Kim, Jae‐Sam Hwang, Mi-Ae Kim","doi":"10.5352/JLS.2020.30.11.999","DOIUrl":"https://doi.org/10.5352/JLS.2020.30.11.999","url":null,"abstract":"The suppression of neuroinflammatory responses in microglial cells, well known as the main immune cells in the central nervous system (CNS), are considered a key target for improving the progression of neurodegenerative diseases such as Alzheimer’s disease, Parkinson’s disease, and Huntington’s disease. Teleogryllus emma is widely consumed around the world for its broad-spectrum therapeutic effect. In a previous work, we performed transcriptome analysis on T. emma in order to obtain the diversity and activity of its antimicrobial peptides (AMPs). AMPs are found in a variety of species, from microorganisms to mammals. They have received much attention as candidates oftherapeutic drugs for the treatment of inflammation-associated diseases. In this study, we investigated the anti-neuroinflammatory effect of Teleogryllusine (VKWKRLNNNKVLQKIYFVKI-NH2) derived from T. emma on lipopolysaccharide (LPS) induced BV-2 microglia cells. Teleogryllusine significantly inhibited nitric oxide (NO) production without cytotoxicity, and reducing pro-inflammatory enzymes expression such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). In addition, Telegryllusine also inhibited the expression of pro-inflammatory cytokines such as interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-α) through down-regulation of the mitogen-activated protein kinases (MAPKs) and nuclear factor kappa B (NF-κB) signaling pathway. The anti-oxidant activities of DSS were analyzed both before and after fermentation. The total lignan content of the DSS extracts fermented with BCH3678 (1,613.8 mg/l) and BCH3572 (1,599.5 mg/l) were relatively high compared to other strains. Anti-oxidant activity was determined according to phenolic compound and flavonoid content and DPPH radical scavenging rate; the highest total phenolic compound content was provided by the DSS with SRCM103716 at 2,803.3 mg GAE/g which returned total flavonoid content of 1,553.1 mg/g as strong correlation of its anti-oxidant activity. The DSS extract fermented by SRCM103716 at 37°C for 24 hr showed the highest DPPH scavenging rate at 66.5%.
