Primary infections were terminated at various levels with pyrantel embonate (adult infections) or ivermectin (larval infections) and mice were then challenged with 500 infective larvae (L3). The level of protection to secondary challenge positively correlated with the intensity of the primary immunizing infection but truncation of larval infection produced significantly better protection than termination of the adult nematode infection. The duration of the primary larval infection (1–6 days) positively correlated with the level of protection to secondary challenge, antibody responses and the proportion of circulating eosinophils. Histological changes in the gastrointestinal tract, peripheral leucocytic changes and antibody responses of the mice to <jats:italic>H. polygyrus</jats:italic> adult somatic antigens indicate both a cellular and humoral basis of host immunity to secondary challenge. Although the TO mice are slow responders in that they harbour chronic infections, immunization by intramucosal killing of the larval stage produced strong protection against secondary challenge infection. The presence of dead immunogenic larval stages within the intestinal wall may well be an important factor, since it exposes the host to stage specific antigens at an appropriate location. To determine the most efficient method for extracting eggs from experimentally contaminated soil, two consecutive studies were undertaken. Four techniques, including washing, sieving, vacuum, and the one recommended by the World Health Organization, were evaluated. Recovery rates of over 85% were recorded with both washing and sieving methods. Using the washing technique, all combinations of the four pre-treatment solutions, distilled water, acetoacetic solution pH 5, 0.1 N sodium hydroxide and 1% Tween 20, and seven flotation fluids with different specific gravities (S.G.) ranging from 1.20 to 1.35 were assayed. There are four types of secretory bodies in the outer syncytial layer as well as serrated overlapping spines and mitochondria. The subtegumentary ‘cells’ are characterized by the presence of four secretory body types as well as giant bodies which may be involved in the elaboration of the secretory bodies or spine material. The normal configuration of the somatic muscles of <jats:italic>T. imbutiforme</jats:italic> show that the muscular machinery necessary for activity once the larval stage becomes excysted is already in place. The sensory endings are found to be of the uniciliate type occurring in groups of up to eight in raised domes distributed over the body. On each of the 18 farms, 45 adult goats were randomly allocated into three groups of 15 animals each: an untreated control group, a group that was orally administered fenbendazole (10 mg kg<jats:sup>-1</jats:sup> body weight) and a group that received orally a levamisole drench (12 mg kg<jats:sup>-1</jats:sup> body weight). Individual faecal egg counts and pooled larval cultures were done 10 days after anthelmintic treatment. Naive lambs were infected with larvae obtained from control and fenbendazole treated groups and were necropsied 35 days after infection for worm recovery. Faecal egg count reductions (FERC) were calculated for fenbendazole and levamisole and, when less than 95 per 100, were considered as indicative of anthelmintic resistance. An <jats:italic>in vitro</jats:italic> egg hatch test (EHT) was conducted with thiabendazole on eggs isolated from pooled faeces of fenbendazole treated goats in nine farms. Faecal egg count reductions indicated the occurrence of benzimidazole resistance in 15 out of 18 farms. Among these farms, nine had EHT values above 0.1 μg thiabendazole ml<jats:sup>-1</jats:sup> confirming the benzimidazole resistance status. Levamisole resistance was detected in two farms through FECR. Based on necropsy results, the prevalence of benzimidazole resistance was higher in <jats:italic>Trichostrongylus colubriformis</jats:italic>, medium in <jats:italic>Haemonchus contortus</jats:italic> and lower in <jats:italic>Teladorsagia circumcincta</jats:italic>. In nine farms the benzimidazole resistance was monospecific whereas multispecific resistance was found in the six remaining farms. A negative relationship was found between FECR for fenbendazole and the average number of anthelmintic treatments given per year on the farm. Despite extensive management including a low number of treatments, the prevalence of benzimidazole resistance was very high suggesting that the repeated and sometimes exclusive use of benzimidazole drugs, even at low frequency, is probably the main cause in developing nematode resistance in dairy goat herds. Gills of these damselfishes were infected with congeneric Monopisthocotylea Monogenea belonging to the genus <jats:italic>Haliotrema</jats:italic>. <jats:italic>Stegastes nigricans</jats:italic> were found to harbour a guild of three <jats:italic>Haliotrema</jats:italic> species whereas only one species inhabited <jats:italic>D. aruanus</jats:italic>. Microhabitat distribution, inter- and intraspecific competition and interspecific associations on the gill were studied. Observations on site preference revealed no spatial segregation between the three congeneric species inhabiting the gills of <jats:italic>S. nigricans</jats:italic>. Juvenile and adult monogeneans of that guild occurred on the same microhabitat. The dominant species <jats:italic>Haliotrema</jats:italic> sp. 1 did not expand on the microhabitat when the intensity of infection increased. Interspecific association tests revealed positive and negative associations. <jats:italic>Haliotrema</jats:italic> sp. 4 expanded its distribution on the gills of <jats:italic>Dascyllus aruanus</jats:italic> when the intensity of infection increased suggesting the likelihood of intraspecific competition. Juvenile and adult monogeneans of <jats:italic>Haliotrema</jats:italic> sp. 4 appeared to segregate as a result of intraspecific competition. This competition may exist to enhance resource availability when the gill habitat is limited. Overlaps between niche breadth and species microhabitat were revealed for monogenean species inhabiting <jats:italic>S. nigricans</jats:italic>. Interspecific competition did not appear to play an important role in the distribution of <jats:italic>S. nigricans</jats:italic> congeneric ectoparasites. Uninfected control tick survival rate was 70%, but only 45% in the primary infection group. Post-secondary infection survival rate (82%) was comparable to controls, indicating that these selected ticks had some protective advantage. Mean <jats:italic>A. viteae</jats:italic> infective larvae recovery from ticks with secondary infections was 31.4% lower than expected, suggesting the development of immunity. SDS–PAGE of haemolymph for proteins induced post-primary infection yielded a stronger signal at 45 kDa than controls, which was further elevated post-secondary infection. Proteins at 48, 22 and 16 to 18 kDa were detected in haemolymph from infected ticks but not seen from controls. Thirty heifer-calves, aged 3–6 months, were divided into three comparable groups, A, B and C. Each group was turned out on a 1.07 ha paddock (a, b and c). The paddocks were naturally contaminated with infective trichostrongylid larvae from infected cattle grazing the previous year. Fungal material was fed to the animals daily during a two month period starting 3 weeks after turnout. Groups A and B were given 10<jats:sup>6</jats:sup> and 2.5×105 chlamydospores per kg of live weight per day, respectively, while group C served as a non-dosed control group. Every two weeks the heifers were weighed and clinically inspected. On the same dates, faeces, blood and grass samples were collected. From mid-July onwards, the number of infective larvae in grass samples increased markedly (P&lt;0.05) on paddock c, whereas low numbers of infective larvae were observed on paddocks a and b grazed by the fungus treated groups. However, the results indicate that administering fungal spores at a dose of 2.5×105 chlamydospores per kg live weight per day did not significantly prevent parasitism in calves, presumably due to insufficient suppression of developing infective larvae in the faeces. Beginning at proglottid 100 (P100), the lengths and widths of proglottids at 100 proglottid intervals were determined, and the surfaces areas were calculated. The relationships of proglottid length and width to proglottid number were linear, but the relationship of proglottid number to surface area (SA) was exponential. The volumes of proglottids were calculated, and the relationship of volume (V) to proglottid number was exponential. The relationship of surface area to volume ratio (SAVR) to proglottid number was exponential; at the anterior end of the worm (P100), the SAVR was 14.6, while at the posterior end of the worm (P1300) the ratio was 4.2. The helminth fauna comprised 23 species, consisting of two monogeneans, 18 digeneans, one larval cestode, one nematode and one acanthocephalan. Didymozoids, represented by 14 species, were the dominant members of the parasite spectrum. The parasite assemblage is characterized by species richness as well as a high diversity. Most of the parasites found, the didymozoids in particular, were host specialists, exhibiting a high degree of host specificity. The fauna comprised three core species, four satellite species and many secondary species, with no potential for interaction among them but the degree of predictability is high in that each infracommunity comprised two or three core species superimposed by a few secondary species. Many host factors such as the varied diet, high vagility, long life span and endothermy appear to have contributed to the development of the species rich and diverse parasite communities in the tuna. Sherma","doi":"10.1017/s0022149x00701167","DOIUrl":"https://doi.org/10.1017/s0022149x00701167","url":null,"abstract":"High performance thin-layer chromatography (HPTLC) was used to analyse the neutral lipids in the rediae, cercariae, and encysted metacercariae of <jats:italic>Echinostoma caproni</jats:italic> from <jats:italic>Biomphalaria glabrata</jats:italic> snails. Visual observations of the chromatograms showed that the most abundant lipid fraction in all stages was free sterol. Quantification of the free sterol revealed mean weights of 2.7 ± 0.64 ng per redia, 0.53 ± 0.023 ng per cercaria, and 0.081 ± 0.0098 ng per encysted metacercaria. Oil Red O staining of the larval stages confirmed the presence of lipids within the rediae and cercariae but did not show lipids in the encysted metacercariae.
