The study aimed to evaluate the bionomic aspects of chocolate mahseer of Nagaland to validate its status. Methodology: Fish Neolissochilus hexagonolepis were collected from March 2019 to February 2020 from Dikhu River, Nagaland. Regression analysis for morphometric study was conducted using the least square method. Logarithmically transformed total length and weight data were utilized to determine the correlation coefficient 'r', coefficient of determination 'r²', and condition factor for male, female, and pooled groups. Results: The highest correlation was seen in standard length ('r' = 0.9968), and the least correlation was observed in eye diameter ('r' = 0.7769). The length-weight relationship exhibited two growth patterns, namely negative allometric growth, which were represented by females (b = 2.807) and pooled (b = 2.868) population, whereas the population of males displayed positive allometric growth (b = 3.015). The correlation coefficient and coefficient of determination of males, females and the pooled sexes showed a positive correlation with relation to its length and weight ranging from 'r' = 0.970 - 0.981 and 'r²' = 0.941 - 0.961, respectively. The condition factor for all the groups ranged from 1.10 - 1.14. Interpretation: Morphometric traits like standard length, snout length, head length etc. (dependent variable) were analyzed on total length (independent variable), signifying a positive correlation among variables. Length-weight relationship displayed high significance between length and weight parameters. The condition factor for all the groups indicated general well-being of fish condition in their environment. Methodology: Seven wheat genotypes (PBW781, RWP-2018-30, PBW821, RWP-2018-31, WH1239, RWP-2018-26, and RWP-2018-32) along with three check varities namely DBW14, WH730, and RAJ3765 were grown under timely sown (TS) and late sown (LS) conditions in the field research area of Wheat Section, CCS HAU, Hisar (2018-19). For generating heat stress, delay in sowing (4 weeks) was done in late sown from timely sown. These wheat lines were assessed for several physiological and yield attributes under both conditions and one genotype was found to be heat-tolerant under late sown condition. Results: PBW821 was classified as thermo-tolerant due to maximum grain yield, higher normalized difference vegetation index, photosynthetic rate, chlorophyll content, and lower canopy temperature compared to other varieties under late sown conditions. Interpretation: Identification of a heat-resistant wheat genotype would be a valuable resource for developing high-yielding cultivars under high-temperature conditions, and these findings might also be used in breeding programs. Methodology: As people are now completely dependent on gadgets in their daily lives, therefore, the researcher solicited consumers who had purchased eco-friendly gadgets from shopping malls and other electronic stores 450 subjects were selected using \"purposive sampling\". Indian consumers in Chennai, Hyderabad, Bengaluru, and Pondicherry are the subjects of the study. Responses were obtained through the use of a questionnaire. To test the research hypotheses, the study used SPSS 25 and AMOS 21. Results: The results indicate that the negative correlation between green washing and sustainable behaviour was mediated by green perceived risk and green scepticism, and these mediators serially affected green washing and sustainable behaviour. Interpretation: This study suggests that firms should integrate transparency into green products without employing greenwash, which would increase consumer trust in green products and reduce green perceived risk and green scepticism among them. Consumers today are aware of greenwashing and have started to investigate it. This research is crucial to protect both the environment and mankind. Methodology: Extracts of fresh and dried neem leaf were prepared using water at three different temperatures viz. room temperature (26 °C) for 24 hr, cold extraction (4 °C) for 24 hr and hot extraction (60°C) for 15 min, respectively. The preliminary qualitative and high-performance thin liquid chromatography analysis was conducted to estimate the phytoconstituents of each extract. The antiparasitic efficacy and 6 hr median effective concentration (EC50) of each extract was estimated against Argulus sp. Results: The qualitative test revealed all the estimated phytoconstituents in aqueous dried and fresh leaf extract prepared at 26 °C. Whereas the hot extract of dried leaf lacked alkaloids, and similarly among the fresh leaf extract, the absence of saponin in the cold, and hot extract lacked terpenoids and tannin. Further, thin layer chromatography analysis revealed more phytoconstituents in dried leaf than fresh neem leaf extracts at all extraction temperatures. Significantly higher 6 hr antiparasitic efficacy of 100 % and 61 % with an estimated median effective concentration (EC50) of 2.75 and 3.24 were found for dried and fresh neem leaf extracts, respectively, extracted at 26 °C in 3.5 g l-1 compared to cold and hot extracts. Methodology: Fresh as well as roasted (over low flame for 15 min) insect samples were used to determine the microbial contaminants. Crushed insect samples were suspended in sterile half-strength nutrient broth solution and were further serially diluted 10-fold (up to 10-10) in isotonic half-strength nutrient broth solution. The total number of aerobic mesophilic microorganisms were determined on plate count agar and expressed as log Cfu g-1. Results: Differences in the number of microbial colonies were observed in fresh and roasted samples. The microbial load ranged from 6.30-8.75 log Cfu g-1 and showed that the highest microbial colonies were present in the fresh samples. The average log Cfu g-1 in fresh insect samples (7.57±0.87) was significantly higher (P<0.05) as compared to roasted samples (7.07±0.76). Interpretation: Edible insects require proper processing before consumption to reduce microbial contamination and further study is needed to identify specific microbes/food pathogens to develop microbial quality and parameters to ensure consumer safety. Methodology: Locally available agro-industrial wastes were utilized as substrate in five formulations including rice straw alone and in combination with card board (7: 3), waste paper (7: 3), rice husk (17:3) and jack fruit saw dust (7:3). After sterilization substrates were inoculated with mature spawn and placed in cropping room. Results: Total yield, biological efficiency and nutritional contents were found better in the fruiting bodies of Pleurotus ostreatus and Pleurotus djamor cultivated on rice straw as the sole substrate than the mixed substrate formulations. Total colonization period (22.6 days for P. ostreatus and 20.2 days for P. djamor) was quite less for both the oyster mushroom cultivated in rice husk mixed substrate among mixed substrate formulations. Among mixed substrate formulations, highest total yield was obtained from waste paper mixed substrate for Pleurotus ostreatus (713.8 g) and card board mixed substrate for Pleurotus djamor (737.7 g). The highest carbohydrate content in Pleurotus ostreatus and Pleurotus djamor were found in card board mixed substrate. Total protein content was high in fruiting bodies cultivated on saw dust mixed substrate for both the oyster mushrooms. Interpretation: Card board, waste paper, rice husk and saw dust mixed substrates showed relatively high fruiting body yield and nutritional values. It can be concluded that these agro-industrial waste appears to be the promising ingredients in combination with rice straw for cultivation of oyster mushroom. Methodology: A comparative study of PNP’s decolorization (with concomitant removal) in three different test conditions was undertaken. The experiment was carried out in one-liter volume Schott Duran bottles. Genomic DNA was extracted and draft genome sequence was determined using Illumina HiSeqX platform. Raw reads were assembled and subjected to subsystem classification using several bioinformatics tools (RAST, PATRIC, and NCBI’s PGAP pipelines). The genome sequence was deposited at the NCBI Genome database and the strain PNPG3 was also deposited at MTCC, IMTECH, Chandigarh. Results: The bacterial strain PNPG3 could carry out decolorization with concomitant removal of PNP in all three sets of experiments, including one set, where only distilled water was used. The best decolorization (with concomitant PNP removal) capacity was recorded for set D (with Minimal Medium, MM; PNP, and free cells) followed by set E (MM, PNP, and immobilized cells) and set B (distilled water, PNP and free cells) conditions. The size of the draft genome sequence of the strain PNPG3 was 6,566,321 bp, with 62.26% GC contents. The genome had 6210 protein-coding sequences and 66 tRNA genes. The predictive presence of different types of proteases and siderophore receptors indicated its possible potential for industrial applications and plant growth-promoting activities. Interpretation: The bacterium Pseudomonas sp. strain PNPG3 has the capacity to decolorize p-nitrophenol even in presence of distilled water and it remains viable for up to twelve days. The genome sequence revealed that the strain harbored genes responsible for the metabolism of aromatic compounds, chemotaxis, protease, and siderophore receptors indicating the versatile nature of the strain. Metal exposure to fishes of River Ganga is a very common phenomenon due to their ubiquitous nature and through discharge of domestic and industrial waste into the river water. Methodology: An in-vitro experiment was performed to study ill effects of metallic stressors on fish. Fishes were divided into seven groups. Group I served as control, Group II, III, IV as 1/20th ,1/10th and 1/5th of 96 hr-LC50 of Cr6+. Group V, VI and VII were similarly treated with Cu at 1/20th, 1/10th and 1/5th of 96 hr LC50 of Cu+2, respectively. All the treatments were done for 15, 30 and 45 days of exposure period. Genotoxicity was evaluated by micronuclei (MN) induction. Enzymatic and non-enzymatic oxidative stress markers, including reactive oxygen species (ROS), superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), glutathione reductase (GR), and glutathione peroxidase (GPx) were evaluated. Histopathological studies of liver and kidney tissue were also performed. Results: Stress biomarkers were more profound in fishes of chromium treated groups in comparison to copper. Particularly, SOD and CAT showed a significant hike in the activity levels. ROS levels in blood cells increased significantly (p < 0.05) in all the treated groups (Group II, III, and IV) in a dose dependent manner as compared with control (Group I). There was significant induction in micronuclei (MN) frequency in all the treated groups for both the metals. The highest frequency of MN induction was recorded in Group IV after 96 hr of the exposure period.Significant histological alterations were observed in liver and kidney of treated fish, changes were more pronounced in chromium treated fish than copper. Interpretation: Acute exposure of chromium and copper induces ROS and generates oxidative stress mediated genotoxicity and histopathological perturbations in the prime organs of fish.Cr and Cu induces the changes in the production and accumulation of enzymatic antioxidant system for the regulation and scavenging of ROS induced by Cr and Cu. Methodology: Inland saline water (15 ‰ or g l-1) was collected from the salt affected waterlogged areas of village Shajrana (30.3346 0N, 74.1196 0E) in District Fazilka, Punjab (India) and diluted with underground freshwater for preparing different salinity levels (2 to 10 ‰). Fingerlings (10 ±2 cm) of C. carpio were exposed to 0 (control), 2, 4, 6, 8 and 10 ‰ salinity after gradual acclimatization with salinity increase @ 1 ‰ hr-1. Results: At the end of 120 days of rearing under saline conditions, 100% fish survival was observed up to 6 ‰ salinity, while 86.66 and 70.00% survival was recorded in 8 and 10 ‰, respectively. However, fish growth declined significantly at all salinity levels (p≤0.05). Gill structure was also affected at all salinity level, but pronounced changes were observed at salinity levels ≥ 6 ‰, including lamellar oedema, epithelial lifting, lamellar hyperplasia, hypertrophy, lamellar fusion, hyalinisation, aneurysm, blood congestion, etc. Interpretation: The results offer referral database to explore optimised economic production of common carp in inland saline areas of the region at salinity levels below 6 ‰. Singh","doi":"10.22438/jeb/44/4/mrn-3018","DOIUrl":"https://doi.org/10.22438/jeb/44/4/mrn-3018","url":null,"abstract":"Aim: To assess the phytoaccumulation efficiency of arsenic in different parts of Typha tripholia (cattail) grown in the natural habitat of a pond located in Rada village of Bijua block, district Lakhimpur Kheri (U.P.) and to develop an adsorbent based removal technology by activated carbon prepared from Typha tripholia (ACTT) using physical activation. Methodology: Phytoaccumulation of arsenic was studied in cattail plants. Rhizomes and leaves of plants were used to prepare activated carbon. The morphological characterization and surface structure were studied by using different modern techniques like Scanning electron microscopy (SEM), Energy-dispersive X-ray (EDX), and Brunauer-Emmett- Teller (BET). The adsorption efficiency and removal of arsenic were studied by using different doses of ACTT, i.e., 0.5, 1.0, 1.5, 2.0, 2.5 g 100 m l-1 in the known solution of arsenate (30 ppb). Results: The arsenic concentration in pond water was 19 µg l-1, which was higher than the permissible limit (10 µg l-1) of WHO, while in the soil it was 123 µg kg-1. The maximum As concentration in the root was 360 µg kg-1, while in the stem and leaf, it was 20 µg kg-1 and 26 µg kg-1, respectively. Characteristics properties of ACTT showed amorphous nature and it contained 81.71 % carbon while other elements like oxygen (10.65 %), chloride (2.12 %) and potassium (5.52%) were also detected. The porosity of ACTT was 1.271 × 10-3 cc g-1 with pore volume 3.7 × 10-3 cc g-1, while the pore size, pore width, and pore diameter were 4.498 nm, 2.0208 nm and 4.498 nm, respectively. Removal of arsenic increased on increasing the dose of activated carbon, and the maximum adsorption (99.6%) was observed at 1.5 g 100-1 ml further it decreased. Interpretation: Thus, Typha tripholia may be used as phytoremediation, a plant-based green technology for the removal of As from contaminated water while ACTT acts as a good adsorbent for As, due to its large surface area and pore space and a high degree of surface reactivity.
