This study helps us recognize and characterize aerobic bacteria isolated from pyogenic samples and establish antibiotic resistance for clinicians to devise an empirical treatment for patients suffering from similar disease. \u0000Methods: This observational study was carried out from November 2019 to May 2020 in the Department of Microbiology at a teaching hospital in northern India and included all patients who presented with suppurative infections during the study period. \u0000Results: Total 201 culture positive samples were included in our study of which 80 (40%) samples were of sterile body fluids, 51 (25%) samples were of pus aspirates and the other samples included wound pus and surgical site infection samples. Escherichia coli was the most common pathogen (30.34%) followed by Klebsiella pneumoniae (17.91%) and Pseudomonas spp (13.43%).Sixty eight (33.83%) isolates were multidrug resistant. Enterobacter spp was completely resistant to ciprofloxacin, ceftazidime and ceftriaxone, and 25% sensitive to Cefoperazone-Sulbactam, thus showing the presence of extended spectrum beta lactamase (ESBL) character. We studied the association of multidrug resistance to certain risk factors like previous antibiotic therapy, days of wound before admission, increased length of hospital stay, patient comorbidities like diabetes mellitus, heart disease, chronic liver disease and immunosupression. \u0000Conclusions: This study reveals Gram negative bacilli to be more commonly causing pyogenic infections in our patients. Local antibiotic susceptibility profile of these pathogens is essential for empirical treatment.

However, awareness of the prevalence of AmpC β-lactamase producing microorganisms such as E. coli could be very valuable for achieving more accurate epidemiological results, as well as controlling their spread in the different hospital wards. \u0000Methodology: A total of four hundred (400) clinical samples comprising of two hundred and seventy-nine (279) urine samples and one hundred and Twenty-one (121) wound samples were collected from patients in different ward at AE-FUTHA. The clinical samples were analyze using standard microbiological culture and identification of Escherichia coli. Detection of phenotypic AmpC β-lactamases production was performed using Cefoxitin-Cloxacillin Double-Disk Synergy Test (CC-DDST). Antibiotic susceptibility studies of AmpC β-lactamases producing Escherichia coli was determined using the Kirby–Bauer disk diffusion method and the results were construed using the Clinical Laboratory Standard Institute (CLSI) zone diameter breakpoints. \u0000Results: Phenotypic AmpC β-lactamases producing E. coli accounted overall prevalence rate of 94.1%. The frequency of AmpC β-lactamases producing E. coli in urine samples were 57(93.4%) comprising of high occurrence rate in Surgical ward 15(100%) followed by medical ward 24(88.9%) but also accounted for overall prevalence rate of 38(95.0%) in wound samples consisting of 9(100 %) from children ward and medical ward 10(100%) followed by Orthopedic ward 13(86.7%) with the least prevalence rate. The AmpC β-lactamases producing E. coli exhibited high percentage of resistance within the range of 50-100% against ceftriaxone, ceftazidime, cefepime, azetronam, Trimethoprim-Sulfamethoxazole Ticarcillin-clavulanic acid but were susceptible to ofloxacin 70.0%, Imipenem 83.3% and amikacin 100%. \u0000Conclusion: The findings of this study is a proof of the occurrence of AmpC β-lactamase producing Escherichia coli among patients admitted to hospital wards and there’s an urgent need for controlling and managing the development of MDR genotype strain. Moreover, to prevent the spread of resistance among AmpC β-lactamase producing Escherichia coli to other strains and improve the effectiveness of antibiotics, it is suggested to establish a precise schedule for antibiotic use in each region based on their antibiotic resistance pattern.

The aim of this study was to develop antifungal Bacillus cultures for cocoa beans fermentation. \u0000Methods and Results: Antifungal activity of 7 Bacillus isolates was investigated using overlay technique. Solid culture media inoculated with bacterial and fungal isolates were cultivated under aerobic conditions at 30 ºC for 5 days. The Bacillus strain with strong antifungal activity was studied and inoculated to 18 kg box fermentations cocoa beans in Cote d'Ivoire in three time-independent replications each including a spontaneous control fermentation. The comparison of inoculated and spontaneous fermentation processes revealed that the Bacillus thuringiensis strain ATCC 10792 don't affected the fermentation process and cocoa bean quality. The anti-fungal in vivo assays revealed that the Bacillus thuringiensis strain ATCC 10792 completely inhibited growth of all fungi on the surface of cocoa beans. The result showed that sensory evaluation of chocolates is not significant differences for all treatment. The sensory evaluation of control and chocolates made from the inoculated cocoa beans with Bacillus showed excellent sensory quality, demonstrating that Bacillus thuringiensis strain ATCC 10792 did not affect aroma, texture and appearance of chocolate. \u0000Conclusion: From this study, it is revealed that the addition of Bacillus thuringiensis strain ATCC 10792 during fermentation cocoa beans suppress fungal growth and improve the quality of cocoa beans. Therefore, the anti-fungal culture Bacillus thuringiensis strain ATCC 10792 is recommended for future applications and its capacity to limit fungal growth during industrial-scale cocoa bean fermentation should be investigated in further studies.

The enumeration of total heterotrophic bacterial count revealed that the soil impacted with crude oil and kerosene yielded low microbial counts ranged from 3.2 × 106 CFU/g, 3.2 × 105 to 2.6 × 103 CFU/g while that of total coliform count obtained from the samples ranges from 2.6 × 105CFU/g to 2.2 × 103 CFU/g. and total fungal count ranged from 2.0 x 102 CFU/g to 1.4 × 102 CFU/g. The genera of bacteria isolated from soils impacted with crude oil and kerosene belong to the genera: Pseudomonas spp., Bacillus spp., Staphylococcus spp., Streptococcus spp., Escherichia coli, Micrococcus spp., Klebsiella spp., Corynebacterium and Salmonella species while Aspergillus spp., Alternaria spp., Candidia spp., Fusarium spp., Trichoderma spp., Mucor spp., Penicillium spp., Cephalosporium spp., Rhizopus spp. are the isolated fungi. The percentage occurrence revealed that Pseudomonas spp. and Escherichia coli had the highest prevalence of 23.1%, followed by Staphylococcus spp. (7.7%), Salmonella spp. (7.7%), Bacillus spp. (7.7%), Micrococcus spp. (7.7%) etc. While the fungal isolates had the following prevalence; Aspergillus spp. (12.5%), Penicillium spp. (12.5%), Candida spp. (12.5%), Mucor spp. (12.5%), etc. The results obtained indicates that crude oil and kerosene have a negative impact on the microbiota of the soil. The combined effect of the compounds is more harmful than their individual effects. This result implies that farmlands impacted with these products would not be suitable for agricultural purposes since the nitrogen fixing bacteria and other soil enriching microorganisms must have been either killed or inactivated by the toxicity of compounds.

The microbiological evaluation was carried out at the Microbiology Laboratory, Cross River University of Technology, Calabar, Nigeria, using pour plate and spread plate methods to ascertain the viable cell counts. The physicochemical composition, proximate properties were determined using standard methods. The enumeration of total heterotrophic bacterial count revealed low microbial count of 2.6 x 103 CFU/g, total coliform count was 2.5 x 102 CFU/g and total fungal count. 2.4 x 102 CFU/g. The value was within permissible limit of 104 CFU/g. The biochemical characterization and identification revealed Staphylococcus spp, Bacillus spp, Salmonella spp, Escherichia coli, and Micrococcus spp are the suspected bacterial isolates. While the fungal isolates are Aspergillus spp, Fusarium spp, Penicillium spp and Rhizopus spp. The percentage occurrence revealed that Staphylococcus spp had the highest prevalence of 33.3%, followed by Escherichia coli (26.7%), Salmonella spp (20%), Bacillus spp (13.3%), Micrococcus spp (6.7%). While the fungal isolates had the following prevalence; Aspergillus spp (50%), Penicillium spp (16.7%), Rhizopus spp (16.7%) and Fusarium spp (16.7%). Proximate analysis revealed moisture (15.0±0.5), ash (35.0±0.5), crude (17.50±0.25), lipid (22.50±0.03), protein (2.218±0.01), and carbohydrate (7.78±0.01). Results for physicochemical analyses showed that Iodine value was 0.372, saponification value was 22.103 and acid value, 1.159. This result indicates that palm kernel oil, melon oil and coconut oil had the best potential for the production of margarine which would be acceptable due to the low microbial count. However, the presence of Staphylococcus spp and Salmonella spp renders the product a potential health hazard in line with FDA guidelines. Therefore, maximum hygienic conditions during production is advocated and further analysis recommended on these samples to discover their maximum importance and industrial utilization.

Mathematical models predicting sporulation rates and germination yields of heat- and ethanol-activated spores were evaluated as a function of the factors selected. The reliability of these models was assessed by comparing the predicted values with those obtained in real food. \u0000Results: It was found that spores originating from high carbohydrates and proteins media had high sporulating and germination rates. It was found that the origin of the spores, mainly the carbohydrate and protein content, resulted in high sporulation rates and that spores from nutrient-rich media germinated faster than those from nutrient-poor media. Germination efficiency varied from strain to strain depending on the method of activation and the composition of the medium in which the spores were produced. The mathematical models obtained were validated in food matrices with correlation of predicted and observed values higher than 0.85. \u0000Conclusion: The ethanolic activation of spore varies depending on the strain and the nutrient content especially the fat content in sporulating medium. A total of three hundred (300) samples each for stool and blood were subjected to non-invasive techniques. The invasive method used in this study was the polymerase chain reaction method (PCR) targeting the 16SrRNA from stool samples while the non-invasive methods used were stool antigen test, urea breath, and serology test. The amplified 16SrRNA fragments of H. pylori from various samples on agarose gel showed their bands at 380 base pairs. Results obtained from this study showed that the PCR method recorded a 54% prevalence rate, the stool antigen test recorded a 13.7% prevalence rate while serology and urea breath test methods recorded a prevalence rate of 14.7% and 11.7% respectively. This study recorded a prevalence rate of 33.6% based on the study location, with females recording the highest prevalence rate in all the diagnostic methods used. The sensitivities of the various methods used against PCR were 90.3% for the stool antigen test method, 64.5% for the urea breath test, and 100% for the serology test method; while their specificities were 26.3% for both the stool antigen test and urea breathe test and 79% for serology test. The serology method showed a likelihood ratio of 4.76 while that of the stool antigen test and urea breath test were 2.354 and 0.462 respectively. In conclusion, this study has shown that the order of accuracy of diagnostic methods can be arranged as follows: PCR> serology > stool antigen test > urea breath test. However, the order might slightly change among related studies.
