Maisuria","doi":"10.20902/ijctr.2019.130125","DOIUrl":"https://doi.org/10.20902/ijctr.2019.130125","url":null,"abstract":"The present work relates to the complexation reaction between Amlodipine Besylate[AML] with Ni 2+ , Mg 2+ , Co 2+ and Ca 2+ cation in dimethylsulfoxide (DMSO), pure methanol (MeOH) and their binary mixtures(DMSO-MeOH and DMSO-Water) by conductometric method. The conductance data show that the stoichiometry of the complexes formed between AML with Ni 2+ , Mg 2+ , Co 2+ and Ca 2+ cation in pure DMSO, pure MeOHas well as in the binary solvent mixtures was 1:1. The stability of AML complexes with Ni 2+ , Mg 2+ , Co 2+ and Ca 2+ metal ion was observed to be sensitive to the nature of the solvent system. In case of DMSO-Water binary solvent systems there was a linear change in LogKf values but in case of DMSO-MeOH binary solvent systems non linear change in LogKf values observed.The negative values of ΔG 0 show that the reaction is spontaneous and ability of the AML ligand to form stable complexes. However, the result shows positive value of ΔH 0 which indicates that enthalpy is not driving force for the formation of the complexes. Furthermore, the positive value of ΔS 0 indicates that entropy is a driving force for the complexation. The Siddha System of Medicine (Traditional Tamil System of medicine) is the foremost of all other medical systems in the world which provide service to the humanity for more than 5000 years in combating diseases and also in maintaining its physical, mental and moral health. Vitex species were used in siddha for its anti- viral activity for several years. However, the present study deals with the Human Immunodeficiency Virus because of its complexity and killing effects. FTIR analysis of Vitex altissima L and Vitex leucoxylon L revealed the presence of 21 and 17 bioactive compounds respectively. These compounds were analysed further for its binding affinity mechanism against one of the virulence causing protein, reverse transcriptase (target protein) of Human Immunodeficiency Virus (HIV) by using molecular docking and bioinformatics tools. Interaction rate was determined between bioactive compounds against the protein target based on binding free energy requirements. Molecular docking was also made to the commercially available drugs (Zidovudine, Stavudine, and Nevirapine) against the target protein. By comparing the results between bioactive compounds in the Vitex species and the commercially available drugs, it was clear that the bioactive compounds were much more effective than the commercially available drugs, thereby suitable for the treatment of AIDS. This method is based on the development of blue coloured chromogen due to reduction of tungstate and/ or molybdate in Folin-Ciocalteu reagent by Drug in alkaline medium. The coloured species has an absorption maximum at 730 nm, 745 nm and 770 nm and the Beer’s law obeys over the concentration range of 12-72 μg/mL, 10-70 μg/mL, 3-21 μg/mL respectively for the drugs CDP, EVZ, PRZ. The chromatography parameters includes stainless steel column Develosil ODS HG-5 RP C18, 5m, 15cmx4.6mm i.d and acetonitrile: potassium dihydrogen phosphate buffer (0.02M, pH 3.0) (55:45v/v) as mobile phase at a flow rate (1.0 ml/minute). The determinations were performed using UV-Vis detector set at 273 nm. This bioflavonoid is the ―nature‘s biological response modifier‖ as it interferes with the various allergens and other reactive compounds. Apple, oranges, tomatoes, onions, black tea and green tea are good sources of quercetin and it is also available commercially. After absorption in the small intestine and colon, quercetin conjugates with glucuronic acid and binds to albumin and passes to liver and benefits the body by its various bioactivities. Quercetin‘s antioxidant activity enhances the radical scavenging activity and metal chelation of the ions but the prooxidant activity depends on its high concentration. Further, quercetin interferes with the formation of leukotrienes from arachidonic acid showing its anti-inflammatory effect. A combined effect of quercetin and bromelain effectively suppresses the allergic reactions and the excessive inflammation resulting from bruising and tissue damage. The mutualistic effect of vitamin C and quercetin protects each other from getting oxidized. A direct relationship was also found to exist between quercetin's antiviral activity and enhancement of cyclic adenosine monophosphate (cAMP), which is a second messenger involved in many biological processes. Quercetin helps in down regulation of mutant gene p53 and inhibits the growth of cancerous cells by putting a check at G1 phase. This also controls the surpassing of the normal regulatory growth by the tumor cells and inhibits the production of heat shock proteins and thus showing its anticancer properties. Owing to the potential pharmaceutical properties of quercetin, the bioactivities, principle uses and mechanisms involved in the treatment of various diseases were reviewed in this paper. Aquatic plants contain some organic compounds that provide definite physiological actions on the human body. In the present study deals with the synergistic effect of Trisodium citrate (TSC)on the inhibition efficiency of methanolic extract of Sargassum swartzii (SS) and Zn 2+ system on mild steel in aqueous environment. The corrosion monitoring techniques adopted in the study were weight-loss methods, electrochemical measurements, FT-IR and surface morphological techniques namely SEM and EDX. It has proved the formulation consisting of 100 ppm of Zn 2+ , 120 ppm of SS and 100 ppm of TSC has 95.77% IE. The high performance of SS Zn 2+ TSC system could be due to synergistic effect that covers a wide surface coverage area on the metal surface and thus retarding the corrosion. Polarization study reveals that this formulation functions as a mixed inhibitor. AC impedance spectra suggest the presence of protective film on the metal surface. FT-IR spectra reveals that the protective film consists of Fe 2+ -SS complex, Fe 2+ -TSC complex and Zn (OH)2. SEM and EDX provide a pictorial representation on the nature of surface film in the absence and presence of inhibitors. The proposed RP-HPLC method uses X terra C18, 4.6 mm, 150 mm 4.6 mm i.d. column (at room temperature), using 0.1% octa-sulfonic acid: acetonitrile 30: 70 v / v, effluent flow rate (1.0 ml / min) and UV detection at 237 nm for oseltamivir analysis. The method was validated according to the ICH guidelines in terms of specificity, linearity, precision and accuracy. The retention time for oseltamivir was 2.31 min. The recovery determinations allowed the calculation of a confidence interval from 99.79 to 101.30% with a relative standard deviation value of 0.5%. LOD and LOQ were estimated at 2.98 and 9.98 µg/mL respectively. The validated method was successfully applied to the determination of oseltamivir in dosage form in capsules (Tamiflu 75 mg, Roche). Oseltamivir was exposed to conditions of acid, basic, oxidative and thermal stress and the stressed samples were analyzed with the proposed method. The chromatographic peak purity results indicated the absence of elution peaks with the main oseltamivir peak, which demonstrated the specificity of the test method for estimating oseltamivir in the presence of degradation products. The complexes of NiL2and CoL2were synthesized by using template condensation synthesis method and characterized by melting point, solubility, elemental analysis, and molar conductance, and magnetic susceptibility, infrared and electronic spectral studies. The complexes, NiL2and CoL2 are soluble in ethanol, partially soluble in Diethyl ether and chloroform and insoluble in hexane and petroleum ether. The complexes, NiL2and CoL2 neither melt nor decompose up to 420 0 C. The molar conductance of NiL2and CoL2 was 42 Scm 2 /mol and 46Scm 2 /mol in respectively. The molar magnetic susceptibility of two complexes was 1.74 BM for NiL2 and 2.76 BM for CoL2. The metal to ligand ratio of both metal complexes was 1:2; both metal complexes are non-electrolytes in ethanol and are paramagnetic at 21 0 C.
