In total 3000 NMRI mice 2PN embryos were divided into six groups (<i>n</i> = 500). The test group was exposed to 20, 40, 80, 160 or 320 μg/ml of CSC. In the control group, CSC was not added to the culture medium during the development of 2PN embryos. The effects of 20 and 80 μg/ml of CSC on genes involved in pluripotency and apoptosis, and also, the aryl hydrocarbon receptor gene was assessed in the blastocysts. Our results showed that CSC had an adverse effect on the viability of mouse embryos at the concentrations of 80, 160 and 320 μg/ml compared with the control group (<i>P</i> < 0.05). In contrast, it had positive effects on the viability of mouse embryos at the concentrations of 20 and 40 μg/ml compared with the control group (<i>P</i> < 0.05). The 20 and 80 μg/ml concentrations of CSC increased the expression of pluripotency, apoptotic, and aryl hydrocarbon receptor genes in the blastocyst embryo stage compared with the control group (<i>P</i> < 0.05). It can be concluded that concentrations higher than 40 μg/ml of CSC have an adverse effect on mouse embryo development in the preimplantation stages. The main mechanism involved in the maintenance of telomeres is based on telomerase activity, a specialized enzyme complex, which is capable of adding TTAGGG repeats at the ends of the chromosomes. Mitochondrial dysfunction may cause progressive shortening of telomeres by promoting the generation of reactive oxygen species. Mitofusin-1 is a protein required for mitochondrial fusion. Mice with the mitofusin-1 (<i>Mfn1</i>) deletion in the oocyte are characterized by accelerated follicular depletion and infertility, associated with defective oocyte maturation and follicular development. We hypothesized whether mitochondrial dysfunction in oocytes with targeted deletion of <i>Mfn1</i> causes telomere shortening. We analyzed telomere length in oocyte and somatic cells in 3-, 6- and 9-month-old <i>Mfn1</i><sup>-/-</sup> and wild-type mice. Immunofluorescence in oocyte mice of TRF1 and H2A.X was assessed to evaluate the interplay between the end-protection functions and the response to DNA damage occurring inside the telomeric repeats. The present study was conducted to use a Bayesian approach for estimation of genetic parameters of production and reproduction traits in Jersey crossbred cattle. Data of Jersey crossbred cattle maintained at Eastern Regional Station, National Dairy Research Institute, West Bengal spread over a span of 41 years were utilized. The marginal posterior medians of heritability for 305-day milk yield (305MY), total milk yield (TMY), peak yield (PY), lactation length (LL), calving interval (CI), total milk yield per day of lactation length (TMY/LL) and total milk yield per day of calving interval (TMY/CI) were 0.31 ± 0.07, 0.29 ± 0.07, 0.27 ± 0.06, 0.16 ± 0.05, 0.15 ± 0.05, 0.29 ± 0.06, 0.27 ± 0.06, respectively. Moderate heritability estimates for 305MY, TMY, PY and production efficiency traits indicate the presence of adequate additive genetic variance in these traits to respond to selection combined with better herd management. Repeatability estimates for 305MY, TMY, PY, LL, CI, TMY/LL and TMY/CI were 0.57 ± 0.08, 0.58 ± 0.08, 0.51 ± 0.07, 0.34 ± 0.06, 0.31 ± 0.06, 0.54 ± 0.07 and 0.49 ± 0.07, respectively. Repeatability estimates for 305MY, TMY and PY were high in the current study, suggesting the use of first lactation records for early evaluation of Jersey crossbred cattle for future selection. Genetic correlations varied from 0.21 to 0.97 and maximum genetic correlation was observed between 305MY and TMY indicating that consideration of 305MY instead of TMY in breeding programmes would suffice. One option is <i>in vitro</i> maturation (IVM) of the immature oocytes as there is not enough time for induction of an ovarian stimulation protocol. The aim was to compare the IVM laboratory outcomes between stimulated and unstimulated (natural) <i>in vitro</i> fertilization (IVF) cycles. In total, 234 immature oocytes collected from 15 cancer patients who underwent an IVM programme (natural IVM) and 23 IVF cycles with a controlled ovarian hyperstimulation protocol (stimulated IVM) were analyzed. The oocyte morphology, zona pellucida (ZP), and meiotic spindle presence were measured using PolScope technology. Also, the rates of oocyte maturation and fertilization were assessed in both groups. The IVM rate was higher in the stimulated cycle (<i>P</i> < 0.05), but the fertilization rate was insignificant in comparison with unstimulated cycles. There were no significant differences in the spindle visualization and ZP birefringence scoring between the groups (<i>P</i> > 0.05). The oocyte normal morphology was better in the stimulated cycle compared with the natural cycle (<i>P</i> < 0.05). The aim of this study was to evaluate the most effective and cost-effective method to reduce the percentage of spermatozoa with defects in chromatin decondensation for use in assisted reproductive technologies (ART) procedures. Sperm samples from 15 sub-fertile males were examined at CFA Naples to determine the sperm decondensation index (SDI), using the aniline blue test, before and after preparation, comparing density gradients with two different swim-up approaches. All three techniques led to a reduction in decondensed spermatozoa with no statistical difference (<i>P</i> > 0.05) between the control and the treated sperm. In contrast, we found a highly significant decrease in SDI (<i>P</i> < 0.01) after the two swim-up methods in all the samples, confirming the efficacy of these methods in lowering the percentage of chromatin compaction damage. There was no statistical difference between the two swim-up methods, however swim-up from the pellet led to improved count, motility and the percentage of normal condensed spermatozoa. Despite multiple attempts, there are no effective tools against these conditions; however, mesenchymal stem cell-based therapy, especially using adipose tissue, has attracted much attention in medicine in light of its advantages such as easy isolation and accessibility. Conversely, it has been suggested that MSC-conditioned medium (CM) can restore injured tissues and has high immunocompatibility. It is also used for research on reproduction, fertility and development. However, IVF embryos are still inferior to their <i>in vivo</i> counterparts. Some substances in seminal plasma appear to have important roles in embryo development, and during the traditional IVF procedure, the seminal plasma is washed away. In this study, extracellular vesicles (EVs) were concentrated from seminal plasma by ultracentrifugation, visualized using transmission electron microscopy, and particle size distributions and concentrations were determined with a NanoSight particle analyzer. We found particles of various sizes in the seminal plasma, the majority having diameters ranging from 100 to 200 nm and concentrations of 6.07 × 10<sup>10</sup> ± 2.91 × 10<sup>9</sup> particles/ml. Addition of seminal plasma EVs (SP-EVs) to the IVF medium with mouse oocytes and sperm significantly increased the rate of blastocyst formation and the inner cell mass (ICM)/trophectoderm (TE) cell ratio, and reduced the apoptosis of blastocysts. Clinical data collected between February 2013 and August 2017 were included in this retrospective study. Cases were classified into five groups by the duration of cryopreservation: Group 1, 6-12 months; Group 2, 13-36 months; Group 3, 37-60 months; Group 4, 61-84 months; and Group 5, >84 months. The embryo survival rate, implantation rate, clinical pregnancy rate, live-birth rate, newborn sex ratio, singleton gestational age, singleton birth weight and malformation rate were compared between the groups. The cryopreservation duration did not significantly affect the rates of clinical pregnancy (<i>P</i> = 0.119) and live birth (<i>P</i> = 0.354), the newborn sex ratio (<i>P</i> = 0.614) or the singleton gestational age (<i>P</i> = 0.212) and birthweight (<i>P</i> = 0.212). Although decreases in the embryo survival and implantation rates were observed in groups 4 and 5 compared with those in groups 1-3, these differences were not statistically significant (<i>P</i> = 0.329, <i>P</i> = 0.279, respectively). These cave-adapted morphs have evolved independently from two different lineages (i.e. 'old' and 'new' lineages); therefore, this model system gives a unique opportunity to explore parallel adaptive evolution in biological traits. The present study corresponds to the first morphological description of the <i>Astyanax mexicanus</i> maturation process of the spermatozoa and oocytes, using thermal and hormonal stimuli to promote spermatogenesis and oogenesis, considering surface and cave morphs from both lineages. We corroborate the relevance of thermal and hormonal stimuli to promote gamete maturation. The hormone Ovaprim (GnRHa + Domperidone) is an effective promoter of ovarian development, maturation end in oocytes and spawning in <i>Astyanax mexicanus</i>. The sperm morphology of <i>Astyanax mexicanus</i> includes the sperm head, the midpiece, and tail or flagellum. We found differences in the spermatozoan total length between environments (<i>F</i> = 9.929, <i>P</i> = 0.05) and linages (<i>F</i> = 49.86, <i>P</i> = 0.005). The oocytes showed a spherical conformation with a mean diameter of 822.4 ± 194.1 μm for the surface populations, and 604.6 ± 38.3 µm for the cave populations. The oocyte chorion presents ridges and grooves that are arranged radially towards the micropyle. A plug in the micropyle zone was observed after fertilization, confirmed by the outer membrane of the chorion, which provides some weak adhesiveness to the substrate. We observed differences in chorion thickness between the contrasting environmental conditions. Bovine H1foo has been identified, and its expression profile throughout oocyte maturation and early embryo development has been established. However, it has not been confirmed if H1foo is indispensable during bovine oocyte maturation. Effective siRNAs against H1foo were screened in HeLa cells, and then siRNA was microinjected into bovine oocytes to down-regulate H1foo expression. H1foo overexpression was achieved via mRNA injection. Reverse transcription polymerase chain reaction (RT-PCR) results indicated that H1foo was up-regulated by 200% and down-regulated by 70%. Based on the first polar body extrusion (PB1E) rate, H1foo overexpression apparently promoted meiotic progression. The knockdown of H1foo significantly impaired bovine oocyte maturation compared with H1foo overexpression and control groups (H1foo overexpression = 88.7%, H1foo siRNA = 41.2%, control = 71.2%; P < 0.05). This decrease can be rescued by co-injection of a modified H1foo mRNA that has escaped from the siRNA target. However, the H1e (somatic linker histone) overexpression had no effect on PB1E rate when compared with the control group.
