Compared with single pregnancy, multiple pregnancy shows a high incidence of maternal complications and premature deliveries and is an important problem in perinatal medicine [2]. Among multiple pregnancies, super twins (triplets or more) and monochorionic multiple pregnancy have particularly high risks. Super twins have been conventionally managed by limiting the number of transferred embryos. After introduction of blastocyst transfer with high selectivity of good embryos, the incidence of monochorionic multiple pregnancy has been increasing. This suggests that the peritoneal environment that contains various free-fl oating cells such as macrophages and endometrial cells derived from refl uxed menstrual blood may contribute to the development of endometriosis. We previously reported that the proportion of active macrophages was elevated in ascites from patients with endometriosis, and in another study, activated macrophages reportedly released several cytokines into the peritoneal cavity. This study focused on the interactions between macrophages and endometrial stromal cells (ESCs) in the development of endometriosis. Methods: The phenotype of macrophages in the ascites from patients with endometriosis was determined by immunocytochemistry. Soluble factors in ascites and conditioned medium from human macrophages and ESCs in co-culture system were quantified by the Cytokine Array Kit. Next, we investigated the significance of macrophages for proliferation and Stat3 activation of ESCs by using co-cultured cells. Furthermore, pSTAT3 expression of peritoneal foci of endometriosis was assessed by immunohistochemistry. Finally, the effects of corosolic acid (CA), a triterpenoid compound, on proliferation and Stat3 activation of ESCs in co-cultures were evaluated. Results: The total number of CD163+ macrophages (M2 macrophages) in ascites from the endometriosis group was signifi cantly higher than that of the control group. The cytokine array study detected that the production of GM-CSF, RANTES, IL-1 receptor antagonist, and MCP-1 were up-regulated in ascites from patients with endometriosis and in conditioned medium from co-cultured cells (c-CM). Coculture with M2 macrophages signifi cantly up-regulated ESC proliferation and Stat3 activation in ESCs in vitro. Immunohistochemically, Stat3 was activated in both epithelial and stromal cells in endometriotic lesions. When the inhibitory eff ect of a Stat3 inhibitor, CA on ESC was investigated, CA inhibited ESC proliferation and Stat3 activation induced by c-CM. Conclusions: These fi ndings indicated that the interactions between M2 macrophages and ESCs via Stat3 activation may play indispensable roles in the development of endometriosis. There are some reports on fertilization and embryonic developmental ability of spermatid. Deficiency of fertilization or developmental arrest frequently appears due to the ability. Functional immaturity is also observed in spite of abnormality of karyotype in spermatids. However, a functional difference between spermatids and spermatozoa is not clarified. In this study, fertilization and developmental ability of the spermatid were examined by the microinjection using immature spermatid including the round spermatid, elongating spermatid, elongated spermatid and intratesticular spermatozoa derived from three different mice strains; ddY, ICR and B6D2F1. Mature female mice were superovulated with PMSG and hCG. Cumulus-oosytes complex were obtained from their oviducts, and cumulus cells were removed from MII oocytes by hyaluronidase and pippeting. Spermatids were collected from mature male testis, and were injected into MII oocytes using a micromanipulator. Manipulated embryos were cultured in CZB medium, and their ability for development was examined. Epididymal spermatozoa from the three strains were used as control group. The fertilization rate was correlated with the spermatogenic stage. In embryonic developmental rate, on the other hand, there were no significant difference between the control and experimental embryos. The data however showed that elongating, elongated and round spermatid slightly deteriorated the embryonic development. Muramatsu","doi":"10.3192/JSIRIB.21.64","DOIUrl":"https://doi.org/10.3192/JSIRIB.21.64","url":null,"abstract":"Midkine (MK) is a heparin-binding growth factor, the expression of which is strong in the midgestation period, is highly restricted in the adult, and is increased in many human carcinomas. In the present investigation, we determined serum levels of MK by an enzyme immunoassay, and found that in many patients with gynecological tumors, the values increased above the cut off value of 500 pg/ml. All control subjects (n=49) including pregnant women exhibited values not more than 500 pg/ml. The percentage of positive cases was as follows; ovarian carcinoma (n=15), 87%; uterine cervical carcinoma (n=18), 67%; uterine endometrial carcinoma (n=10), 50%; ovarian cyst (n=25), 72%; leiomyoma (n=29), 72%. CA-125 and MK levels were not correlated in ovarian carcinoma and ovarian cyst; a combination of the two assays may be helpful in increasing specificity or sensitivity. Furthermore, increased MK levels were found even in patients with early stages of uterine cervical and endometrial carcinomas.
