Penelitian ini bertujuan untuk mengidentifikasi dan menganalisis keanekaragaman spesies kupu-kupu yang terdapat di kampus Universitas Sam Ratulangi. Pengambilan sampel dilakukan dari bulan Februari sampai Juni 2018 di kampus Univesitas Sam Ratulangi, Manado. Metode penelitian yang digunakan yaitu metode survei dengan pengambilan sampel secara purporsif. Teknik pengambilan sampel dilakukan dengan menggunakan teknik sweeping yang diterapkan secara acak sepanjang 150 m. Hasil penelitian menunjukan bahwa kupu-kupu di kampus Universitas Sam Ratulangi ada 29 spesies yang termasuk dalam 5 famili yaitu Papilionidae, Nymphalidae, Pieridae, Lycaenidae dan Hesperidae. Spesies kupu-kupu yang paling banyak ditemukan yaitu Zizina otis. Famili yang banyak ditemukan yaitu famili Nymphalidae dengan jumlah spesies sebanyak 10 spesies.Kata Kunci: keanekaragaman, kupu-kupu, Nymphalidae, Universitas Sam Ratulangi Abstract Butterflies are one type of insect from the Order of Lepidoptera which has a variety of combinations of colors and as one of the pollinators in the process of flower fertilization. This research aims to identify and analyze butterfly species diversity that exist within Sam Ratulangi University. The sampling was taken from February until June 2018 at Sam Ratulangi University, Manado. The research method was survey through purposive sampling method. The sampling was conducted by using sweeping technique that was performed randomly as long as 150 m. The results of the research shows that there are 29 species of butterflies that exist within the area of Sam Ratulangi University and they are categorized into 5 families, they are: Papilionidae, Nymphalidae, Pieridae, Lycaenidae and Hesperidae. The most common butterfly species is Zizina otis. Polietilen glikol (PEG) mampu menurunkan potensial air dalam larutan, sehingga dapat digunakan untuk menginduksi kekeringan. Penelitian ini bertujuan untuk mengkaji ada tidaknya perbedaan respons fisiologis berdasarkan kandungan air daun pada tanaman padi lokal Sulawesi Utara yang mengalami kekeringan dengan induksi PEG 8000. Hasil penelitian ini menunjukkan bahwa faktor varietas (Superwin, Burungan, Ombong, dan Temo), faktor konsentrasi PEG 8000 (potensial air medium 0, –0,25 dan –0,5 MPa), faktor waktu perlakuan (0, 6 dan 12 jam), interaksi antara faktor varietas dan konsentrasi PEG, interaksi antara faktor varietas dan waktu perlakuan, interaksi antara faktor konsentrasi dan waktu perlakuan, interaksi antara faktor varietas, konsentrasi PEG, dan waktu perlakuan dalam cekaman kekeringan tidak menyebabkan perbedaan kandungan air daun yang nyata. Dalam penelitian ini kandungan air daun tidak dapat dijadikan sebagai indikator respons fisiologis pada keempat varietas padi lokal Sulawesi Utara terhadap cekaman kekeringan. Polyethylene glycol (PEG) decreases the water potential in the nutrient solution, therefore, it can be used to induce drought. The purpose of this research was to evaluate physiological response of North Sulawesi local rice under PEG 8000-induced drought based on the leaf water content. The results of this study indicated that varieties factor (Superwin, Ombong, Burungan and Temo), the factor of PEG 8000 concentration (medium water potential -0.25 and -0.5 MPa), the treatment period factor (0, 6 and 12 hours), the interaction between varieties and PEG 8000 concentration, interaction between varieties and treatment period, the interaction between PEG 8000 concentration and treatment period, interaction among varieties, PEG 8000 concentration and treatment period under drought did not result in significant differences in leaf water content. Penelitian bertujuan mengisolasi kedua gen tersebut pada tanaman Pandanus sp asal Riau. Metode penelitian meliputi isolasi DNA total, elektroforesis, PCR, purifikasi gel, sekuensing, dan analisis data sekuen DNA menggunakan program BioEdit version 7.0.0, BLASTn, dan MEGA6. Sekuen DNA penyandi 18S rRNA (420 bp) dan ubiquitin (493 bp) telah diperoleh. Sekuen 18S rRNA Pandanus sp memiliki kemiripan sebesar 99% dengan beberapa spesies Pandanus. Sekuen ubiquitin Pandanus sp memiliki kemiripan sebesar 87%-89% dengan sekuen ubiquitin dari beberapa tanaman dan alga hijau. Dendrogram yang diturunkan dari kedua sekuen juga mendukung hasil tersebut bahwa sekuen 18S rRNA dari Pandanus sp. membentuk satu kelompok dengan sesama sekuen 18S rRNA, begitu pula dengan sekuen ubiquitin Pandanus sp. membentuk satu kelompok dengan sesama sekuen ubiquitin. Kedua gen berpotensi dijadikan kontrol internal setelah dilakukan validasi.Kata kunci: RNA ribosom 18S, housekeeping gene, Pandanus sp., Riau, ubiquitin. Abstrak This research aimed to isolate genes encoding 18S rRNA and ubiquitin in Pandanus sp. from Riau. The research method included total DNA extraction, polymerase chain reaction, electrophoresis, gel purification, sequencing, and DNA sequence analysis using BioEdit version 7.0.0, BLASTn, and MEGA6 programs. Sequences encoding 18S rRNA (420 bp) and ubiquitin (493 bp) had been obtained. The BLASTn analysis showed that 18S rRNA of Pandanus sp. had 99% similarity to some species of Pandanus. Ubiquitin sequence in Pandanus sp. had 87%-89% similarity to ubiquitin sequences of some plants and green algae. Dendrograms generated from both sequences also supported the results that 18S rRNA sequence of Pandanus sp. was the same group as 18S rRNA on Pandanus genus. Similarly, ubiquitin sequence of Pandanus sp was the same cluster as other plants. Kehadiran logam-logam berat di air dan sedimen dapat menurunkan kualitas air sungai. Penelitian ini bertujuan menganalisis kehadiran logam-logam berat timbal (Pb), kadmium (Cd), tembaga (Cu) dan seng (Zn) di air dan sedimen Sungai Lowatag. Penelitian ini menggunakan metode purposive sampling pada tiga stasiun yang tersebar dari hulu ke hilir. Pengambilan sampel sedimen dilakukan secara komposit di tiap stasiun. Sampel air diulang sebanyak tiga kali di tiap stasiun. Kehadiran logam berat-logam berat pada sedimen dengan konsentrasi berbeda-beda yaitu Cu berturut-turut dari Stasiun I, II dan III yaitu 13; 17 dan 13 mg/kg dan Zn yaitu 73; 48 dan 56 mg/kg. Kehadiran logam berat Pb di sedimen dari Stasiun I, II dan III yaitu 28; 19,1 dan 28,2 mg/kg masih di bawah Nilai Ambang Batas (NAB) menurut RNO/1981 sebesar 10 mg/kg – 70 mg/kg. Kehadiran logam berat Cd berturut-turut 3,83; 2,69 dan 4,03 mg/kg telah melampaui NAB menurut RNO/1981 (0,1 mg/kg – 2,0 mg/kg). Kehadiran logam-logam berat Pb, Cd, Cu, dan Zn di air masih di bawah NAB menurut PP No.82/2001. Kata-kata kunci: logam berat, kualitas air, sedimen, Sungai Lowatag, Sulawesi Utara AbstractThe qualified river water is required by organisms for their sustainability. The occurrence of heavy metals in surface water and sediments can degrade the quality of river water. The aim of this study was to analyze the occurrence of lead metals (Pb), cadmium (Cd), copper (Cu) and zinc (Zn) in water and sediment of Lowatag River. The purposive sampling method was applied to survey three stations from upstream to downstream. Sediment sampling was collected compositely at each station. Water samples were collected three times in each station. The occurrence of heavy metals in the surface sediments with different concentrations of Cu were 13; 17 and 13 mg /kg and Zn were 73; 48 and 56 mg/kg respectively from Station I, II and III. The occurrence of heavy metal Pb in the sediments of Stations I (28 mg/kg), II (19.1 mg/kg) and III (28.2 mg/kg) were below the Threshold Limit Value (TLV) according to Du Reseau National D’ Observation (RNO) year 1981 (10 mg/kg-70 mg/kg). The occurrence of heavy metal Cd in the sediments of Stations I (3.83 mg/kg), II (2.69 mg/kg) and III (4.03 mg/kg) were exceeded TLV according to RNO/1981 (0.1 mg/kg-2.0 mg/kg). Stress oksidatif ini disebabkan karena radikal bebas berlebih. Tubuh memerlukan antioksidan untuk mengurangi pengaruh radikal bebas dan meredam dampak negatifnya. Penelitian ini bertujuan untuk mengevaluasi daya antioksidan ekstrak kulit buah jamblang (Syzigium cumini (L.) Skeels) dengan menggunakan metode DPPH. Berdasarkan hasil pengujian daya antioksidan ekstrak kulit buah jamblang diketahui bahwa ekstrak kulit buah jamblang memiliki kandungan antioksidan yang tergolong aktivitas sedang dengan nilai IC50 sebesar 169.3. Hasil penelitian ini menunjukkan bahwa kulit buah jamblang berpotensi untuk dikembangkan sebagai sumber antioksidan alami bagi manusia.Kata Kunci : antioksidan, DPPH, ekstraksi, jamblang, radikal bebas, stress oksidatif AbstractOxidative stress can induce many diseases. Oxidative stress is caused by excessive free radicals in the body. The body required antioxidant to decrease and hush the negative effects of free radicals. The aim of this study was to evaluate the antioxidant potency of extract rind of jamblang (Syzigium cumini (L.) Skeels) using DPPH method. The result showed that the jamblang rind extract had moderate antioxidant activity and the IC50 value was 169.3.
