This zooplankton is widely favored by marine fauna larvae because of its small size can fits well with various larval mouth; thus, it is easily preyed by larvae. This study aimed to determine the variation of rotifer B. rotundiformismorphometry if cultured with different feed at 40 ppt. The use of 40 ppt salinity is expected to provide a variable morphometric size because B. rotundiformishas a polymorphism property. Microalgae used as feed for rotifer B. rotundiformiswere Prochloronsp. and Nanochloropsis oculata. Microalgae were cultured with Hirata medium. In the early stages, B. rotundiformiswas cultured at optimum temperature (28 ºC) and salinity 20 ppt, then it was cultured at salinity 40 ppt. Salinity adaptation was done by raising the salinity of the medium by 2 ppt every two days in a 10 ml reaction tube containing 10 individuals. After adaptation, B. rotundiformiswas transferred in a 1000 ml container with a density of 50 individuals. For the morphometric aspect, the total length, the length of the lorica, the width of the lorica and the anterior width were measured. The result showed the morphometric of rotiferB. rotundiformisfed with microalgae Prochloronsp. at 40 ppt salinity was smaller than that of the rotifer fed with N. oculata. Based on that finding it can be concluded that B. rotundiformis fed with Prochloronsp. at a salinity of 40 ppt has the potential to be developed as feed for fish larvae. Further investigations on how to accelerate the cultivation of microalgae Prochloronsp. as feeding for B. rotundiformisare needed.RotiferBrachionus rotundiformismerupakan golongan zooplankton yang digunakan sebagai makanan bagi larva ikan. Zooplankton ini banyak disukai oleh larva fauna laut, karena ukurannya kecil yang cocok dengan berbagai bukaan mulut larva. Penelitian ini bertujuan untuk mengetahui variasi morfometri rotifer B. rotundiformis, jika dikultur pada salinitas yang tinggi (40 ppt) dengan pemberian pakan berbeda. Penggunaan salinitas 40 ppt diharapkan bisa memberikan ukuran morfometrik yang bervariasi, karena rotifer jenis ini memiliki sifat polimorfisme. Therefore, extraction procedure that successfully works in one species of algae mayfail for another type of algae. The present study was aimed to examine several DNA extraction techniquesand rbcL (ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit)gene amplifications of Gracilaria sp. collected inBahoi, North Minahasa (126043’48’’N 12501’33”E). DNA genom of Gracilariasp. was extracted using conventional method (CTAB, Cetyltrimethyl ammonium Bromide), and commercial extraction kits (innuPrep Plant DNA Kit and Geneaid Genomic Plant Mini Kit). Amplification of rbcLgene employed 2 primers (rbcL-aF; ATGTCACCACAAACAGAGACTA AAGC, rbcL-aR; GTAAAATC-AAGT CCACCRCG, and rbcL-1F ATGTCACCACAAACAGAAAC, rbcL-724R TCGCATGTA-CC TGCAGTAGC under 2 different annealing temperatures (45 and 500C). Genomic DNA of Gracilariasp. was successfully extracted using Geneaid DNA Mini Kit (Plant) indicated by a DNA band on the agarose gel. RbcLgene of Gracilaria sp. could be amplified using primer 1F-724R and annealing temperature at 500C indicated bya sharp DNA band at 300-400 bp (1kb marker, Solis Biodyne) as a partial amplification of the target gene.Kualitas hasil ekstraksi DNA dan amplifikasi gen pada alga dipengaruhi oleh beberapa faktor diantaranya adalah karakter dan komponen penyususun dinding sel alga itu sendiri. Oleh karena itu, prosedur ekstraksi yang berhasil dilakukan pada pada satu jenis alga dapat saja gagal dilakukan untuk jenis alga lainnya. Carrageenan is a polysaccharide extracted from seaweed or some species of red algae (Rhodophyceae). Seaweed growth is strongly influenced by two factors: internal and external factors. But twthat determine the success of the seaweed growth is the management carried out by people working on it. Banggai Regency is one of the largest seaweed production centers in Central Sulawesi. The main objective of this studyis toexamine the potential of seaweed cultivation (Kappaphycus avarezii) by looking at the growth and the carrageenan, inBanggai waters, Central SulawesiProvince. The temperature range obtained during this study r25to 31ºC. The results of carrageenananaliysis wasvery different due to differences inlocation, showed by content. The highest and lowest ashcontentwere obtained from two locations, 1,8% (Jayabakti) and 2.8% (Liang), respectively.Rumput laut merupakan sumberdaya pesisir yang sangat potensial. Karagenan merupakan polisakarida yang diekstraksi dari beberapa spesies rumput laut atau alga merah (rhodophyceae). Pertumbuhan rumput laut sangat dipengaruhi oleh dua faktor yaitu faktor internal dan faktor eksternal. Namun yang sangat menentukan keberhasilan pertumbuhan rumput laut yaitu pengelolaan yang dilakukan oleh manusia. Kabupaten Banggai merupakan salah satu sentral produksi rumput laut terbesar di Sulawesi Tengah. Tujuan utama penelitian ini mengkaji tentang potensi budidaya rumput laut (Kappaphycus alvarezii) yang dikembangkan dengan melihat pertumbuhan dan analisis karaginannya di perairan Kabupaten Banggai Provinsi Sulawesi Tengah. Kisaran suhu yang didapat selama penelitian ini adalah berkisar 25–31ºC. Hasil analisa rendemen karagenan ini sangat berbeda yang disebabkan oleh perbedaan lokasi memberikan pengaruh nyata terhadap kandungannya. The result showed that edible film processed fromcarrageenan with different concentrations have different physical properties. The higher the concentration of carrageenan used in the manufacture of edible film, the stronger the solubilityof the edible film, the stronger the attractiveness, andthe thicker and more resistant. Edible filmdari bahan karaginan merupakan salah satu bentuk kemasan primer bagi bahan pangan. Sifat fisik kemasan menjadi komponen penting yang dipertimbangkan dalam memilih kemasan. Penelitian inidilakukan untuk mengetahui pengaruh konsentrasi karaginan terhadap sifat fisik kelarutan, ketebalan, kuat tarik, persentasi perpanjangan putus dan laju transmisi uap air. Hasilnya diperolehedible filmyang diproses dari karaginan dengan konsentrasi yang berbeda memiliki sifat fisik yang berbeda. This study reveals that theecological condition of Olele waters is good, and it was shown bythe average coral cover which ishigher than 50%. Similarly, fish species is in very high abundance, with a total of 36 species and a total number of more than 12.993 fish, wherePseudanthias tukais the highest population. In terms of ecotourism feasibility development, total of Pirkins Score were 3,2, andthis can be categorized as moderate, meaning that Olele coastal area can be developed as ecotourism area. Finally, strategies for ecotourism development of Olele coastal area are; a) using Olele coastal resources for ecotourism destiny by promoting conservation values, b) infrastructure ecotourism development needs to be improved, c) integratrated ecotourism management policies should be included in policy for development of Kabupaten (disrict) government level; d) development of ecotourism need to cooperation between district government and private sectors.Tujuan penelitian ini adalah mengevaluasi kondisi terumbu karang dan ikan karang yang berada di perairan pesisir Olele, mengkaji kelayakan pengembangan ekowisata di kawasan pesisir Olele dan merumuskan strategi pengembangan kawasan ekowisata. Dari hasil penelitian dan analisis data, kondisi ekologi perairan Olele berada pada kategori baik: karang pada stasiun 1 rata-rata memiliki tutupan karang hidup >50%, artinya bahwa keragaman karang tinggi. Sama halnya dengan spesies ikan, jumlah species sebanyak 36 spesies dan total jumlah individu sebanyak 12.993 dimana spesies terbanyak yaitu Pseudanthias tuka. Selanjutnya analisis kelayakan pengembangan ekowisata, total nilai scoringPirkins 3,2 atau berada pada level moderat, artinya dapat dikembangkan menjadi kawasan ekowisata. This study provides information on the condition of coral reefs on the eastern coast of Minahasa Regency, especially in Kamenti and in Toloun. Sampling withSCUBA was conducted using lifeform categories and transects in the form of a 50 m measuring tape with 'Line Intercept Transect' techniqueat thedepthsof 5 m, 10 m and 15 m. The condition of coral reefs in the eastern coastal waters of Minahasa Regency is generally 'good'. The percentage of coral cover in Kamenti was categorized'good', while in the waters Toloun are in a state of 'average'. Genera richness on both sites can be quite high, intotal there are44 genera with 35 genera in Kamenti and 38 genera in Toloun. Kamenti station with the depth of 15 m hasthe highest conservation value because in addition to having the highest number of genera (32 genera), it has also the highest Shannon index (3.35), the highest genera richness index (8.29), the highest genera evenness index (0, 97) and the lowest dominance index (0.12). There is no apparent correlation between the percentage of live coral cover with the number of genera or the Shannon index. But the high percentage of live coral cover is not always identical with the high genera richness; the maximum genera richness is at the intermediatecover level.Perairan pantai sebelah timur Kabupaten Minahasa Provinsi Sulawesi Utara merupakan salah satu kawasan perairan yang berpotensi untuk dijadikan Kawasan Konservasi Perairan. Penelitian ini memberikan informasi tentang kondisi terumbu karang di pantai timur Kabupaten Minahasa khususnya di Kamenti dan di Toloun. Sampling dengan menggunakan SCUBA dilakukan dengan metode kategori lifeformdan menggunakan transek berupa pita meteran sepanjang 50 m dengan teknik ‘Line Intercept Transect‘ pada kedalaman 5 m, 10 m dan 15 m. Kondisi terumbu karang di perairan pantai timur Kabupaten Minahasa umumnya ‘baik’. Persentase tutupan karang batu di Kamenti berada pada kategori ‘baik’ sedangkan di perairan Toloun berada pada kondisi ‘sedang’. Kekayaan genera pada kedua lokasi penelitian dapat dikatakan cukup tinggi yakni secara total terdapat 44 genera dengan masing-masing 35 genera di Kamenti dan 38 genera di Toloun. Stasiun Kamenti kedalaman 15 m adalah yang paling tinggi nilai konservasinya karena selain memiliki jumlah genera terbanyak (32 genera), juga memiliki indeks Shannon tertinggi (3,35), indeks kekayaan genera tertinggi (8,29), indeks kemerataan genera tertinggi (0,97) dan indeks dominasi terendah (0,12). The establishment of a marine conservation area does not necessarily solve its management problems. The study aimed to examine the sustainability status of the MPA management of Uwedikan village and to formulate a managementstrategy. Research method used wasa descriptive method through case study approach. Data collection employed questionnaires-basedinterviews on respondents. The data were analyzed by using Rapid Appraisal for Fisheries Status (Rapfish) to obtain the sustainability status of the MPA in Uwedikan. The Rapfish outcome wasthenanalyzed using Diagnosis and Therapy Analysis of Law (DTAL) to formulate a strategy for the existing management.Kawasan Konservasi Perairan Daerah (KKPD) di Desa Uwedikan merupakan kawasan konservasi yang telah ditetapkan melalui keputusan Bupati Banggai sejak tahun 2008. Penetapan suatu kawasan konservasi laut tidak serta merta dapat menyelesaikan permasalahan pengelolaannya. Penelitian bertujuan untuk mengkaji status keberlanjutan penatakelolaan kawasan konservasi perairan daerah Desa Uwedikan dan merumuskan strategi pengelolaannya. Metode penelitian adalah metode deskriptif melalui pendekatan studi kasus. Pengumpulan data melalui wawancara kuesioner mendalam terhadap responden. Data collection covered primary and secondary data. Primary data was carried outthrough direct observation on production planning, production process, and control on the mini purse-seine fishing unit and through direct interviews with mini purse-seine owner and fishermen. Secondary datawas obtained from Marine and fisheries Service of North Halmahera. Literature study was conducted as well. The data were analyzed to find outthe business pattern practiced by mini purse-seine fishermen by usinginteractive model of analysis and time series analysis-based average percentage methodwas usedto get the fishing season.This study concluded that business pattern of fishermen in South Tobelo district was done by imposingthe operational cost on the investment owner in both peak and lowfishing seasons. Moreover, the profit sharing is divided by50% for townersand 50% for fishermen after operational cost, and 20% for the FAD if other FAD was used. The fishing season of the mini purse-seine in South Tobelo district occurrsin March, August and September, while the lowseason occursin January, February, June, November, and December.Penelitian ini menggunakan metode deskriptif dan dilakukan selama 5 bulan. Pengumpulan data dilakukan baik pada data primer maupunsekunder. Data primer dilakukan dengan pengamatan langsung terhadap perencanaan produksi, proses produksi dan pengawasan produksi unit penangkapan pajekodan wawancara terhadap nelayan dan pemilik kapal pajeko. Data sekunder dikumpulkan dari Dinas Kelautan dan Perikanan Kabupaten Halmahera Utara disamping itu dilakukan juga study pustaka. Untuk mengetahui pola usaha yang dijalankan oleh nelayan pajekodigunakan interactive model of analysis. Untuk analisis pola musim penangkapan ikan menggunakan metode persentase pata-rata (The Average Percentage Methods) yang didasarkan pada Analisis runtun raktu (Times Series Analysis). Dari hasil Penelitian ini dapat disimpulkan bahwa pola usaha yang digunakan oleh nelayan di Kecamatan Tobelo Selatan dilakukan dengan membebankan biaya operasional pada pemilik baik musim puncak maupun musim paceklik tiba. Disampaing itu system bagi hasil dibagi 50% untuk pemilik dan 50% untuk nelayan setelah dikeluarkan biaya operasional dan 20% untuk rumpon apabila menggunakan rumpon milik orang lain. This study aimsto analyze the policy strategy for developingMinapolitan Region of the City of Bitung through SWOT analysis and determinepriorities for the development strategy of Bitung city Minapolitan Region. The research method is a survey study, data were collected through interviews and direct observation and literature. SWOT analysis is used to identify strengths, weaknesses, opportunities and threats. Furthermore, to determine the alternative development strategy used OSPM. This requires that fishermen have the motion or power that can determine and influence the behavior of groups and their members in achieving their goals effectively. Fishermangroups should always be dynamic in running fishing business. This study aims to 1) analyze the factors that affectthe dynamics of traditional fishing groups in the coastal reclamation area of Manado City, and 2) determine the dynamics model of traditional fishermen groups in the coastal reclamation area of Manado City. This research wasconducted using survey method. F test statistics wasused for mixedinfluence test and t-test statistic for partial test. Multiple linear regressions was conductedto describe the dynamics of traditional fishermen group in the coastal reclamation area of Manado City. The result of this research concluded that internal (X1) and external (X2) factors together or partially hasan effect on dynamics of traditional fisherman group of coastal reclamation area of Manado City. Partially, internal sub factor (education level and experience of fishing effort) and external sub factor (the availability of capital aid and information availability) have real effect ondynamics of traditional fisherman group of coastal reclamation area of Manado City. The dynamicequationmodel of traditional coastal reclamation area of Manado CityisY = 0,770774 + 0,344691X1+ 0,388735X2.Kelompok nelayan merupakan wadah kerjasama dari nelayan dalam satu wilayah untuk dapat mencapai nelayan yang berkualitas. Hal ini mengharuskan nelayan memiliki gerak atau kekuatan yang dapat menentukan dan mempengaruhi perilaku kelompok dan anggota-anggotanya dalam mencapai tujuan secara efektif. Kelompok nelayan harus selalu dinamis dalam menjalankan usaha penangkapan ikan. Penelitian ini bertujuan untuk 1) menganalisis faktor-faktor yang mempengaruhi dinamika kelompok nelayan tradisional di kawasan reklamasi pantai Kota Manado, dan 2) menentukan model dinamika kelompok nelayan tradisional di kawasan reklamasi pantai Kota Manado. Penelitian ini mengunakan metode survey. Statistika Uji-F digunakan untuk uji pengaruh secara bersama-sama dan Statistika Uji-t untuk uji parsial. Regresi linear berganda dilakukan untuk menggambarkan persamaan dinamika kelompok nelayan tradisional di kawasan reklamasi pantai Kota Manado.
