Many studies, demonstrating altered expressions of specific miRNAs in diverse types of human neoplasia, suggested that they may play a key role in tumorigenesis. Recently, miRNA genes were found to be abnormally expressed in several types of cancer, including endometrial cancer. However, miR-23b and miR-203 expression in endometrial cancer has yet to be studied in Korea. As such, the purpose of this study was to analyze miR-23b and miR-203 expressions in endometrial cancer and to evaluate the relationship between miR-23b and miR-203 expressions. A retrospective study was carried out on the formalin-fixed, paraffin-embedded tissues of 42 endometrial cancer tissues using quantitative real-time PCR. In endometrial cancer tissues, miR-23b expression levels (2.70±4.45) were higher than miR-203 expression levels ( − 2.34±4.08). Endometrial cancer tissues showed an overexpression of miR-23b in 30 (71.4%) of the 42 endometrial cancer cases, whereas miR-203 was underexpressed in 29 (69.0%) of the 42 cases. There was a significant association between miR-23b and miR-203 expressions in endometrial cancer tissues (p=0.0005). These findings suggest that miR-23b and miR-203 expressions may be involved in endometrial carcinogenesis. The purpose of this study is to develop an ELISA kit suitable for evaluating the expression of skin barrier proteins. Proteins were obtained from the skin via AriNo and D-Squame patches. The efficiency of protein collection from the skin, using the Arino patch, was shown to be more effective than using D-Squame; while the efficiency of lysis using 0.1% Triton-X100 was higher than that of other lysis solutions, including 0.1 M Tris-HCL, 0.1% Tween-20, and 5 mM KOH. Recombinant skin barrier proteins, such as filaggrin and involucrin, were produced by molecular biological methods. Monoclonal antibodies against filaggrin and involucrin were produced by immunization of mice, fusion of spleen cells and myeloma cells, as well as a selection of antibody-producing hybridoma cells. The filaggrin expression in the skin of subjects suffering from atopic dermatitis was lower than that in normal mice. Involucrin expression was not altered between normal individuals and subjects with atopic dermatitis. This study examined the relationship between the LAP index and ALT (alanine aminotransferase) level and the effect of the LAP index on the ALT level. Cross-sectional studies were conducted on healthy adult males without a history of liver disease. From January 2015 to June 2017, 13,854 adults between 20 and 70 years of age, who underwent health screening at a general hospital in Gyeonggi-do, were enrolled in this study. The LAP index was calculated as [waist circumference (cm) − 65]×[triglyceride (mmol/L)] for males. Serum ALT abnormalities were set at 40 IU/L or more in males. All subjects underwent measurements of the anthropometric indicators and physiological examinations. A significant increase in ALT with increasing LAP quintiles was observed (p < 0.001). The LAP index correlated with ALT (r=0.238, p < 0.001). Insulin (r=0.449, p < 0.001) and HDL-cholesterol (r= − 0.369, p < 0.001) were strongly correlated with LAP. In addition, the LAP index is a factor affecting the ALT level (p < 0.001). As the LAP index increased, the risk of an elevated ALT was higher (p < 0.001). Lee","doi":"10.15324/KJCLS.2017.49.4.350","DOIUrl":"https://doi.org/10.15324/KJCLS.2017.49.4.350","url":null,"abstract":"용혈 지수(hemolysis index)는 생화학 장비에서의 용혈을 위한 반정량 지표이다. 용혈 지수의 특성이 상업 플랫폼마다 다르기 때문에 용혈 지수의 표준화 또는 일치화는 현재 되어 있지 않다. 진단검사의학에서 일치화(harmonization)는 측정 절차와 상관없이 동일한 결과와 동일한 해석을 할 수 있는 능력을 말한다. 임상 환자의 혈액 중에서 무작위로 추출한 40개의 검체를 인위적으로 체외 용혈(in vitro hemolysis)시킨다. 혈액 자동화 장비인 XE-2100 (Sysmex, Japan)에서 측정된 혈청 헤모글로빈을 기준으로 검체의 혈청을 0∼300 mg/dL 사이에서 20mg/dL 차이로 각각을 희석시킨다. 희석된 혈청 헤모글로빈을 생화학 자동화 장비 Hitachi-7600 (Hitachi, Japan)으로 측정하여 용혈 지수와 혈청 헤모글로빈을 평가하였다. 용혈 지수와 혈청 헤모글로빈은 선형성을 보였으며 용혈 지수 1은 대략 혈청헤모글로빈 20 mg/dL 정도였다. 용혈 거절 기준을 결정하기 위해 용혈 지수를 세 그룹으로 나눈다: 용혈 지수 0∼1, 용혈지수4∼6, 용혈 지수 9∼15. 또 다른 임상 검체 40개의 검체를 Hitachi-7600 (Hitachi, Japan)로 각각의 임상 검사 종목을 측정한 후, 각각의 검체를 주사기의 피스톤을 앞·뒤로 움직여 인위적인 체외 용혈을 시키고 Hitachi-7600 (Hitachi, Japan)로 측정하였다. 세 그룹 사이의 임상 검사 종목의 용혈로 인한 백분율 차이(percentage difference)를 ANOVA 또는 Kruskal-Wallis test 분석하였다. 사후 검정에서 용혈 지수 0∼1인 그룹과 용혈지수 5∼6인 그룹 사이에 유의한 차이가 나는 종목들은 Glucose, creatinine, total protein, AST, direct bilirubin, uric acid, phosphorus, triglyceride, LDH, CPK, magnesium, potassium이였다. 많은 임상 검사 종목들이 용혈지수 4∼6인 그룹과 참조 그룹인 용혈지수 0∼1인 그룹 간에 통계적으로 유의미한 차이를 보이기 때문에 용혈 기준은 대략 용혈지수 5 (혈청 헤모글로빈 100 mg/dL)가 적합하다 판단된다.","PeriodicalId":10080,"journal":{"name":"临床检验杂志","volume":"33 1","pages":"350-358"},"PeriodicalIF":0.0,"publicationDate":"2017-12-31","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"76891751","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

These bacteria are opportunistic pathogens that can contaminate people, and in doing so, cause various social economic problems. This study proved that NTM are also present in air conditioners of multiuse facilities in frequently visited populated areas. The results of this study are meaningful because they showed that pathogenic microbes can live in the surroundings and cause diseases to people with impaired immunity. This study used the AFB stain, L-J medium culture, and PCR method for the detection of MTB and NTM in air conditioner dust. MTB was not detected in any of the collected samples, while NTM was detected in 2 out of 40 samples (5%). Most people living in the modern environment cannot avoid being exposed to air conditioners so special attention is needed for sanitary inspections and the management of air conditioners. Moreover, it is important to study NTM through various individuals, accumulate the related data and establish the methods and standards of hygiene management system to reduce NTM infections in public areas.
