The fragments were separated by chromatography on DEAE-cellulose columns and in some cases by subsequent paper electrophoresis. The amounts of mono- and dinucleotides from various unfractionated and partially fractionated soluble RNA's were compared (Table I). In hydrolysates of highly purified serine transfer RNA fractions 7 di-, 7 tri-, 7 tetra-, 2 penta- and 2 hexanucleotides - coming from 2 serine transfer-RNAs - were identified and quantitatively determined (Tables II and III), as well as some oligonucleotides, which occur in smaller amounts and probably come from contaminations of the serine transfer-RNA fractions. 3 oligonucleotides could not be identified, and one only partially. The effects of 5-fluorouracil, not reversible in the presence of uracil, on <em>Staphylococcus aureus</em> (strain Duncan) were studied.</p></span></li><li><span>2.</span><span><p>2. The logarithmic growth of <em>S. aureus</em> (strain Duncan) in the presence of 5-fluorouracil (100 μg/ml) and uracil (200 μg/ml) was retarded by 90% after 60-min incubation.</p></span></li><li><span>3.</span><span><p>3. Up to 4% of the UMP in the ribosomal RNA was replaced during 60-min growth, by 5-fluorouracil ribotide: 14% of UMP in the soluble RNA was replaced during this period.</p></span></li><li><span>4.</span><span><p>4. The proportions of cell wall, hexose and polysaccharide were unaffected, but the lipid content fell slightly.</p></span></li><li><span>5.</span><span><p>5. Total nucleic acid fell by 45% but the total nitrogen (less nucleic acids) remained constant.</p></span></li><li><span>6.</span><span><p>6. {"title":"Characteristics of ribosomes isolated from roots of Zea mays","authors":"Theodore C. Five ribosomal components (40 S, 60 S, 80 S, 120 S, and 150 S), with the 80-S component predominating, were present in the cytoplasmic extract. Sedimentation coefficients at infinite dilution were determined for all except the 150-S component.</p><p>Purified ribosomes contained 43% RNA which was characterized by a high guanine and relatively high cytosine content. The 80-S component dissociated into 60-S and 40-S particles when pH of the medium was increased from 6.5 to 8.0 or higher. Magnesium (0.5–2 mM) prevented the dissociation.</p><p>In marked contrast to the 70-S ribosome of <em>Escherichia coli</em>, the 80-S ribosome of corn did not require added magnesium to maintain its integrity in a solution of nearly neutral pH and low ionic strength. In fact, 5 mM magnesium in such a solution caused the association of corn ribosomes into large random aggregates.
