The control group consisted of rats without pathology. A catheter previously implanted into the jugular vein of the animals, allowed repeated blood sampling and monitoring of the pharmacokinetic profile of the substance in each animal. Pharmacokinetic studies usually involve healthy animals, but the ischemia/reperfusion injury of the liver disrupted its functions, potentially altering drug metabolism and pharmacokinetics. In this study, the changes in cerebral angiogenesis and the characteristics of angiogenesis in the prefrontal cortex of Alzheimer's disease (AD) mice were investigated. AD was induced by injection of β-amyloid into the hippocampus. The animals underwent cognitive testing, and their brains were analyzed using immunohistochemical techniques to evaluate angiogenesis. The results showed an increase in the proliferation of endothelial cells and altered ratio of tip, stalk, and phalanx cells. Vascular branching and density in the prefrontal cortex were decreased, indicating an unproductive form of angiogenesis in AD. Obtained results highlight the link between changes in angiogenesis and cognitive decline, confirming the importance of vascular dysfunction in AD progression.</p>","PeriodicalId":9331,"journal":{"name":"Bulletin of Experimental Biology and Medicine","volume":" ","pages":"371-375"},"PeriodicalIF":0.9,"publicationDate":"2025-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"143413467","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The most potent stimulation of migration and proliferation of EC and FB was observed at angiogenin concentration of 125 ng/ml in a serum-free medium. Angiogenin had little effect on apoptosis/necrosis of EC and FB. It promoted IL-1β production by EC in a serum-free medium and did not affect FB. Angiogenin had the opposite effects on the production of TNFα and IL-10 by cells: it suppressed the production of these factors by EC in a serum-free medium and enhanced their production by FB in media with and without serum. In a serum-free medium, VEGF production by EC increased and by FB drops. NO production by EC in response to angiogenin stimulation did not depend on the culturing conditions, while NO production by FB increased in complete culture media. Thus, the presence of FCS in the culture medium considerably modulated the effect of angiogenin.</p>","PeriodicalId":9331,"journal":{"name":"Bulletin of Experimental Biology and Medicine","volume":" ","pages":"367-370"},"PeriodicalIF":0.9,"publicationDate":"2025-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"143405877","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

A single injection of 5×106 LLC cells into the right lateral subcostal region caused the growth of the primary tumor and its metastasis to the lung. For reducing metastasis and mortality after resection, the primary tumor should be removed with subcutaneous fat on day 8 after inoculation. Antitumor and antimetastatic effects and reduced mortality were achieved by intraperitoneal injection of carboplatin (63.3 mg/kg) and paclitaxel (13.3 mg/kg); chemotherapy was administered twice. The combination of the two approaches increased the survival: the antitumor and antimetastatic effects were observed in 60% of mice with LLC relative to resection alone or treatment with cytostatics alone. However, chemoresistance was formed in 40% of mice, the values of the tumor growth inhibition index and the metastasis inhibition index decreased. This was accompanied by the appearance of cancer stem cells in the circulation with the potential for the formation of spheroids in vitro.
