To study the response of the sciatic nerve trunk’s hemomicrocirculatory bed to a single administration of cryopreserved placenta and intraperitoneal administration of λ -carrageenan. Material and methods. An experimental study was performed on the sciatic nerve trunk of 100 sexually-mature rats. Histological research methods were used. It was established that when transplanting a cryopreserved placenta, all parts of the hemomicrocirculatory bed (HMCB) respond. Thus, the resistive part of the sciatic nerve trunk HMCB during cryopreserved placenta transplantation was characterized by a gradual increase in the mean diameters with the maximum value on the 5th-7th days and their complete recovery on the 10th day of the study. In the simulation of acute aseptic peritonitis there is a sharp decrease in diameters on the 2nd-3rd days, followed by their increase on the 5th-14th days and recovery on the 30th day. Results. In transplantation of cryopreserved placenta, the exchange link of the HMCB reacted significantly by increasing the mean diameter of the capillaries on the 5th day. In the simulation of acute aseptic peritonitis, the mean diameter of the capillaries increased on the 1st day and did not return to the control group limits on the 30th day of the study. Capacitive link responded to transplantation of cryopreserved placenta by an increase in diameter with the maximum value on the 5th day and recovery on the 10th day of the study (p <0.05). When simulating acute aseptic peritonitis, there is also an increase in diameters with the maximum value on the 7th day. On the 30th day of the study, the index value did not reach the limits of the control group. Conclusions. A single administration of cryopreserved placenta fragment causes short-term changes in the hemomicrocirculatory bed with the maximum values on the 5th-7th days of the study, in contrast to a single injection of 5 mg of λ-carrageenan (Sigma -USA), which leads to reactive inflammation and lasts for over 30 days.","PeriodicalId":89801,"journal":{"name":"Journal of clinical & experimental pathology","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2020-09-13","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"91237856","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The objective of the work was to study the activity of superoxide dismutase and catalase in the myocardium of male and female animals with prolonged use of dexamethasone, high NaCl content in water and the possibility of using L-carnitine as a protective agent. Material and methods. The experiment was performed on 96 mature white non-linear rats (age 5-6 months, weight 0.17-0.23 kg). Long-term glucocorticosteroid therapy was modeled by per os dexamethasone intake. Half of the animals received drinking water with a high NaCl content. Correction was carried out by per os L-carnitine intake. Long-term use of dexamethasone has led to a decrease of the catalase and superoxide dismutase activity. Increased NaCl content in water exerted a slight negative effect in conjunction with glucocorticoids, more in males. Results. During prolonged use of dexamethasone L-carnitine completely resumed the activity of superoxide dismutase in female and male animals. In female rat catalase activity was restored in all groups of animals. In male rat with a high salt content (4%) in water, the catalase activity could not fully recover.","PeriodicalId":89801,"journal":{"name":"Journal of clinical & experimental pathology","volume":"29 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2020-05-07","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"73242623","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

To characterize the state of theautonomic nervous system we used A.M. Vein vegetative symptoms scale, Ashner-Danyini,Danielopulu (orthoclinostatic) tests.Results. Indices’ analysis of the carried out investigation of the general vegetativesymptoms have shown that the skin color changes during excitement, physical activity inpatients with somatoform autonomic dysfunction according to hypertensive type, cardiacneurosis and hypertension had a negative tendency, that is, the skin was pale, whatreflects an increase of the sympathetic nervous system tone and forms a decrease fromthe state of eutonia in scores by 1.7 times, 1.6 times and 2.0 times respectively (p<0.05).Pulse rate veritably increased (51.6%, 41.9% and 54.8% (p<0.05)) in patients of the 1st,3rd and 4th groups, in patients of the 2nd group it veritably decreased (29.0% (p <0.05))when conducting orthostatic test. During clino-orthostatic test, the pulse rate changed inthe diametrically opposite direction: in patients of the 1st, 3rd and 4th groups it sloweddown (21.9%, 28.1% and 31.3%, respectively (p< 0.05)), in patients of the 2nd groupit probably accelerated (21.9% (p <0.05)). To carry out a comparative analysis of the frequency of PAI-1 gene G43A polymorphism in citizens of Chernivtsi region who suffer from various forms of ulcer disease, and to study the possible relationship between different types of genotype and the development of complications. Material and methods. 60 patients with ulcer disease were examined: 42 (70%) men, 18 (30%) women. 37 (61.67%) patients had a duodenal ulcer, the rest ones (38.33%) had a gastric ulcer. Uncomplicated ulcer disease was diagnosed in 12 (20%) patients. 5 (8.33%) patients were found to have an ulcer perforation. In 43 (71.67%) patients there was an ulcer complicated by acute bleeding. In 29 (67.44%) patients the bleeding was stopped. In 14 (32.56%) patients there was a relapse of bleeding. PAI genotyping by G43A mutation was performed by using polymerase chain reaction (PCR). PCR amplification of the corresponding RAI gene fragment was performed by using a specific pair of primers 5’-CCA ACA GAG GAC TCT TGG TC-3’ and 5’- CAC AGA GAG AGT CTG GCC ACG-3’. PCR was performed by using a CFX96 amplifier (Bio-Rad, USA). PCR results were carried out by electrophoresis in 2% agarose gel using a tris-borate buffer. For DNA fragments visualization the gel was stained with ethidium bromide and photographed using ultraviolet light on the GelDoc 2000 installation (BioRad, USA). To determine the length of the obtained fragments, their electrophoretic mobility was compared with the mobility of the DNA marker Gene Ruler DNA Leader Mix (Thermoscientific). The statistical dependence between the values was tested by determining the Fisher criterion, χ2-criterion by Pearson, in particular, the correspondence of the genotype distribution to the Hardy-Weinberg equilibrium. Results. 94.12% patients without acute bleeding and 91.67% patients without acute complications were found to have GG genotype. One female patient (8.33%) without any acute complications was found to have homozygous AA genotype by the mutant allele. No patient with an ulcer perforation was a carrier of allele A. None of the patients without bleeding had a heterozygous GA genotype. GA genotype was found in 25.58% of patients with an acute ulcerative bleeding, which is statistically significantly higher than in patients without any bleeding.
