In this study, an oleic acid (OA)-induced hepatocyte steatosis model of orange-spotted grouper (Epinephelus coioides) was established for the first time. The model was used to test the effect of taurine on steatosis hepatocytes in Control, High-fat (0.4 mM OA) and Taurine (0.4 mM OA + 2 mM taurine) experimental groups of fish. Hepatocyte samples were subjected to transcriptome analysis. A total of 99634 unigenes was assembled, 69982 unigenes were annotated and 1831 differentially expressed genes (DEGs) in Control vs High-fat group, and 526 DEGs in the High-fat vs Taurine group were identified, of which 824 DEGs (Control vs High-fat) and 237 DEGs (High-fat vs Taurine) were observed to be upregulated, and 1007 DEGs (Control vs High-fat) and 289 DEGs (High-fat vs Taurine) were downregulated after taurine intervention. These genes are involved in peroxisome proliferator-activated receptor (PPAR) and 5' AMP-activated protein kinase (AMPK) signaling pathways, fatty acid elongation, primary bile acid biosynthesis, glycerophospholipid and glycerolipid metabolism. The findings provide new clues in understanding the regulatory role of taurine in lipid and fatty acid metabolism of fish. To estimate the influence of copper mining on river biota, the quantitative and qualitative macroinvertebrate community structure was analyzed, and the ecological status of the river was assessed using standard biological indices. Data was gathered at 6 sites along the river. A total of 75 macroinvertebrate taxa were recorded. The reference site had the highest number of recorded taxa (48) and a high biodiversity (diversity index H=2.3). In contrast, the lowest number of taxa (8), a low diversity index value (H=1.15), and a very poor overall ecological status were recorded at site 2, which is heavily influenced by mining discharge. The Ephemeroptera, Plecoptera and Trichoptera group (EPT) constituted 46.30% of the community at site 1 but was completely absent at site 2 where Diptera (70%) and Oligochaeta (20%) were dominant, reflecting a strong impact of the mining industry on the upper river stretch. Adult male albino rats were administrated PSO (per os, 2 mL/kg b.w./day) and a 12% ethanol solution in water, ad libitum, with an average intake of 8.14 g of ethanol/kg bw/day for 6 weeks. Congestion, hepatic central vein dilation, portal vein branch dilation, Kupffer cell hyperplasia, fatty liver changes, hepatocyte focal necrosis were observed after daily alcohol intake. All observed changes were reduced when PSO was ingested with ethanol. PSO intake itself induced discrete cellular edema, congestion and slight dilatation of the central and portal vain branches. Chronic ethanol intake elevated catalase (CAT) activity and glutathione reductase (GR) protein expression; concomitant PSO intake had no effect on CAT activity or GR protein expression. PSO intake decreased the activities of GR, glutathione-S-transferase (GST) and xanthine oxidase (XOD) in the liver, probably due to the ingestion of antioxidants. Intake of PSO and ethanol significantly decreased cytosolic superoxide dismutase (SOD1) and increased NF-?B protein expression compared to ethanol intake, suggesting that the protective effects of PSO were mediated by the NF-?B signaling pathway. Successful regeneration of damaged periodontal tissues depends on the proper functionality of periodontal ligament mesenchymal stem cells (PDLMSCs), especially the production of extracellular matrix proteases. We investigated the influence of IL-17 on ECM remodeling through modulation of urokinasetype plasminogen activator (uPA) and MMP2/MMP9 expression in human PDLMSCs at mRNA, protein and activity levels using by RT-PCR, Western blotting and zymography, respectively. Investigation of the involvement of MAPKs in these processes in PDLMSCs was determined by Western blotting, as well as by utilizing specific p38 and MEK1/2 inhibitors. Our results show that IL-17 activates MAPK signaling in PDLMSCs. Moreover, IL-17 had no effect on MMP9 expression, but it stimulated uPA and MMP2 gene and protein expression in PDLMSCs through the activation of the ERK1/2 MAPK signaling pathway. The obtained data suggest that IL-17 contributes to ECM degradation in the periodontal ligament by stimulating uPA and MMP2 expression and activity in PDLMSCs. This study aimed to evaluate the acute toxicity of the aqueous extract of C. cainito (CE) bark based on OECD guidelines in two different in vivo experimental models: acute single-dose oral toxicity in adult Wistar rats and the zebrafish embryo acute toxicity test. All concentrations of CE (500-4000 mg/kg) tested during a 14-day period in both male and female rats showed no effect on behavior, body weight, organ weights, biochemical and hematological parameters. In contrast, CE significantly delayed zebrafish embryo hatching and decreased embryo survival rates in a dose-dependent manner. Hatched larvae were notably sensitive to CE-induced toxicity compared to unhatched fish embryos. Acridine orange staining showed that CE induced apoptosis in the yolk sac region that is responsible for supplying nutrients to support larval growth and development. According to OECD guidelines, CE was identified as GHS category 5, a substance with low to no acute toxicity. It is a cysteine protease highly expressed in Carica papaya fruit latex, but also present in papaya leaves. Purification procedures mostly deal with the latex and include a combination of precipitation and/or chromatographic techniques. Due to its solubility, structure and activity characteristics, the pH and salt content play significant roles in handling papain extracts. Here we report a simple, rapid and easily scalable procedure for papain purification from papaya leaves, which contain different contaminants as compared to papaya latex. Sodium chloride precipitation of contaminants at pH 5 followed by ammonium sulphate precipitation resulted in the removal of other leaf proteins and protein fragments from papain solution and about a 3-fold purification. Thus, overcoming the side effects of CPZ is of great importance. Because agmatine (AGM) can act as a free radical scavenger, it is an interesting compound as an adjunct to CPZ therapy. The aim of our study was to investigate the enzymatic parameters of oxidative stress in the hippocampus and striatum of rats after CPZ treatment, and the potential protective effects of AGM. Rats were injected as follows with (i) 1 mL/kg b.w. saline; (ii) a single intraperitoneal (i.p.) dose of CPZ (38.7 mg/kg); (iii) CPZ (38.7 mg/kg) and AGM (75 mg/kg); (iv) AGM (75 mg/kg). CPZ induced an increase in superoxide anion radical (O2 ?-) concentration, while the activities of the antioxidant enzymes, superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR), were lowered in both the hippocampus and striatum. Cotreatment with CPZ and AGM protected the examined brain structures by reversing the antioxidant enzyme activities to the control values. Following CPZ treatment, the effects were more pronounced for SOD and GPx in the hippocampus, and for SOD, CAT and GPx in the striatum. The full effect of restored superoxide production was achieved in the striatum, which points to the role of CAT. Emotions were considered as irrational experiences beyond the domain of logical perception because of their intertwinement with mood, temperament, creativity, motivation and personality. Through the centuries, emotions have been the focus of research among great classical philosophers, doctors, neuropsychologists, neuroscientists, neurologists and psychiatrists. The neurophysiological basis of behavior, such as emotional facial expression, and autonomic events in the physiological theory of William James and James-Lange and modified by Cannon-Bard, was followed by the two-factor theory of emotions of Schachter-Singer and Lazarus? higher-order cognitive evaluation. Four components that influence each other represent the concept of emotions and complete the overall emotional experience, and these are: autonomous (increase in heart rate, blood pressure); somatic (body language, facial expressions); cognitive (control, management), and subjective feeling (emotion, individual experience). The interplay between emotions and cognition has been the subject of research. Emotions can be evoked reflexively by simple physical stimuli (bottom-up), but can also be complex reactions involving cognitive, physiological and behavioral reactions (top-down). The European ground squirrel, Spermophilus citellus, an endangered flagship species of the open grasslands of central and southeastern Europe, is no exception. Considering anthropogenic changes affecting its habitats in northern Serbia, we studied the species? feeding habits by subjecting its feces to microhistological examination. In addition to identifying plant remains in the feces, we examined the vegetation of the four study sites located inside a local mosaic of steppe and saline pastures. Our findings indicate that these remaining European ground-squirrel sites are diverse and stable grasslands covered by native flora that represents a complete dietary base for the species. The diet is dominated by a small number of plant species, specifically Achillea millefolium, legumes and grasses, with sporadic contributions from a number of other species. The diet of S. citellus also differs by location and season in that grasses dominate during the early season, while A. millefolium takes precedence later in the year. This dietary information is essential for the development of future species and habitat management strategies. Vukomanovic","doi":"10.2298/ABS210121012V","DOIUrl":"https://doi.org/10.2298/ABS210121012V","url":null,"abstract":"Perinatal asphyxia (PA) is a condition characterized by a gas exchange disorder due to a lack of blood flow or gas exchange, with potential multiorgan dysfunction. Our study aimed to determine the correlation between biochemical markers and echocardiography findings in a group of asphyxiated newborns. The prospective cohort study included 120 neonates (52/120 with PA) treated at a tertial referral pediatric center, from 2012 to 2014. A moderate-strong positive correlation was recorded between the transtricuspid pressure gradient (TRPG) and serum lactate, and between TRPG and NT-proBNP in the PA group (P<0.001) on the 1st day of life. A moderate positive correlation was found between NT-proBNP, lactate and troponins on one side, and TRPG on the other in the PA group after the 2nd measurement. Multinomial regression analysis showed that the lactate level was an independent factor for survival on the 1st (odds ratio (OR) 41.3, 95% confidence interval (CI) 2.14-797.1) and 3rd (OR 136.4, 95% CI 2.27-8206.7) days. Our research confirmed a significant correlation between echocardiographic and biochemical parameters of the myocardial lesion and cardiac function.
