This highly active class of anticancer agents possesses a unique mechanism of action with some inherent shortcomings, which might have been solved by liposomal formulation. Recent studies have revealed the protective action, increased tumor delivery and prolonged plasma exposure of these liposomal formulated drugs. These advances in pharmaceutical development could increase the levels of activity of these agents, as well as increase their clinical utility as new emerging anticancer therapies.

","PeriodicalId":80125,"journal":{"name":"Pharmaceutical science & technology today","volume":"3 6","pages":"Pages 205-209"},"PeriodicalIF":0.0,"publicationDate":"2000-06-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1016/S1461-5347(00)00268-6","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"21685154","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Parenteral administration by high-dose injection can, however, cause serious side effects. Significant improvement in the therapeutic index of IFNs could be achieved with oral administration. Using this route, dose-related side effects are not seen, and efficacy is maintained in both animal studies and human clinical trials. Oral IFN administration appears to mimic a natural innate immune response. As such, it may represent an alternative delivery strategy to make better use of these critical cytokines.

","PeriodicalId":80125,"journal":{"name":"Pharmaceutical science & technology today","volume":"3 6","pages":"Pages 193-197"},"PeriodicalIF":0.0,"publicationDate":"2000-06-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1016/S1461-5347(00)00264-9","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"21685152","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This is because the drug distribution then follows that of the carrier, rather than depending on the physicochemical properties of the drug itself.
