{"title":"Activity of Na+/K+-ATPase and of Ca++-ATPase under the action of adenosine triphosphate in experimental myocardial hypertrophy.","authors":"C Moisin,&nbsp;N Balta,&nbsp;V Filcescu,&nbsp;I F Dumitriu,&nbsp;G Stoian,&nbsp;G Petec","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>This paper presents the results of our research on the mechanisms involved in the modifications occurring in the activity of ionic pumps (Na+/K+-ATPase and Ca++-ATPase) at the level of the sarcolemma, the sarcoplasmic reticulum and mitochondrial membrane of the myocardial cell in experimental myocardial hypertrophy induced by isoproterenol. We also studied the effects of concomitant administration of adenosine triphosphate. Thus, we found the activity of the sarcolemmal Ca++-ATPase intensely increased under the action of isoproterenol, while Ca++-ATPase activity in the sarcoplasmic reticulum decreased significantly. At the same time, the Na+/K+-ATPase activity decreased both in the sarcolemma and in the sarcoplasmic reticulum. Administration of adenosine triphosphate induces opposite effects, of lower intensity, upon the activity of the two ATPase, that tend to offset the the effects of isoproterenol. This was found in the group of rats given concomitantly both isoproterenol and adenosine triphosphate. A better understanding of the processes involved in those modifications of membrane ATPase activity allows us to consider their different behaviour to isoproterenol and adenosine triphosphate as the expression of intrinsic mechanisms by means of which the myocardial cell intervenes in maintaining a physiological concentration of calcium ions (Ca++) that is necessary both in order to avoid a failure of the contractile apparatus and in order to preserve the mitochondria role of ATP supplier. In the present study we tested the differences between the background EEG activity (BEA) of the \"O\" and \"A\" rat types during restrained wakefulness and anesthesia with Ether and chloral hydrate (CHL). The differentiation of the psyhoneuroendocrine rat types was made using the level of painful sensitivity. 13 hypersensitive \"A\" and 14 hyposensitive \"O\" rats were selected from a 91 adult (140 g) male Wistar population using the distribution of the tail retraction time (TRT) during a tail-flick test. The epidural EEG activity, was quantified within the 1-30 Hz band by six numerical parameters: root mean square (RMS), mean spectral frequency (MSF), spectral edge frequency at 95% (SEF95) and the relative spectral power (RSP) within the Theta (4.5-7.5 Hz), Alpha (7.5-12 Hz) and Beta (12.5-30 Hz) bands. The quantification method used was able to detect statistically significant differences between the two psyhoneuroendocrine rat types during consciousness and light ether anesthesia but failed to show any differences during the deep CHL anesthesia. The particularities were shown to be topographically related with the fronto-parietal regions were \"O\" type showed a higher SEF95 during the awake restrained state. The pain sensitive \"A\" type showed a significant intrahemispheric theta RSP asymmetry both during consciousness and ether anesthesia while no such theta gradient could be shown for the \"O\" type. The differences between the \"A\" and \"O\" types are enhanced under light Ether anesthesia to which the \"A\" type is more resistant. {"title":"Incidence of morphofunctional lesions in the cervical region of patients with gall-bladder dyskinesia.","authors":"M Uluitu,&nbsp;C Teleianu,&nbsp;N Stoicescu","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>During research performed in humans of both sexes, aged 17-30, suffering from disorders of gall-bladder kinetics, we found a constant association between this disorder and a cervical syndrome with occipital cephalea, spasmophilia, pre-molar and molar dental decay, even to edentation. We believe the mechanism inducing those disorders originates in the malabsorption of vitamin D2, caused by the absence of biliary salts in the intestine. Our findings could be of interest for the therapeutic management of such cases. In conclusion, we describe a new clinical framework establishing a pathological pattern that is functionally and morphologically coherent. The pattern includes the motor-biliary condition that induces disorders in the mechanism of calcium metabolism control. It also includes major neuromuscle hyperexcitability in the pre-vertebral region associated with the biped posture. Functional and morphological lesions in the cervical region of the spine, with Arnold-type manifestations and spasmophilia prevail in the final clinical picture. This picture is very early associated with multiple dental caries that may progress to edentation. The significant perturbation of proteic biogenesis, the inhibition of HeLa tumoral cells cultures development, as well as the existence of a dose--response relationship argue the behaviour of these products as in vitro active cytostatic agents. Li-GLA 15 micrograms ml-1 was administered to both normal and LP-BM5 MuLV retroviral infected murine bone marrow cultures. After 2 weeks of treatment, numbers of progenitors being produced by infected/treated cultures were reduced to some 10% that of normal cultures. In the remaining 4 weeks, numbers of CFU-GM and BFU-E hematopoietic progenitors returned within normal range. The efficacy of Li-GLA in relieving retroviral hematopoietic bone marrow suppression correlates to a reduction in interleukin-4 (IL-4) secretion, normally elevated in association with LP-BMP5 infection. These data indicate that this reduction in bone marrow suppression of LP-BMP5 infected cells may be due to a killing of infected cells by the Li-GLA, rather than stimulating hematopoiesis as with other lithium compounds. Mixed saliva was sampled without stimulation during 2 periods of the day: 07:30-08:00 before breakfast and 17:30-18:00 before dinner. We determined: salivary flow rate, pH with Merck indicator and, after homogenization, the thiocianat with the FeCl3 method and LDH activity (the Norbert method adapted in our laboratory for saliva). Our study showed the same diurnal changes in flow rate and salivary pH in both diabetic and control groups: minimal values in the morning and maximal ones in the afternoon. In non-smoking diabetic patients the salivary thiocianat had maximal values in the morning and minimal ones in the afternoon; similar behaviour, but less obvious was observed in smoking diabetic patients and in the control group regardless of the smoking habit. LDH activity showed unsignificant diurnal variations in the diabetic patients. In the control group we found a significant decrease of LDH activity in the afternoon. While the heavy water induced relaxing effects, the deuterium-depleted water increased basal tone and vasoconstrictor responses at phenylephrin, noradrenalin and angiotensin.
