{"title":"Dysregulation of miR-330-3p is Involved in the Occurrence and Development of Pulmonary Arterial Hypertension Caused by Congenital Heart Disease.","authors":"Xiaobing Yin, Jin Li, Yiran Luo, Tianzong Li, Ya Wang, Xiaolan He, Xia Tan, Manxia Liu, Xinghui Liu, Jianmei Shen","doi":"10.14744/AnatolJCardiol.2025.4807","DOIUrl":"10.14744/AnatolJCardiol.2025.4807","url":null,"abstract":"<p><strong>Background: </strong>The study aimed to investigate the expression of miR-330-3p and its clinical and functional performance in congenital heart disease-associated pulmonary hypertension (CHD-PAH).</p><p><strong>Methods: </strong>The expression of miR-330-3p in CHD-PAH and hypoxiatreated human pulmonary artery smooth muscle cells (HPASMCs) was assessed using reverse transcription quantitative polymerase chain reaction (RT-qPCR). The receiver operating curve was conducted to evaluate the clinical diagnostic value of serum miR-330-3p in CHD-PAH. In cytology, CCK-8 and Transwell migration assays were performed to assess the func-tional role of miR-330-3p in hypoxia-induced HPASMCs. The online TargetScan database and dualluciferase reporter assays were employed to explore the downstream target of miR-330-3p.</p><p><strong>Results: </strong>Compared with healthy controls and patients without PAH, miR-330-3p expres-sion was upregulated in patients with PAH. Serum miR-330-3p expression has relatively high area under the curve (AUC) values in differentiating CHD-PAH patients from con-genital heart disease (CHD) patients and healthy individuals. Silencing miR-330-3p weakened the increased cell proliferation, migration, and inflammation caused by hypoxia in HPASMCs. KLF-10 was identified as a putative target of miR-330-3p. Knockdown of KLF-10 could partially reverse the influence of miR-330-3p knockdown in hypoxiainduced HPASMCs.</p><p><strong>Conclusion: </strong>Upregulation of miR-330-3p might have diagnostic value for predicting individuals suffering from CHD-PAH. {"title":"Protective Effect of Amifostine on Radiotherapy-Applied Cardiovascular Tissue.","authors":"Gökay Taylan, Murat Çaloğlu, Vuslat Yürüt Çaloğlu, Tülin Yalta, Nurettin Aydoğdu, Kenan Yalta, Meryem Aktoz","doi":"10.14744/AnatolJCardiol.2025.4996","DOIUrl":"10.14744/AnatolJCardiol.2025.4996","url":null,"abstract":"<p><strong>Background: </strong>The present study evaluates the protective effect of amifostine (AMI) on acute toxicity in large vessels and the heart in rats with radiotherapy (RT) applied to the thorax.</p><p><strong>Methods: </strong>Twenty-one Wistar albino rats were randomly assigned to 3 groups: Alone RT (n = 7), amifostine plus RT (AMI+RT, n = 7), and control (n = 7) groups. The rats in the RT and AMI+RT groups received a single dose of 20 Gy radiation to the entire thorax. Prior to irradiation, AMI was administered intraperitoneally at a dose of 200 mg/kg, 30 minutes before the procedure. Five days after irradiation, the levels of p53, CD68, and COX in the vascular tissue (aorta) were measured, along with the levels of malondialdehyde (MDA) and glutathione (GSH) in the aortic and heart tissues.</p><p><strong>Results: </strong>The results showed that the level of MDA significantly increased after irradiation, but GSH levels did not change (P < .001 and P = 0.138). Malondialdehyde levels were significantly reduced by AMI, and GSH levels increased (P = .031 and P = .007). When comparing the control group with AMI + RT, MDA and glutathione levels were similar (P = .314 and P = .136). The aim of this study was to evaluate the effectiveness and safety of ultrasound-assisted catheter-directed thrombolysis (USAT) compared to anticoagulation therapy alone in patients with IHR PE.</p><p><strong>Methods: </strong>A total of 425 patients diagnosed with acute PE and determined as IHR, 203 of whom underwent USAT, and 222 patients receiving only anticoagulants as the control group, were included. Baseline and post-treatment right ventricle (RV) function in echocardiography, tomographic RV/left ventricle (RV/LV) ratio, Qanadli score (Qs), and % changes from baseline were taken as primary effectiveness outcomes. For safety outcomes, major and minor bleeding and in-hospital all-cause death were adopted. Propensity score analysis was performed to reduce confounders and bias.</p><p><strong>Results: </strong>The USAT treatment was found to be associated with improved RV function and decreased Qs, but no significant effect was observed on the RV/LV ratio and its change. Bleeding events were more frequent in the USAT group (P < .001 for both), and no difference was observed in terms of mortality.</p><p><strong>Conclusion: </strong>The study, based on real-life data, has shown that a moderate-dose, slow-infusion tissue-type plasminogen activator regimen is superior to anticoagulant therapy alone in terms of reducing pulmonary arterial thrombus burden, restoring RV dysfunction, and improving clinical outcomes in acute PE patients at IHR.
