{"title":"[An immunoenzyme method of diagnosing familial hypercholesterolemia].","authors":"I V Fuki, S N Preobrazhenskiĭ, Kh A Kurdanov, V S Repin, Kh A Khashimov","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>A new modification of enzyme immunoassay: enzyme-linked-immunoreceptor assay (ELIRA)--was used to study the activity of LDL-receptors on cultured fibroblasts from 10 patients with elevated plasma cholesterol levels, IHD, accelerated atherosclerosis and xanthomatosis. Four patients were found to have heterozygous form of familial hypercholesterolemia. We have also shown that the results of ELIRA were quantitatively similar to the data obtained by traditional radioisotopic method. {"title":"[Comparison of methods for computer and visual assessment of stenosing lesions of the coronary arteries. 1].","authors":"E V Pomerantsev, M L Blank, A A Smirnov, S A Abugov, A P Savchenko","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>A system of computer analysis of coronary arteries stenotic lesions developed in the USSR Cardiology Research Centre, Academy of Medical Sciences of the USSR, is described. Reliability of the computer analysis data was assessed, and computer and visual methods were compared on the basis of phantom study. The study was performed in 5 models of stenotic coronary arteries, the main parameters of which were measured mechanically as well as by computer and visual analysis of their images. Both methods proved to give highly reproducible intraobserver and interobserver results. {"title":"[Calcium metabolism of the thrombocytes in ischemic heart disease].","authors":"V G Kiktev, M Iu Men'shikov, E M Bugriĭ, A V Kuz'min, Iu A Karpov","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Changes in Ca++ concentration were studied in platelets of patients with acute myocardial infarction, unstable or stable angina pectoris and those of healthy donors by means of fluorescent probe Quin-2 AM. Influence of aggregation inductors on the process of Ca++ level increase in these cells was also investigated. Intracellular Ca++ concentration increased in patients with acute myocardial infarction in the presence of PAF 2.10(-7) M (1337 +/- 255 nM) and in the presence of ADP 10(-5) M (1767 +/- 296 nM). A certain increase in calcium responses to stimulators was observed also in patients with unstable or stable angina pectoris. Dynamic follow up of patients with acute myocardial infarction starting from the 14th day demonstrated significant fall in intracellular Ca++ concentration (from 1767 +/- 296 nM to 834 +/- 186 nM, p less than 0.01). Platelet sensitivity to inductors during the course of therapy did not change significantly in patients with stable angina pectoris and tended to decrease in those with unstable angina pectoris (from 707 +/- 274 nM to 410 +/- 95 nM, p greater than 0.05). Cells with little changes were coincided with apparently changed cardiomyocytes in which hypercontracted or atrophied sarcomeres with distended Z-lines, glycogen disappearance and swollen or hypertrophied mitochondria were seen. Myocardial content of ATP did not change but that of phosphocreatine was decreased by 45 percent. The pump function of the isolated heart was moderately lower that was associated with almost twofold increase in left ventricular diastolic stiffness suggesting a deterioration of its filling. A rise in Ca++ concentration in the perfusate exerted similar positive inotropic effect on hearts of both groups which was associated with a prominent fall of diastolic stiffness. The results obtained in vivo allow a following series of comparative antihypertensive and vasodilating activity: alpha rANF greater than APII greater than AP7-27. alpha rANF, APIII and APII did not differ significantly in vasodilating activity on isolated vessels; analogue 7-27 was less active. Sensitivity to verapamil tested by EC50-half-maximal inhibition of responses to hormones can be weakened in vitro by adrenalin and strengthened by papaverin. Thrombocytes of hypertensive patients were individually sensitive to combination of papaverin and verapamil. Computer tomography is proved to be a valuable noninvasive method for elucidation of retroperitoneal space organs pathology which can cause arterial hypertension. Hormonal-active tumors are better identified when patients are purposely selected according to the catecholamine and VMA levels as well as the state of renin-angiotensin-aldosterone system.
