Two isoforms have been detected in human erythrocytes, and six in nucleated avian erythrocytes. Multiple alternative splicing of human reticulocyte mRNA for protein 4.1 implies the possibility of even more isoforms. Structural and functional dissimilarities between isoforms, and their differential expression during the course of erythropoiesis are not well understood. Here, we show that only protein 4.1b is present in the immature reticulocytes of anaemic patients. The ratio of 4.1a to 4.1b in mature reticulocytes of healthy individuals was 1.25. This ratio was 2.0 in healty mature erythrocytes, and 0.7 in peripheral red blood cells from anaemic patients. It is presumed that the transition occurs via a short-lived oligomer whose formation is accelerated by ATP. In the context of this model, the non-Michaelis-Menton kinetics of the enzyme can be explained by interprotomer interactions. After solubilization of the enzyme with octaethylene glycol dodecyl ether, the hydrolysis of ATP follows ordinary Michaelis-Menton kinetics. The validity of the model is also supported by radiation-inactivation experiments with a nucleotide (GTP) which does not accelerate the E2----E1 transition, as well as by experiments with a low concentration of ATP. {"title":"Investigation of antigenic structure of attenuated and virulent Venezuelan equine encephalomyelitis virus by means of monoclonal antibodies.","authors":"I A Razumov,&nbsp;E V Agapov,&nbsp;A V Pereboev,&nbsp;E V Protopopova,&nbsp;S D Lebedeva,&nbsp;V B Loktev","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>A comparative study of the antigenic structure of virulent strains and attenuated vaccine strains of Venezuelan equine encephalomyelitis virus (VEEV) by means of monoclonal antibodies has made it possible to investigate the antigenic structure of the envelope glycoproteins E1 and E2, and to specify their role in the development of antiviral immunity. On the E1 glycoprotein there are five nonoverlapping antigenic sites consisting of eight epitopes that are recognized by monoclonal antibodies; six sites consisting of twenty epitopes were found on the E2 glycoprotein. The monoclonal antibodies against four sites protect the animals from lethal infection with the virulent strain, Trinidad donkey. Out of the thirteen epitopes identified as being responsible for antiviral immunity, three are changes in the TC-83 strain, and six belong to two sites in the strain 230. The role of gluconeogenesis.","authors":"V P Shelepov,&nbsp;V A Chekulaev,&nbsp;G R Pasha-Zade","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The interrelationship between glycogen biosynthesis and do novo glucose formation in rats was studied by determining the oxaloacetic acid, ATP, and glycogen levels in the tissues of healthy starved rats and during the growth of transplantable hepatomas with different rates of growth. From the results obtained it was proposed that glycogen biosynthesis and de novo glucose formation are mutually enhanced and coupled processes. The validity of this conclusion was confirmed by in vitro model experiments involving the use of isolated-liver perfusion. Results of a multiple regression analysis indicated that the efficiency of gluconeogenesis and glycogen deposition is determined by the supply of oxygen to the liver, by the oxaloacetic acid content of this organ, and also by the degree of reduction of the glucose precursor. It was also shown that the ability of glucocorticoids and glucagon to enhance gluconeogenesis is dependent on the supply of oxygen to the liver. To identify proteins involved in the initiation of RNA replication at different stages of infection a highly specific affinity labelling technique was used. In samples of the nuclear fraction taken from cells 45 h after infection (late stage), affinity labelling with aldehyde-containing derivatives of ATP and elongation of this label with [alpha-32P]GTP identified a polypeptide with a molecular mass of about 69 kDa. By means of affinity labelling with aldehyde-containing analogues of GMP, GDP, and GTP as initiation substrates and [alpha-32P]ATP as the elongation substrate, a polypeptide of 100 kDa was selectively modified in the nuclear fraction of cells at the early stages of infection (8 h). These proteins were immunostained with TBEV-specific antibodies, and were identified as the nonstructural TBEV proteins NS3 and NS5, respectively. Collagens precipitated from the culture medium and intracellular collagens were detected by enzyme-linked immunosorbent assays with rabbit polyclonal monospecific antibodies. The assay could detect 10 ng protein per well, and intraassay coefficients of variation were less than 5%. Secreted collagens types I and III were also analysed by electrophoretic separation of [3H]-labelled alpha-chains under nonreducing and reducing conditions. These methods revealed a structural abnormality in one patient with clinical features of Marfan syndrome, and led to the postulation that the abnormality resulted from substitution of cysteine for another amino acid in collagen type I.
