Its possible causes may be free-hemoglobin (F-Hb), stroma freed from destroyed red blood cell, and their combined action. Especially, a great amount of F-Hb is liable to flow out during extracorporeal circulation. The plasma fractionation using a membrane has so far been applied to remove F-Hb (1). The molecular weight of F-Hb of 68,000 which is nearly equal to that of albumin has made it still difficult to separate F-Hb effectively. Upon transfusion, this purine may be uptaken by the endothelial cell and oxidized in a post-ischemic or post-anoxic environment with production of oxygen-derived free radicals. We have tested this hypothesis with a isolated perfused rat heart model monitoring the recovery of the heart function from 20 min anoxia in the presence of 0.1 mM hypoxanthine or xanthine. Addition of 0.1 mM guanine minimized the fraction of hypoxanthine to be salvaged. The presence of hypoxanthine in the vascular space impaired the recovery of the end-diastolic pressure, left ventricular developed pressure, contraction rate, and coronary perfusion pressure. We conclude that intravascular hypoxanthine is oxidized by the endothelial cell xanthine oxidase contributing to the post-anoxic reoxygenation injury. Since the injury led by equimolar xanthine was nearly half of that observed for hypoxanthine, this injury appears to be correlated to the stoichiometry of the oxygen-derived free radical generating reaction.</p>","PeriodicalId":77498,"journal":{"name":"Biomaterials, artificial cells, and artificial organs","volume":"18 2","pages":"309-20"},"PeriodicalIF":0.0,"publicationDate":"1990-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.3109/10731199009117309","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"13519426","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Liposomes of varying lipid composition were prepared by detergent dialysis. All liposome suspensions showed non-Newtonian viscous behavior, as did erythrocyte suspensions. The viscosity of suspensions of Hb liposomes with a bilayer consisting of egg yolk lecithin and phosphatidylserine was almost identical to erythrocyte suspension viscosity. When cholesterol was included in the lipid component of the Hb liposomes, the dependence of viscosity on the shear rate decreased (flattening of the curve), but overall suspension viscosity became higher than that of erythrocyte suspensions.
