During the first three days after the operation enzyme activity is higher in nuclei obtained from the intact (left) side of the brain while later on enzyme activity is more pronounced in nuclei obtained from the right (sympathectomized) side of the brain. The SOD obtained have been shown to have three isoenzymes (a, b, c). A study of the catalytic and macromolecular properties of the SOD obtained from grey and white matter of cerebral hemispheres as well as their optical and electron paramagnetic resonance spectra indicates that both proteins possess similar properties. {"title":"[Molecular mechanisms of mediator secretion].","authors":"R N Glebov,&nbsp;G N Kryzhanovskiĭ","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>A review is presented discussing problems of transmitter localization in nerve endings, their replenishment in synaptic vesicles (SV), reuptake of transmitters and their degradation products from the synaptic cleft, the structural variations in the presynaptic membrane (pre-SM) during rest and excitation and the role of contractile proteins in the mechanism underlying transmitter secretion. A hypothesis is proposed about the universality of the mechanisms involved in transmitter release and utilization and the key role of the membrane ATP-ase system in these processes. During depolarization of the pre-SM the increase in membrane permeability is ascribed to the loosening of protein-lipid bonds and inhibition of ATP-ase activity involved in transport mechanisms. During depolarization the number of complementary contacts between SV and the pre-SM increase probably through the action of myosin-like and actin-like proteins localized on the SV and pre-SM, respectively. The release of transmitters and polypeptides is thought to be initiated by an increased concentration of Ca2+ in the synaptoplasm which induces contraction of the actomyosin-like complex. Changes in the Na-gradient brought about by the activity of Na, K-ATP-ase, are involved in the active reuptake of transmitters from the synaptic cleft. The newly-synthesized transmitters and those taken up from the synaptic cleft are stored in the SV by a Mg-ATP-ase dependent mechanism. Transmitters are stored in SV bound to acid polypeptides containing nucleotides. The data obtained have shown that in mitochondrial fraction the level of ammonium is high while its endogenous formation is less pronounced. A large amount of ammonium is formed from ADP and some from glutamic acid, especially in the presence of ADP. The level of ammonium is, on the contrary, low in the synaptosomal fraction and endogenous ammonium formation is high. In this fraction also there is ammonium formation from both ADP and GA but to a smaller extent than in the mitochondrial fraction. In the evaporated dialysate the enzymatic activity of the soluble protein fraction is completely recovered. The reduction of peptide hydrolase activity following dialysis is probably due to the decrease of ionic strengths. A comparison of the effect of the neurohormone with that of theophylline (10 mM) showed that very small amounts of the neurohormone brought about inhibition of enzyme activity that was equal to that observed from much larger concentrations of the theophylline. At this purification the major band on disc-gel electrophoresis is associated with the enzyme activity, although several minor components are also present. The arylamidase is -SH dependent and puromycin inhibited. It has activity toward lysyl and arginyl beta-naphthylamides as well as toward the beta-naphthylamides of a large variety of neutral amino acids. The enzyme is strongly substrate inhibited, making the relative order of reactivity to aminoacyl beta-naphthylamides dependent upon the substrate concentration chosen for comparison. It also shows activity toward certain di-, tri-, and oligopeptides. When more than one residue is cleaved, the release is sequential, starting from the amino terminus of the peptide. It appears therefore that pig brain neutral arylamidase is an aminooligopeptidase of broad specificity. We suggest that beta-naphthylamides are model substrates representing the N-terminal end of peptides with three or more residues.
