Antigen-antibody reactions were located on the shell of filarial eggs as well as on the cuticle and the interior of L3 and microfilariae. Using sera from Meriones unguiculatus, Mastomys natalensis and golden hamsters infected with D. viteae obtained at 7, 14, 21 and 28 weeks p.i. it was observed that the intensity of the immunostaining on several developmental stages of the filariae decreased with increasing duration of infection. This effect was more pronounced in the case of M. unguiculatus and M. natalensis than in the case of golden hamsters. For detection of surface antigenicity the simple procedure of drying the test material on microscopic slides proved to be time saving and equally specific as compared to the embedding in Epon of the filarial stages and the successive preparation of semithin sections. However, embedded filarial stages presented more intensive immunostaining than nonembedded stages. The T. cruzi strains are interspecifically distinguishable with the lectins from Euonymus europaeus, Tridacna crocea, Tridacna maxima and the human blood-group testserum anti-B from the T. vespertilionis strains. While the T. vespertilionis strains did react with anti-B and E. europaeus, the T. cruzi strains did not agglutinate. The T. cruzi strains were agglutinated by the lectins from T. crocea and T: maxima while the bat-trypanosomes showed no reactions. Using these lectins it was not possible to distinguish the bat-flagellates intraspecifically. With the lectins from Triticum vulgaris and Arachis hypogaea the T. cruzi strains could be distinguished. While the Ténuantépéc strain did agglutinate with A. hypogaea, T. cruzi strain Guatemala did react only with the lectin from T. vulgaris. The bat-trypanosomes were agglutinated only by A. hypogaea but not by T. vulgaris. Ten nodules contained adult parasites with an average sex ratio of 1.6 for female to male worms. The median worm burden was two filariae per person. The onchocercomata of two patients with severe localized onchocerciasis comprised one or two pairs of microfilariae producing worms whereas the microfilaria load in the skin of these patients was estimated to have been less than 100 000 microfilariae. One nodule contained no intact microfilariae but more than 10 000 small granulomas with all stages of degenerating microfilariae whereas the density of 0.08 live microfilariae per milligram in the skin near to the nodule was very low. None of the patients had received antifilarial drugs before nodulectomy. It is concluded, therefore, that these patients with severe localized onchocerciasis possessed the capacity naturally to kill microfilariae. In the nodules of these patients many degenerating microfilariae were observed which were surrounded by eosinophils or macrophages or which lay in small abscesses with neutrophil leucocytes or in small granulomas. The study sample consisted of 40 Yanomami inhabiting a savannah area at 950 m above sea level and 21 Yanomami residents of a tropical rainforest area at an altitude of 250 m. Stained intrauterine microfilariae, still within the egg case, exhibited a diffuse distribution of the enzyme in the early stages of embryonic development and a negative reaction at a more developed stage. Four of the five enzyme staining patterns described by Omar (1978) were found in the 3157 microfilariae examined from skin snips. Their distribution was: Type I--17.2%, Type III--0.5%, Type IV--75.6% and Type V--6.6%. No examples of Type II were observed. The results indicate that acid phosphatase patterns of the Upper Orinoco Onchocerca strain most resemble those of strains from Guatemala and Yemen, and are different from the African strains found in Upper Volta and Liberia. The enzymes tested were: nonspecific esterase (NSE, E.C.3.1.1.), phosphoglucomutase (PGM, E.C. 2.7.5.1), glucose-6-phosphate dehydrogenase (G-6-PD, E.C. 1.1.1.49), glucosephosphate isomerase (GPI, E.C. 5.3.1.9), malate dehydrogenase (MDH, E.C. 1.1.1.37), alcohol dehydrogenase (NADP+) (ADH, E.C. 1.1.1.2). Their enzyme types were related to those of T. cruzi. The comparison of enzyme patterns of the six enzymes has shown that each species was characterized by species-specific enzyme profiles. Among the stocks of the European species, T. dionisii, and T. vespertilionis, variations of the enzyme patterns of PGM, G-6-PD and GPI suggesting that the final status of this subspecies is probably not yet established. In relation to T. cruzi it has been found that T. dionisii showed identical enzyme profiles with group II of T. cruzi. For T. vespertilionis no enzyme types identical with T. cruzi were detectable. Their antigenicity in natural infections of Meriones unguiculatus was investigated by immuno-recognition with pooled sera of infected hosts. The antibodies were labelled by radioiodinated protein A and visualized by autoradiography. Immunorecognized protein bands were demonstrable in all stages, some of them being stage specific, others common to several or all stages. Patients who had already received chemotherapeutical treatment during the first 5 years following the onset of the disease developed fewer cases of lagophthalmus in both types of leprosy than patients who remained untreated for longer than 5 years. A comparable positive influence of early chemotherapy on the incidence of posterior synechiae of the iris could not be proven. Though only one among 59 male animals yielded a positive result, 46.5% of the female cattle showed to be infected with the parasite. Comparing the presence of adult worms in the gastrosplenic ligament and that of larval stages in the skin a correlation of 87.5% could be obtained. Most of the microfilariae were concentrated in the umbilical region but with severely infested animals they also were found in the skin of the udder, head, ear and neck. Evidence of infection was obtained by clinical, radiologic, echographic, and/or computerized axial tomography examinations in only one of three DD5-negative individuals, and in 2 of 4 students showing a questionable reaction of identity with arc 5 in DD5, who were positive in LA and/or IHA. In contrast, cysts could be demonstrated in all DD5-positive cases, whether positive or negative in the agglutination procedures. These cysts in DD5-positive cases included the smallest cysts (1-1.2 cm wide) detected in surveys to date. Comparison of data obtained in the immunodiagnostic survey and in a review of hospital records was used to assess the relative contribution of each method in estimating the prevalence of infection.
