Our results point to a statistically significant difference in the oestrous phase of the posterior cortex and ovarium levels, comparing control rats with the enucleated ones. In the dioestrous phase, the modifications that we have encountered are at the amygdala and ovarium. Various pharmacological agents, such as histamine, phencyclidine, phenoxybenzamine (PBZ) and gamma-aminobutyric acid (GABA), were tested in this paradigm for evaluation on incidence of ovulation, number of ova released and degeneration in the ova shed. A dose of 25 iu of pregnant mare serum gonadotropin (PMS) was found to be most effective in inducing superovulation in these rats. The percentage of degenerative eggs obtained was least in this dose group. Histamine and GABA appeared to cause an increased number of oocytes to be released, while phencyclidine reduced the number of animals responding to PMS treatment. PBZ was effective in completely blocking ovulation only when given before the LH surge occurred in the treated animals. The quality of ova released was affected by GABA, phencyclidine and PBZ. GABA and phencyclidine reduced the incidence of degenerative oocytes while treatment of animals with PBZ at the time of PMS administration increased the degenerative oocytes. The significance of the microscopic study of seminal fluid in this syndrome is well emphasized. The possible relationship of the histological pattern to spermatogenic function and chromosomal constitution is discussed. Our conclusions are that most patients were azoospermic and the the volume of the ejaculate was low or absent. The decreased density of seminal fluid was the result of the absence of spermatozoa. The seminal pH remained normal during the study. With an increasing degree of reduction of spermatozoa, a decreasing testosterone level and increasing LH and FSH levels could be demonstrated. In normozoospermia, between 40 and 140 X 10(6)/ml, a direct correlation was found between FSH and sperm count, and, in the group between 40 and 100 X 10(6)/ml, a direct correlation between T and sperm count. {"title":"[Validation of the radioimmunoassay for rat luteinizing hormone (LH) (author's transl)].","authors":"A M Aranda Iriarte","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>A rat LH radioimmunoassay (RIA) using the immunoreactants provided by Dr Parlow on behalf of the Rat Pituitary Hormone Program of the NIAMDD of the NIH is described. The instructions suggested by the NIH have been modified in order to reach a higher sensitivity. It is possible to use prepipetted frozen standards and the same labelled antigen over a period of 1 month, or longer, after labelling, provided the preparation is purified immediately prior to its use. These procedures, as well as the addition of antibody and buffer simultaneously, shorten considerably the time spent on a given RIA and decrease the inter- and intra-assay variability. The specificity of the method is also described. There is good parallelism between the standard curve and serial dilutions of plasmas and hypophyses of rats. Results show significant differences in the oxidative metabolism of the amygdala in both male rats and ovariectomized females on comparing the fasting group with controls; moreover, the frontal cortex of male rats also shows significant differences when comparing both groups. The weight of the adrenal glands is significantly lower in ovariectomized female rats under fasting conditions than in controls. Circulating oestradiol was measured by radioimmunoassay after chromatographic separation on microcolumns of Sephadex LH-20. Mean values of oestradiol concentrations and coefficients of variation for two 4-hour periods were calculated for each subject and for the whole group. Data were statistically evaluated by means of the Wilcoxon signed-rank test. Rapid fluctuations in plasma oestradiol levels were observed in all subjects. The pattern of the fluctuations was found to be irregular and different in each subject.
