{"title":"Endocrine studies in postmenopausal women during oral replacement therapy with unconjugated oestrogens.","authors":"J M Vasquez, C A Samaras, C Nezhat","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Two groups of postmenopausal women were seen at monthly intervals during a three-month trial of continuous therapy with oral unconjugated oestrogens. Ten women in the first group were administered daily Hormonin No. 1 containing oestriol (E3) 0.135 mg, oestradiol (E2) 0.3 mg and oestrone (E1) 0.7 mg. Eight women in the second group received Hormonin No. 2 containing E3 0.27 mg, E2 0.6 mg and E1 1.4 mg. E1, E2, E3 and dehydroepiandrosterone (DHA) as well as follicle-stimulating hormone (FSH) and luteinizing hormone (LH) were measured by radioimmunoassay. Maturation index of vaginal smears and clinical effects were also evaluated. Oral replacement therapy with these unconjugated oestrogens produced a significant elevation of E1 (p less than 0.05) and E2 (p less than 0.05) to values corresponding well with the premenopausal range measured in our laboratory. Postmenopausal levels of FSH and LH showed only a moderate but significant decrease (p less than 0.05). There was consistent relief of vasomotor symptoms. One case of endometrial focal adenomatous hyperplasia uncovered during the period of treatment was transformed to functional secretory endometrium after an appropriate course with progestogens. {"title":"Ovestin vaginal cream and suppositories for the treatment of menopausal vaginal atrophy.","authors":"R Trevoux, W H van der Velden, D Popović","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Eighty-two postmenopausal women with vaginal atrophy and associated symptoms were treated with either Ovestin vaginal cream (54 women) or vaginal suppositories (28 women) containing 0.5 mg of E3/dose, daily for 3 weeks. A maintenance dose of 0.5 mg of E3 twice weekly was applied by all patients for 5 weeks, and by 27 of them for up to 16 weeks. Variables studied were clinical and colposcopic findings, the Maturation Index (MI) and Maturation Value (MV), cervical mucus ferning (F) and spinnbarkeit (S) and endometrial biopsies (obtained pretreatment and after 3 weeks in 15 of the patients). Furthermore, the bio-availability of circulating unconjugated E3 for up to 8 hours following a single dose of Ovestin cream was studied in 10 of the patients. Clinical and colposcopic findings indicated that the treatment had a very favourable effect in all patients. This was reflected in the pronounced change in vaginal smears, indicating a strong oestrogenic effect. There was a slight to moderate effect on cervical mucus. Endometrial biopsies showed that endometrium remained atrophic in all 15 patients in whom biopsies were obtained. A maintenance dose of 0.5 mg of E3 twice weekly appeared to be sufficient to maintain the beneficial effect. The effects of the medication on the climacteric symptoms, the subjective sensations, the plasma FSH levels and endometrial histology were studied. In the treated group compared with the control group the relief or improvement of the following climacteric symptoms were recorded: perspiration, palpitations, irritability and backache. A favourable effect on the subjective sensations was noted in both groups, although no significant difference for the group which received Org OD 14 was found. At the end of the treatment with Org OD 14, the FSH levels were found to be greatly reduced in comparison with the basal values; this, however, was not the case with the placebo group. With regard to endometrial histology, no sign of hyperplasia was found in any of the patients. No relevant side effects or symptoms of estrogenic or androgenic stimulation were recorded. {"title":"Fertility after operation for uterus bicornis.","authors":"N T Spirtos, A V Comninos","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Sixty-three properly selected patients with sterility due to uterus bicornis underwent operations to restore fertility, Of these 63, 12 suffered from primary sterility and 51 from secondary sterility. The 51 patients with secondary sterility had had a total of 127 pregnancies before operation. Of these 127 pregnancies, 122 ended in early or late abortion and five in premature delivery, giving 5 live births and a fetal wastage rate of 96%. Of the 63 patients, 42 became pregnant after operation. Of these 42, 14 had more than one pregnancy. The total number of pregnancies was 59. Of these, 32 went to term, 15 ended prematurely and 12 ended in abortion. The 59 pregnancies resulted in 45 live births. {"title":"Effects of sialoadenectomy and castration in female rats.","authors":"A Menéndez-Patterson, A Velasco-Plaza, B Marin","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Previous work on the role of the submaxillary glands in the control of the oestrous cycle in rats has been extended to castrated rats in order to avoid the overlapping between sexual and salivary hormones. Animals were sacrificed 30 days after sialadectomy or pseudosialadectomy. The data show that simultaneous castration and sialadectomy increases significantly the glucaemia level and decreases the weight of the adrenal glands. Non-simultaneous castration and removal of the submaxillary glands decreases the weight of the parotid glands. This effect decreases when both actions are simultaneous. On the other hand, castration produces an important decrease in QO2 uptake in tested structures. Removal of submaxillary glands produces a significant increase of QO2 in hypothalamus and thyroid glands. Simultaneous castration and sialadectomy at the anterior cortex, posterior cortex and parotid gland level shows similar results with respect to desalivated rats; other structures show results similar to the castrated group values. {"title":"Effect of sex, testosterone propionate and trienbolone acetate on the rate of growth and myofibrillar protein degradation in growing young rats.","authors":"S Santidrian, J R Thompson","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The urinary output of N tau-methylhistidine (3-methylhistidine: 3-Mehis) was determined to evaluate the effect of testosterone propionate (TP, 1 mg/100g body weight per day) and trienbolone acetate (3-oxo-17 beta-hydroxy-4,9,11-estratriene acetate, TBA 1 mg/100g body weight per day) on the rate of myofibrillar protein breakdown in young growing intact and adrenalectomized (AdX) male and female rats. Injections were given subcutaneously for 14 days. Body weight changes were recorded daily throughout the experiment. Compared with the intact control rats, male-AdX rats had a significant (p less than 0.05) reduction in both growth and 3-Mehis output (and, therefore, a reduction in the rate of myofibrillar protein degradation). Administration of the anabolic steroids to these rats had no significant effect on growth but increased the rate of 3-Mehis output to the normal range in the case of TP, and above the normal range (p less than 0.05) in the case of TBA. However, compared with the intact female rats, female-AdX rats showed a significant (p less than 0.05) increase in 3-Mehis output. Administration of either TP or TBA to these animals significantly (p less than 0.05) increased growth and reduced 3-Mehis output. The response of myofibrillar protein degradation in AdX rats to TP or TBA administration is strongly dependent on the sex of the animals. Ten groups of mothers were studied, including 234 control subjects. The injectable progestational contraceptives caused an improvement in the amount of milk and its protein concentration, whereas the IUCDs did not influence the lactation pattern. Sulpiride was an ineffective contraceptive but a potent lactation stimulant. {"title":"Endocrine profiles of sows during the oestrous cycle.","authors":"G Enne, L Perotti, G Delrio, P Inaudi, M d'Istria, R Pierantoni, F Citarella, M A Musaro, C Monittola, A R Genazzani","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>We measured the plasma concentrations of luteinizing hormone (LH), oestradiol (E2), progesterone (P), 17 alpha-hydroxy progesterone (17P), androstenediona (A) and testosterone (T) at oestrus and during the oestrous cycle for four consecutive cycles in a group of 15 normal sows. The results show that at oestrus the peak LH value was preceded, 24 hours earlier, by an E2 peak, and indicate that the LH rise begins when E2 concentrations reach their highest value. During diestrus, concentrations of LH and E2 were constantly low, P and 17P were characterized by lowest concentrations during the oestrous period which showed significant (p less than 0.001), progressive increases from the second day after the LH ovulatory peak, to reach their highest values after 8-14 days. The 17P decrease in proestrus precedes that of P.T and A concentrations showed a significant (p less than 0.001) increase 2 days before the LH ovulatory peak; high plasma concentrations of both androgens were maintained until the LH peak occurred. Measurements taken in consecutive cycles in the same animals showed a high reproducibility of the hormone concentrations examined, which showed similar patterns and values in each of the cycles studied.
