{"title":"Properties of hog cholera viruses recently isolated in Japan.","authors":"M Shimizu,&nbsp;Y Shimizu","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Hog cholera (HC) viruses newly isolated in Japan in 1980 and 1981 were examined for pathogenicity and serological properties by the neutralization test with antisera against bovine viral diarrhea-mucosal disease (BVD . MD) and HC viruses. Five of 23 isolates examined were neutralized poorly by BVD . MD antibody, but well by HC antibody. On the contrary, 15 isolates were neutralized readily and two isolates moderately by BVD . MD antibody. The other one reacted poorly with either HC or BVD . MD antibodies. The isolate neutralized poorly by BVD . MD antibody was more highly pathogenic than those neutralized readily. All the chicks hatched suffered from anemia and died at 10 to 15 days of age with bone marrow aplasia. Specific pathogen free laying hens were inoculated with CAA, and eggs were collected from them over a period from 1 to 28 days after inoculation. Two of 67 chicks hatched from the eggs revealed anemia at 14 days of age. CAA was recovered from 3 of 40 chicks. When passaged 19 times in MDCC-MSB1 cell cultures, it produced anemia of the same severity in chicks as it did before passage. Titration of the infectivity of CAA was performed successfully with subcultures of MDCC-MSB1 cell cultures which had been inoculated with serial tenfold dilutions of infected material. In it, no infected cultures could be subcultured. The propagation of CAA was also proved in the MD cell line, MDCC-JP2, and the avian lymphoid leukosis (LL) cell line, LSCC-1104B1, but not in the two MD cell lines, MDCC-RP1 and MDCC-BP1, or in the two LL cell lines, LSCC-1104X5 and LSCC-TLT. For the preparation of antigen, Mycoplasma hyopneumoniae was grown in broth medium containing horse serum for 14 days by shaking culture. Anticomplementary activity of antigen was eliminated by addition of complement and heating at 56 degrees C for 30 minutes. Storage of this antigen at 4 degrees C or at room temperature for 12 months had no significant effect on antigen titer. In the complement fixation test with this antigen, complement-fixing antibodies could be detected up to 41 weeks after experimental inoculation by KIO4 treatment of serum. In it, closed-colony mice of the CFW strain were inoculated intravenously with seven strains of the virus at 7 days of pregnancy. After that, an attempt was made to recover the virus from placenta and fetus, so that the infection rate of each strain might be determined. As a result, the infection rate was high for both placenta and fetus in the case of the AS-6 and Sagara strains both of which had undergone three passages in the mouse brain. The placental infection rate was high and the fetal infection rate relatively low in the case of the JaGAr01 and Fuji strains which had undergone 7 and 150 passages, respectively, in the mouse brain. The infection rate was very low for both placenta and fetus in the case of the Nakayama-Yakken strain which had undergone more than 100 passages in the mouse brain. There was no difference in the severity of viremia after inoculation between the AS-6 and Fuji strains. Both placental and fetal infection rates were low in the case of the JaTH160 strain which had undergone passages in mice by intraperitoneal inoculation and which presented a strong peripheral infectivity and induced a severe viremia after inoculation. Neither placental nor fetal infection occurred in the case of the S- strain used as live virus vaccine. It was recovered from the brain and rectal contents even 49 days PI, when the experiment was finished. In chicks inoculated with CAA at 28 or 42 days of age, CAA was also proved to multiply in all the organs, except the brain. It disappeared from these chicks earlier than from the chicks inoculated at 1 day of age. The mixture was refluxed with hydrochloric acid, and the supernatant was obtained. The supernatant was then adjusted to the pH of the isoelectric point of casein and the resulting precipitate collected. The precipitate was further hydrolyzed with papain at a neutral pH. After acidification of the reaction mixture, the supernatant and the precipitate were separated. The supernatant fraction, which was toxic to induce gizzard erosion, was submitted to a gel filtration of Sephadex G-25, and resolved into five fractions. Gizzard erosion-inducing toxicity was found only in the second fraction. The non-toxic second fraction was also prepared by the same treatment from the non-toxic material of heated casein. The characters of these toxic and non-toxic second fractions were compared by using several analytical methods. Thin layer chromatography, as well as gel permeation chromatography, revealed that more than eight components having widely distributed molecular weights existed in both second fractions. By reversed phase chromatography, these fractions were found to consist of various peptides (or peptide-like compounds). Some characterizations, however, were performed. It was visible around bacteria which had been fixed in a ruthenium red-containing fixative. It was composed of electron-dense spikes and extended outwards from the bacterial surface. It was not found around bacteria which had been treated with a fixative devoid of ruthenium red. Bacteria were seen scattered among blood cells.
