{"title":"[An evaluation of the biochemical typing of Klebsiella cultures].","authors":"S I Pokhil, L A Kosobutskiĭ","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The paper deals with the results of analysis of 219 strains of Klebsiella using 12 tests--fermentation of adonite, asparaginic acid, sodium citrate, dulcite, d-tartrate, glutamic acid, inosite, L-proline, sodium malonate; reactions with methyl red, Foges-Proscauer, with 5-ASA. A scheme for determining fermentovars of Klebsiella is suggested which includes the tests--fermentation of adonite, dulcite, d-tartrate, glutamic acid; color reaction with 5-ASA. The groups of Klebsiella different in origin are characterized by nonhomogeneous distribution of different fermentovars. Due to the data obtained both intracellular and extracellular RNAses of A. laidlawii possess similar specificity to different polynucleotides. Both RNAses preferentially break cytidine-bonds. {"title":"[The determination of the location of the antigens of the causative agent of melioidosis at the ultrastructural level by using monoclonal antibodies].","authors":"V I Kapliev, N N Piven', N P Khrapova","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The indirect immunoperoxidase method with the use of monoclonal antibodies has been employed to determine the localization of antigens of melioidosis agents. The formed reaction products were then detected by the electron microscopic analysis. The most important in melioidosis pathogenesis antigens 6 and 8 (AG 6 and AG 8) were determined to localize on the cell wall and in the capsule-like mucous layer, respectively. Monoclonal antibodies to AG 8 due to their marked cross-linking properties promote stabilization, preservation and detection of labile melioidosis agent. {"title":"[The chromatographic properties of the DNA-dependent DNA polymerases from Acholeplasma laidlawii PG-8].","authors":"S V Bezuglyĭ, I G Skripal', V V Babichev","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The DNA-dependent DNA-polymerase (DNA polymerase I which is not sorbed on the column with DEAE-cellulose, and DNA-polymerase II, which is absorbed by this column and is eluted from it by 0.3 M of NaCl), have been isolated from Acholeplasma laidlawii PG-8. DNA-polymerase I in homogeneous state was obtained as a result of the stepwise treatment by heparin-sepharose (elution at 0.35 M of NaCl) and poly-U-sepharose (elution at 0.3 M of NaCl). It was presented on the electrophoregram by one polypeptide with molecular weight of 72 kDalton. {"title":"[The use of a CHO cell culture in studying Vibrio cholerae grown under different conditions].","authors":"O I Sal'nikova, V V Lobanov, N Ia Shimaniuk","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Studies of biological activity of cholera vibrios in cultures of chinese hamster ovary cells (CHO) have revealed their strong dependence on culture conditions. Elongation of CHO cells is caused only by choleragenic strains. Under stationary conditions of culture the vibrios were found to release haemolisin into the medium and had a cytotoxic effect. Most of cytotoxic supernatants exhibited a neuraminidase activity. Proteolytic activity was less dependent on the vibrio culture conditions. Rickettsiella was observed till the 66th day in the primary cultures and till the 16th day after the first passage. Binary division of the cells has been found only in the primary cultures; a complex cycle of the Rickettsiella development ceased at the stage of formation of the crystal-forming cells. {"title":"[The level of spontaneous phage production and sensitivity to melioidosis phages of museum cultures of Pseudomonas pseudomallei].","authors":"I I Denisov, V I Kapliev","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The phage-producing activity and sensitivity of museum cultures of a melioidosis agent to melioidosis phages have been studied. Most cultures show spontaneous phage-production, though its level differs in strains. Some of the melioidosis cultures demonstrate lack of phages. The fact that these strains have different sensitivity to chloroform lysate of cultures producing phages indicates their possible lysogenic state. Electron microscopy has shown the presence of at least tow morphological types of phages in P. pseudomallei C-141. They are typified by their sensitivity to bacteriocins and by their production using colicinogenic and indicating strains from collection of P. Fredericq [correction of Frederick], D. G. Kudlaĭ, N. I. Koshanova as well as klebocinogenic K-type cultures of Klebsiella previously suggested by the authors. Investigation results have shown sufficient stability of a bacteriocinotype of the cultures confirmed by the population analysis.
