{"title":"[The statement of nuclear volume, DNA content and nuclear form in tissue sections (author's transl)].","authors":"H J Anton,&nbsp;R Thelen","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>15 days after amputation of the forelimb of young postmethamorphic Triturus vulgaris DNA-content and volume of the nuclei of the mesodermal blastema cells have been investigated in longitudinal sections of 2 microns in thickness by Feulgen-photometric and electronic determination of area. Discrimination of the DNA-synthesis phase was carried out using microautoradiography after \"in vivo\" labeling with tritiated thymidine. Volume and DNA-content of the nuclear sections have been found to be positive correlated. The DNA-synthesising part of the population shows characteristic differences to the mean value of volume and DNA-content of the remaining population. The factor circumference 2/area is suitable to characterize the differentiation degree of cells.</p>","PeriodicalId":76159,"journal":{"name":"Microscopica acta. {"title":"[Cell activation and nuclear volume (author's transl)].","authors":"H J Anton,&nbsp;W Elsen","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Freezing dried semithin sections (1 micron) coloured with nuclear fast red are suited for determination of volume and activation state of cells and nuclei by means of electronic image analysis. Additional investigations of the proteinsynthesis with help of autoradiographs after incorporation of tritiated amino acids affirm the informations. Magnification of the nucleus together with a simultaneous increase of the proteinsynthesis per unit volume characterize the Stratum basale near the amputation level as the most active area of the regeneration epidermis. The cell volume is correlated with the alterations of the nuclear volume. Determination of the mean relative volume of nuclei with aid of measurements of nuclei sections suffices to get informations about cell activity.</p>","PeriodicalId":76159,"journal":{"name":"Microscopica acta. This is true of both theory and practice. Indeed, it is often forgotten that Delesse not only introduced the stereological method, but also specified the necessary conditions for its use. To help make good this deficiency methods of stereological sampling are reviewed. In particular, the ratio estimate is discussed and examples are given. Estimation of the percentages of each cell class in populations of blood cells and cells from vaginal smears have problems in common. In most cases the bias of the ratio estimate is trivial. The formula for approximate calculation of the standard error of the ratio estimate requires more sampling units than most people realize. In addition, in artifical smears, a more even distribution of the cells is recommended.</p>","PeriodicalId":76159,"journal":{"name":"Microscopica acta. Under hypoxia the vascular volume was 50% higher than under hyperoxia.
