Population studies show several positive associations with antigens of the HLA-A and HLA-B series. Negative association have been found with antigens of the HLA-C series. Moreover the distribution of the RM3 factor is in Hardy-Weinberg equilibrium with that of the HLA-C alleles. However, serological investigations (namely the 'lysostrip' technique) bring evidence that RM3 serum does not appear to contain antibodies directed against any of the antigens of the HLA-A, B and C series. The study of a segregant family, showing a crossing over between the HLA-A and HLA-B loci, shows that the RM3 factor segregates on the side of the HLA-A locus. HL-A 27 was the only antigen with a significantly increased frequency. This increase concerned predominantly male patients in whom the disease developed at the beginning of puberty. In this group of sixteen boys, the polyarticular form was more frequent and tended to be associated with sacroliitis, while the rheumatoid factor was negative in all but one. The frequency of HL-A 27 in this group was 65.5%. The patients were also examined by the lymphotoxic test with anti H-2 alloimmune sera known to exhibit a high degree of association with some HL-A antigens. On JRA lymphocytes these associations were confirmed for HL-A 2 and anti-H-2f and for HL-A 27 and anti-H-2p. The strongest reactions were observed with lymphocytes from male patients around the age of puberty. Two different immunization doses of the conjugate were used, respectively, 2 x 40 mug and 2 x 200 mug in complete Freund's adjuvant or in alum adjuvant. There were two groups of control males, non-immunized and immunized with BSA in adjuvant. In the pooled immune sera, antibodies to testosterone were determined by radioimmunoassay; their titre ranged between 7 and 10. On histological sections of testes, inhibition of spermatogenesis (manifested by a sower frequency or even absence of tubules producing mature sperm, reduced frequency of tubular cells and their degenerative changes) was observed in almost all males immunized with the higher dose of the conjugate. In such animals, increased frequency of interstitial cells (except vascular elements) and enlarged nuclei of Leydig cells were found. No MLR reactions could be detected among guinea-pig families of a closed colony which had been bred to homozygosity for their GPLA antigens. By contrast, animals which differed in their I region showed strong MLR reactivity. Animals with identical I regions as judged by four serologically-defined specificities only, but differ with respect to B locus determinants, yield appreciably lower MLR responses. The female animals were found to have a higher concentration of both antigens and of HDL than the male animals. The amounts of antigens and of HDL were not obviously normally distributed, but appeared to be more bimodally distributed. The relationship between the amount of antigens and amount of HDL were tested for each animal. A significant positive correlation was found between the amount of R 67 antigen and HDL both for male and female animals in the Albino rabbit population, and for male animals in the Dutch rabbit population. A comparative study of rabbit and human serum high density lipoprotein.","authors":"A L Borresen","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Different classes of rabbit serum lipoprotein were prepared by ultracentrifugal flotation at densities 1-006, 1-063 and 1-21 g/ml. Agarose gel electrophoresis on rabbit whole serum and the serum fractions with different densities showed that this technique separates the different lipoprotein classes reasonably well. The electrophoretic mobility of the different lipoprotein classes of rabbit serum seems to be similar to that of the human lipoproteins, with the exception of alpha1-lipoprotein which had a greater mobility than human alpha1-lipoprotein. The chemical composition of rabbit high density lipoprotein (HDL)p was fairly similar to that of human HDL although the former seems to be richer in triglycerides. HDL was, after isolation by ultracentrifugal flotation at density 1-21, delipidated and submitted to gel filtration on Sephadex G-200 in 8 M urea. The major protein fraction of rabbit apo HDL corresponds in elution volume to that of the major fraction of human apo HDL, apoA-I. A protein fraction corresponding to human apoA-II does not seem to be present in rabbit HDL in demonstrable amounts. The two different genetic polymorphisms known in rabbit HDL were found to reside in two different polypeptides. This conclusion was reached in experiments where a quantitative radial immunodiffusion technique was employed.
