The detection and dosage of the toxic in the urine of patients have been carried out by the mean of a technic perfected by the same authors. {"title":"Factors influencing the indomethacin-induced intestinal lesions in the rat.","authors":"L Mariani","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The influence of various experimental conditions such as partial hepatectomy, administration of hepatotoxic drugs such as carbontetrachloride or ethionine, and starvation on the ulcerogenic activity of indomethacin at intestinal level has been studied in the rat. While lesions produced by oral administration of high doses of indomethacin are reduced by starvation, the incidence of ulcers was significantly increased by partial hepatectomy and by hepatotoxic drugs reaching the maximum of severity (intestinal perforation) in partially hepatectomized animals. These findings suggest that ulcerogenic activity of indomethacin is strictly correlated to the drug metabolising capacity of the liver.</p>","PeriodicalId":75837,"journal":{"name":"European journal of toxicology and environmental hygiene. The higher effect observed in vivo might be due to the activity itself of the chromone cycle which added up to that of the carbo-hydroxamic acid function. This latter alone appears in vitro. P. F. rats inhaling 0.55 ppm of acrolein, compared to 173 control rats, are reported. In what concerns the respiratory apparatus, this dose of acrolein affects its defense mechanisms, leading to a greater susceptibility to the airborne Salmonella enteritidis infection, compared to the control group. These phenomena disappear spontaneously when intoxication is prolonged more than three weeks. On the contrary, the general toxic action, manifesting itself through diminished weight growth and under-nourishment, compared to control group, lasts as long as intoxication and disappears only after acrolein inhalation has stopped. The sample is heated in a closed vessel at 80 degrees C and the head-space vapour injected in a gas chromatograph coupled to a mass spectrometer. Mass spectra are recorded every 4 seconds and stored by a Data Acquisition System. The reconstructed spectra enable very easy identification of the peaks of the chromatogram. The technique for extraction and chromatographic analysis is developed. The limit values for a correct determination are : 0,2 ng for nicotine and 1 ng for cotinine. The results show that the transit of nicotine and cotinine into the blood is very fast. They confirm the rapidity of the urinary excretion of nicotine, the slowness of that of cotinine, and the relation between the eliminated amounts and the urinary flow.</p>","PeriodicalId":75837,"journal":{"name":"European journal of toxicology and environmental hygiene. Every cosmetic substance may induce accidental intoxications. Most often, fluid cosmetics are absorbed either by children or by feeble-minded subjects. No relation between serum lipids and red cell lipids content have could be demonstrated. A relation is shown between morphology and lipids red cells content, different anatomopathologic states of liver and erythrocyte morphology. In steatosis red cells phospholipids (specially phosphatidyl choline) are increased. In cirrhosis red cells show an increased of both cholesterol and phospholipids content.
