Elimination of the easily mobilizable pool of lymphocytes was almost complete within approximately 6 days after commencing RECIB and was reflected by a reduction of approximately 50% in the numbers of paracortical lymphocytes. Blood lymphocyte counts were reduced to approximately ⅓ to ¼ of the pre-irradiation values. Thymectomized calves had lower pre-irradiation blood lymphocyte counts than non-operated controls, however, the reduction in the number of blood lymphocytes was proportionally similar. Continuation of RECIB beyond one week did not result in a further depletion of blood lymphocytes. The lymphocyte counts in the follicular cortex and in the medulla of lymph nodes were not significantly changed by the various procedures. RECIB of 8–16 days duration did not alter the number of germinal centers in non-thymectomized calves, however, in thymectomized animals treated with RECIB, the number of germinal centers was reduced.</p></div><div><p>Mit Hilfe der wiederholten extrakorporellen Bestrahlung des peripheren Blutes (RECIB, repetitive extracorporeal irradiation of the circulating blood) läßt sich ohne Gewebeschädigung und ohne Hämolyse der mobilisierbare Anteil der Lymphozyten zerstören. Diese Technik wurde — allein oder in Kombination mit Thymektomie bzw. Imuran-Behandlung — zu Vergleichszwecken bei Kälbern angewandt. Innerhalb der ersten sechs Tage nach Beginn der RECIB war fast der ganze leicht mobilisierbare Anteil der Lymphozyten ausgeschaltet. Dies zeigte sich u. a. in einem Absinken der Lymphozytenzahl auf ungefähr die Hälfte im Bereich der parakortikalen Zone der Lymphknoten sowie auf ein Drittel bis ein Viertel der Ausgangswerte im peripheren Blut. Thymektomierte Tiere wiesen schon vor der RECIB tiefere Blutlymphozytenwerte auf als die nicht operierten Kontrolltiere, zeigten aber eine proportional entsprechende Abnahme dieser Werte nach RECIB. Eine Fortsetzung der RECIB über eine Woche hinaus bewirkte keine weitere Verstärkung der Blutlymphopenie mehr. Die Lymphozytenzahl im Bereich des follikulären Kortex und der Medulla von Lymphknoten wurde durch die verschiedenen Behandlungsmethoden nicht nennenswert beeinflußt. Die Zahl der Keimzentren veränderte sich trotz 8–16tägiger RECIB bei den nicht thymektomierten Tieren nicht wesentlich, im Gegensatz zu thymektomierten, RECIB-behandelten Kälbern.</p></div>","PeriodicalId":75583,"journal":{"name":"Beitrage zur Pathologie","volume":"160 1","pages":"Pages 38-49"},"PeriodicalIF":0.0,"publicationDate":"1977-04-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1016/S0005-8165(77)80004-8","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"12057128","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The 8- and 10-year-old boys presented the characteristic clinical symptoms of the syndrome in their entirety. Both had a highly increased excretion of heparan sulfate in urine. The elder boy died and was autopsied and diagnosed as having Sanfilippo disease Typ A by a drastic reduction of heparan sulfate sulfamidase activity in organ extracts. Histochemically, highly water-soluble, sulfated acid mucopolysaccharides were demonstrated in liver, spleen, and cerebrum of the deceased child. Chemical analyses revealed a 12-fold increase of sulfated mucopolysaccharide in the patients liver and a 4.5-fold increase in the cerebrum when compared with normal controls. The sulfated polysaccharide consisted mainly of heparan sulfate, which was of low molecular size, heterogeneous in charge, and rich in its sulfamino hexose content. In addition, the gangliocytes of cerebrum and cerebellum had accumulated glycolipids. Electron microscopically the storage cells were overloaded with lysosomal residual bodies. The mitral valve was also involved in the storage process, which is a rare manifestation of the Sanfilippo syndrome. Acid mucopolysaccharides were deposited intracellularly as well as extracellularly in the mitral valve tissue. Polarisation microscopically there was found a change from normally positive to negative birefringence in the connective tissue ground substance of the mitral valve when containing stored mucopolysaccharides.</p></div>","PeriodicalId":75583,"journal":{"name":"Beitrage zur Pathologie","volume":"160 1","pages":"Pages 58-72"},"PeriodicalIF":0.0,"publicationDate":"1977-04-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1016/S0005-8165(77)80006-1","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"73823806","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
