Masutani","doi":"10.15761/JTS.1000370","DOIUrl":"https://doi.org/10.15761/JTS.1000370","url":null,"abstract":"Poly(ADP-ribose) polymerase (Parp) inhibitor 3-aminobenzamide (3-AB) pretreatment suppresses G1 arrest and enhances G2 arrest after gamma-irradiation in mouse embryonic fibroblast C3D2F1 3T3-a cells. 3-AB partially inhibits Waf1/Cip1/p21 and Mdm2 mRNA inductions, which are transcriptionally activated by p53, suggesting that poly(ADP-ribosylation) is involved in the downstream of p53 dependent signal transduction after gamma-irradiation in C3D2F1 3T3-a cells. In this study we further examined the involvement of poly(ADP-ribosylation) in cell cycle arrest. Effect on G1 arrest suppression was lost when 3-AB was added 6 hrs after irradiation. When C3D2F1 3T3-a cells were synchronized by serum starvation, and gamma-irradiated, the peak time of DNA synthesis was not changed but the ratio of DNA synthesis was decreased in gamma-irradiated cells, where 3-AB pretreatment slightly enhanced the decrease of this ratio. 3-AB decreased G1 arrest in mouse embryonic fibroblast Swiss3T3 cells dose-independent manner whereas G1 arrest was not affected at low doses in FM3A and NRF49F cells. To confirm the inhibitory effect of 3-AB on Parp activity, NAD level change was measured after gamma-irradiation. We observed NAD decrease induced by gamma-irradiation was prevented by the 4 mM 3-AB, suggesting sufficient inhibition of cellular Parp activity at 4 mM concentration. These results suggested that the effect of Parp inhibitor on G1 arrest after gamma-irradiation depends on cell phenotypes. out by incubation of the cells with medium containing 0.25% FBS for 44 hrs and released cells by changing the medium containing 10% FBS and 4 hrs later, cells were treated with chemicals. The aim of the study was to determine the contribution of impaired jugular veins to the status of the brain tissues insufficiently drained by extracranial veins. Thirty- eight mice were used for our experiments, 22 mice underwent to the bilateral ligation of the internal and external Jugular veins “(ligated group)”, 16 mice underwent to the same operative procedures and postoperative precautions but without jugular veins ligation ‘‘(sham-operated group)’’. Following the surgery, mice were monitored every day for the development of clinical symptoms. After 20 days and 3 months post-surgery, mice were sacrificed. Each brain was cut out sagittal to divide the two hemispheres. The samples were subjected to both histological and immunohistochemically staining using slides to define edema, inflammatory reaction, iron and fibrinogen deposit and demyelination. Of the 22 ligated group 7 showed a Grade 1 of clinical impairment. Histopathological assays showed the presence of brain edema and micro-hemorrhages. A significant higher number of positive inflammatory cells in the brains of ligated Group in comparison to sham group (p<0.05) was detected. No evidence of demyelination nor iron or fibrin deposition in Ligated group was demonstrated. In conclusion, jugular vein ligation altered cerebral hemodynamics without causing significant neurological symptoms. Lindley","doi":"10.15761/jts.1000460","DOIUrl":"https://doi.org/10.15761/jts.1000460","url":null,"abstract":"Background: The IMPACT implementation support platform is a measurement and feedback system specifically designed to scale evidence-based programs and practices (EBPPs) and support high-quality implementation. The purpose of this study was to evaluate use of the IMPACT software system in terms of its acceptability, appropriateness, feasibility, and likelihood of adoption. Methods : Seventy-eight school-based providers (across 49 schools) participated in this study. Demographic and background information was collected in the fall of 2019. Thereafter, providers delivered the Social Skills Group Intervention (S.S.GRIN) EBPP with students as usual and used IMPACT to enter and track progress and process data for their S.S.GRIN groups. In the spring of 2020, providers completed a series of ratings to evaluate their experience with IMPACT. Providers' usage of IMPACT during the study was also tracked. Results : Providers' ratings of IMPACT for each implementation outcome were significantly higher than average ( p < 0.001), with Satisfaction, Superior Innovation, and Interface Quality being especially positive. There was a significant interaction effect where ratings were higher for providers who reported disliking implementation data tracking for S.S.GRIN at the start of the study. Greater usage was significantly associated with higher ratings of providers’ capacity for tracking implementation data and a higher likelihood of recommending IMPACT to others. No significant differences in the patterns of results were found for demographic subgroups. Conclusion : IMPACT was seen as acceptable, feasible, and appropriate by school-based S.S.GRIN providers who reported likely continued use (adoption) of the innovation. IMPACT was particularly well received by those who started the study with negative impressions of implementation data tracking for S.S.GRIN. This study supports the potential utility and value for supporting ongoing implementation of school-based programs. Tsai","doi":"10.15761/JTS.1000321","DOIUrl":"https://doi.org/10.15761/JTS.1000321","url":null,"abstract":"Systemic Lupus Erythematosus (SLE) is a multifactorial systemic autoimmune disorder that can produce numerous abnormalities in cellular and humoral immune responses, including abnormal autoantigen and autoantibody production by dysregulated B cells. There are many known ligands of the AhR (aryl hydrocarbon receptor) in air pollutants, such as 6-formylindolo[3,2-b] Carbazole (FICZ) and 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). This study investigated the effect of FICZ/TCDD on the autoantigen/autoantibody and inflammatory cytokine production of B cells in Dp-allergic SLE. Dp2-induced autoantigen/autoantibody from B cell lines and PBMC derived from Dp-allergic SLE were used to evaluate the effects of FICZ/TCDD. Autoantigen/autoantibody and inflammatory cytokines were measured after cells were stimulated with FICZ/TCDD in conjuction with Dp2. The results showed that both FICZ/TCDD can activate AhR expression and induce the mRNA expression of IL-8. There were no effects on the production of IFN- α , IL-6, or autoantigen/autoantibody, except enolase-1. In the presence of Dp2 there were no augmentation effects of FICZ/TCDD on the expression of AhR or any of the autoantigens/autoantibodies from B cell lines. Although Dp2 could induce IL-8 production from PBMC derived from Dp-allergic SLE and non-SLE patients, there were no differences between the two groups and FICZ also showed no augmentation effects on IL-8 production. In conclusion, although FICZ/TCDD can induce AhR expression and IL-8 production from B cells derived from Dp-allergic SLE. There were no augmentation effects on Dp2-induced autoantigen/autoantibody or inflammatory cytokine production by FICZ/TCDD. Recently, platelet-rich plasma (PRP) injections have emerged as alternative cell-based therapies for the treatment of AGA. Its efficacy is strictly linked to the release of growth factors (GFs) via alpha-granules degranulation. Among their well-known activity, more recently, GFs are acquiring importance as regards their involvement in the regulation of hair growth cycle. Thanks to the advent of modern biotechnology synthetic polypeptides mimicking growth factors have been developed opening to new therapeutic approaches also in the dermatological field, including hair growth disorder. Objective: The aim of the present study was to evaluate the efficacy of a cosmetic product (TR-M-PRP plus) mimicking PRP composition by means of biomimetic peptides in subjects affected by AGA. Materials and methods: 60 AGA subjects were treated for three months end evaluated, at the end of the study and after one month of follow-up, as regards hair growth by evaluating total and anagen hair count, anagen/telogen ratio, % of miniaturization, Hair Mass Index (HMI), and hair shaft diameter. Results: TR-M-PRP plus treatment produced a statistically significant (p < 0.001) clinical improvement compared with PLACEBO in total and anagen hair counts. The treatment with TR-M-PRP plus resulted in a time- increased improvement in the anagen/telogen ratio, reduction of 5 of miniaturization and increasing of HMI and Hair shaft diameter. Material and methods: A retrospective analysis covered medical records of 33 pregnant women hospitalized in 1998-2017 operated on for pathologies affecting genital organs. The following parameters were assessed: age of patients, gestational age at the time of treatment, type of treatment, intraoperative histopathological diagnosis, and further course of pregnancy. Results: Gynecological pathologies were diagnosed in 33 out of 44 (75%) pregnant women undergoing surgical operations/procedures in the 20-year period. Those included one case of pre-invasive carcinoma of the uterine cervix in early pregnancy, two cases of mucous cystadenoma of borderline malignancy detected intraoperatively on histopathological examination. In 25 women, internal pathologies were operated on by laparotomy, and in two women by laparoscopy. Vaginal procedures and operations, i.e. ovarian and peri-salpingeal cyst punctures and cervical amputations were performed in 3 and 1 patient respectively. Serous, follicular, and corpus luteum ovarian cysts were the most numerous (11-33.3%) in the group of gynecological diseases. Operations for diseases and injuries of external genital organs were performed in 2 women. One of the women had a miscarriageon the 4th day after gynecological surgery. Another one left the Department with her pregnancy intact on the second day after surgery and did not report for further consultation. There were 3 (9.7%) premature births in 31 of 33 pregnant women operated on for gynecological diseases. Conclusions: Surgical treatment in pregnancy due to genital disorders should be individualized, taking into account clinical symptoms, gestational age, results of imaging and laboratory examinations, and the woman’s preferences. However, the molecules involved in osteoclastogenesis are still incompletely understood. The pre-osteoclast cell lines RAW264 and 264.7 are widely used to study osteoclast differentiation, however, their response to RANKL stimulation is relatively lower than that of primary pre-osteoclast. Here, we established novel RAW264 subclones. The subclones showed various responses to RANKL stimulation and their tartrate-resistant acid phosphatase (TRAP) activity was strongly correlated with the mRNA expression of two-pore channel subtype 2 (TPC2). TRAP activity in Clone15 and 10 was higher and lower when compared with that of RAW264.7 cells, respectively. We conclude that the subclones established in our study are useful tools to study osteoclast biology.
