The present study was undertaken to determine the time course of lesions produced in the lungs, spleen, liver, kidneys and bone marrow from 1 to 168 hours following a single (1.5mg) intravenous injection of P. aeruginosa LPS--1R (S-form), and its mutant strains, 557 (R-form) and 605 (more-R-form)--in rats. The lesions consisted of atelectasis, infiltration by polymorphonuclear leukocytes, intraalveolar hemorrhage and perivascular edema of the lung, hyperplasia of the white pulp of the spleen, necrosis of hepatic parenchyma, vacuolization of cells of the adrenal zone fasciculata, and hyperplasia of bone marrow. Immunoperoxidase stain for the R-form and the more-R forms of LPS of P. aeruginosa showed similar distribution kinetics within hepatocytes and Kupffer cells, while the S-form required 24 hours to become evident within hepatocytes. Double staining revealed that myc protein and nucleic acids are mutually exclusive and that myc protein is located in the nucleoplasm. Immunoelectron microscopic observation confirmed that the myc protein is part of the nuclear sap. To study the relationship between myc protein and hepatitis B virus (HBV) marker expression, we have utilized the cell line HepG2 transfected with the HBV genome as well as the corresponding tumors induced in nude mice. The ONMP Ras 0, perfectly complementary to the specific target region in normal c-Ha-ras, inhibits cell-free translation of p21 from a normal c-Ha-ras mRNA template in a dose-dependent manner. At 200uM Ras 0, p21 translation is inhibited by almost 90% in a rabbit reticulocyte lysate; at 100uM, p21 is reduced by over 60%. Ras I and Ras II contain, internally, a single and double base mismatch, respectively. The inhibitory activity of these ONMPs is reduced in proportion to the number of mismatches. When the target mRNA encodes the activated c-Ha-ras differing by a single nucleotide at the twelfth amino acid codon from normal c-Ha-ras, the magnitude of the inhibitory effect of Ras I increased significantly because Ras I is now perfectly complementary to its target mRNA. In turn, Ras 0, now only partially complementary, is considerably less effective at the same concentrations. Culture supernates of human umbilical vein endothelial cells (ECSN) at concentrations of 2.5 to 25% stimulated human foreskin fibroblast PGE2 production 6 to 180-fold. Following molecular sieve chromatography, peak activity eluted with an Mr of 14-18,000. In a standard IL-1 assay, neither ECSN or 14-18,000 Mr fractions possessing PGE2 stimulatory activity were able to stimulate murine thymocyte proliferation in response to PHA. Immunoperoxidase staining of endothelial cells demonstrated intracellular IL-1; after addition of exogenous IL-1 endothelial cells also stained for tumor necrosis factor (TNF). IL-1 and TNF are known to be synergistic in stimulating fibroblast PGE2 synthesis. Thus, elaboration of TNF by endothelial cells may allow detection of IL-1 in fibroblast PGE2 assays when the concentration of IL-1 is inadequate to stimulate thymocyte proliferation. Plastic embedding was used in this study to demonstrate platelet aggregates in the pulmonary vasculature of mice exposed to complement activation. The degree of platelet aggregation in the excised lungs was graded semiquantitatively in a total of 75 mice. Control Balb/c mice had a mean aggregation score of 0.16 while mice which received 0.3 ml zymosan-activated plasma (ZAP) intravenously (iv) had a score of 2.2. Injection of 0.3 or 0.4 ml of ZAP to which epsilon-aminocaproic acid was added prior to incubation with zymosan resulted in a score of 2.6 or 4.7 respectively. Balb/c (C5 sufficient) and AKR/J (C5 deficient) mice injected iv with 1 mg zymosan had scores of 2.9 and 3.2, respectively. Cobra venom factor (CVF) injected iv to Balb/c mice induced a dose-dependent aggregation. Serum TSH and CT values were determined in 50 male rats at ages 2, 6, 12, 18 and 24 months and 50 female rats at 18, 21 and 24 months. The animals were sacrificed at the end of the experiment to evaluate thyroid C cell pathology. Male rats showed an increase in the serum TSH values at 6 and 12 months of age, a decline at 18 months and a significant increase from 18 to 24 months of age. In female rats, a significant increase in serum TSH concentration was also noted from 18 to 24 months of age. In both sexes, 24 month old animals with thyroid C cell hyperplasia (CH+ group) had a significantly higher level of serum TSH as compared to animals with normal distribution of C cells (CH- group). Concentration of serum CT showed a progressive increase with age in both sexes. Male rats with thyroid C cell pathology had significantly higher levels of serum CT at 24 months of age as compared to rats with normal C cell distribution. In female rats, however, serum CT concentrations in two groups were not statistically significant. One group of experimental animals (N = 31) received a single intraperitoneal injection of 0.1 mg/kg of S enteritidis endotoxin. The second experimental group (N = 34) received multiple intraperitoneal injections of 5 to 10 mg/kg of E coli endotoxin at 15 minute intervals over a period of 75 minutes. Within four hours of injections, histopathologic evaluation of all the tissues revealed interstitial edema, widening of blood capillary lumina, accumulation of polymorphonuclear leukocytes and platelets in capillary lumina and marked swelling, vacuolization and focal desquamation of endothelial cells in microvasculature. Hepatocytes showed loss of glycogen, vacuolization and degeneration in the centro-portal region. Islet cells of pancreas also revealed swelling and vacuolization. In the small intestine, hemorrhagic pools of blood were frequently seen in lamina propria and the apical portion of villi showed degeneration and breakdown. Lungs showed focal hemorrhage, collapse of alveolar architecture and swelling of endothelium in larger artereoles. The ability of interferon to produce a similar effect in vivo was tested in rhesus monkey placentas which are known to contain C-type virus particles. Samples of placenta from 6 monkeys treated with recombinant leukocyte A interferon (25 x 10(6) units/kg) for 7 to 21 days and from 6 control monkeys were examined in the electron microscope. Budding and immature C-type virus particles were present in every placenta examined from both interferon-treated and control monkeys. Mature particles were found in placentas from 3 of 6 monkeys in both treated and control groups. As we had previously demonstrated the efficacy of demethylating drug in activating the formation of retroviral particles, we used 5-azacytidine to activate endogenous retrovirus expression. Proceeding to a more detailed survey we were able to observe various types of retrovirions. All of them were present in the different activated cell lines. They were: intracisternal R-type particles (IRP), intracisternal A-type particles (IAP), intracytoplasmic A-type particle (ICAP), intramitochondrial A-type particle (IMAP) and M-432 virus-like particles. Some of these particles have never been described before in Syrian hamster cells.
