A single administration of hydrogen sulphide donor as drops into cavity of the eye has caused the hypotensive effect, which was pretty weak in the normotensive eyes and pronounced in the eyes with ocular hypertension. Long-term administration ofNaHS solution by the scheme, during administration of adrenaline as a model inductor, has largely prevented IOP rise. It was shown that ginkgolic acid, a constituent of lipophylic Ginkgo biloba extract, has numerous biological activities. In the present study we have focused on the features of ginkgolic acid action on αl and α2 glycine receptors that make part of the inhibitory system of the brain. Using whole-cell configuration of patch-clamp recording we analysed effects of ginkgolic acid on different subunits of glycine receptors. Experiments were performed on cultured Chinese hamster ovary cells (CHO cells), transfected with αl and α2 glycine receptor subunits. Ionic currents were induced by the fast application of different glycine concentrations. After 20-40 sec of pre-treatment with ginkgolic acid (25μM) currents mediated by al glycine receptors reversibly increased from 364±49 pA, (n=34) to 846±134 pA, (n=34). EC(50) for glycine has changed from 36±6 μM (control) to 17±2 μM. In contrast, the application of ginkgolic acid on glycine receptors formed by α2 subunits did not provoke potentiation. Our results demonstrate that ginkgolic acid is a subunit specific modulator of glycine receptors. The semiquantitative method of damage rate of microcirculation channel and liver parenchyma was used, leukocyte liver infiltration and cellular composition of infiltrates were investigated also. Primary liver reaction to the Con-A was the inflammatory changes in the vascular bed, followed by disturbances in the parenchyma.The sufficient increasing of leukocyte migration to the liver was revealed. Besides, the neutrophile infiltration was increased first with a maximum at 6 hours of the experiment (63,9 ±4,6%, p<0,001 to the control level) ,and then the lymphocyte infiltration was increased with creation of manycellular lymphocytemacrophage infiltrates (62% at 48 hours comparing to 6 hours of experiment) and sufficient quantity of plasma cells population (4,9%, p<0,05 comparing to 6 hours of experiment). The obtained data gives the base to suggest that the elevated infiltration of liver tissue by leukocytes, particularly by lymphocytes and monocytes, together with necrotic death increasing creats the conditions for effective intracellular interaction and immune response to autoantigenes. We found an increase serum level of EMAP-II and decrease of endothelium-dependent dilatation in type 1 diabetes. It was significant correlation between EMAP-II and HbAc1, blood glucose, total cholesterol, LDL, triglycerides and inverse correlation between EMAP-II and HDL, endotheliumdependent dilatation. The development of obesity is tightly connected with dysregulation of intrinsic gene expression mechanisms controlling majority of metabolic processes, which are essential for regulation many physiological functions, including insulin sensitivity, cellular proliferation and angiogenesis. Our objective was to evaluate if expression of angiogenesis related genes VEGF-A, CYR61, PDGFC, FGF1, FGF2, FGFR2, FGFRL1, E2F8, BAI2, HIF1A, and EPAS1 at mRNA level in adipose tissue could participate in the development of obesity and metabolic complications. We have shown that expression level of VEGF-A, PDGFC, FGF2, and FGFRL1 genes is decreased in adipose tissue of obese men with normal glucose tolerance (NGT) versus a group of control subjects. At the same time, in this group of obese individuals a significant up-regulation of CYR61, FGF1, FGFR2, E2F8, BAI2, and HIF1A gene expressions was observed. Impaired glucose tolerance (IGT) in obese patients associates with down-regulation of CYR61 and FGFR2 mRNA and up-regulations of E2F8, FGF1, FGF2, VEGF-A and its splice variant 189 mRNA expressions in adipose tissue versus obese (NGT) individuals. Thus, our data demonstrate that the expression of almost all studied genes is affected in subcutaneous adipose tissue of obese individuals with NGT and that glucose intolerance is associated with gene-specific changes in the expression of E2F8, FGF1, FGF2, VEGF-A, CYR61 and FGFR2 mRNAs. It was shown that animals with alcohol intoxication under of the admission of the main substrate NOS - L-arginine activity of antioxidant protection enzymes was increased in twice on the fond of TBA-positive products decrease contents. Established in hemolisate red blood cells in rats with alcohol inrotoxication value of total NOS activity decreases by 65% compared to control. Not selective inhibitor Nto-nitro-L-arginine methyl ester (L-NAME), which is a structural analog of L-arginine, reduced output level of total NOS activity by 23.4% in the control and 25% under conditions of pathology. The consumption of rats L-arginine NOS total activity increased in the two study groups. It was established that the development of obesity in rats was accompanied by violation of homeostasis in the glutathione-dependent system of blood serum. It has been shown that animals on high-calorie diet-along 14 weeks had a decrease (1,2) in GSH concentration, a decline (1,7) in GP activity and the increase (1,7) of GT activity compared with control values. {"title":"Modulation of the hippocampal propensity to non- synaptic epileptiform synchronization in low-calcium model of epilepsy.","authors":"O S Zapukhliak, O V Netsyk, O S Rasulova, D S Isaev","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The CA3 and CAI regions are the main stages of the \"three-synaptic pathway\", which plays a role in the generation of hyper-synchronous events in the hippocampus. Under certain experimental conditions, this brain structure might support pathological epileptiform synchronization that is independent of active chemical synaptic transmission. In present work, we estimated the conditions that would facilitate non- synaptic synchronization of the hippocampus. Non-synaptic epileptiform activity was induced in hippocampal slices by the omission calcium ions from the extracellular milieu. The propensity of hippocampal regions to nonsynaptic interactions was estimated by measuring the delay time neededfor the development of low-Ca²⁺ discharges in the CA3 and CAI. Next, an increase of neuronal excitability was induced by the pre- incubation ofhippocampal slices in 4-aminopyridine (4-AP) and by the reduction ofextracellular osmolarity. Pre-incubation of hippbcampal slices with 4-AP under normal osmotic conditions resulted in decreased latency for non-synaptic discharges in the CA3, but not in the CAl. Exogenous melatonin (Unipharm Inc., USA) was administered to experimental group of animals daily'at a dose 5 mg/kg. The duration of the experiment was 28 days. It was revealed that the exocrine part of the.pancreas responds differently to the effects of melatonin at different times of the year. Thus, after administration of melatonin in the spring increase of the size of acinus, the height of the epithelium (by 7 %), area exocrinocytes (by 58 %), of their nucleus (by 20 %) and cytoplasm (69 %), the amount of nucleoli in cells (18 %), reduction the amount of connective tissue elements. Melatonin introduction in the autumn decrease in the size of acinus, height and area of exocrinocytes, growth the number of exocrinocytes in the acinus, nucleoli and width layers interlobular connective tissue in the gland. This may indicate that melatonin increases in the spring of the synthetic activity of the exocrine pancreas, whereas in the autumn (for the majority of the morphometric parameters) - somewhat reduces its functional state. The administration of melatonin in the spring (mostly) and in the autumn periods increased the functional activity of the endocrine pancreas.
