{"title":"The overlapping effects of thyrotropin and gonadotropins on chick embryo gonads in vitro.","authors":"M A Shahin,&nbsp;O Török,&nbsp;G Csaba","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Pieces of 12- and 15-day-old chick embryo testes and ovaries were cultured in vitro in the presence of thyrotropin (TSH), gonadotropins (FSH + LH) and adrenocorticotropin (ACTH) for different periods. All the explants of treated gonads differentiated into typical testes or ovaries according to their genetic sex. The gonads of 12-and 15-day-old chick embryos showed a good response to both thyrotropic and gonadotropic stimulation. On the other hand, they did not respond to adrenocorticotropic stimulation. Fifteen-day-old chick embryo testes were grown in tissue culture in the presence of the said hormones. Gonadotropins and TSH enhanced the growth and migration of testicular cells as compared with the control or ACTH treated group. In addition, they maintained the germ cells on the upper surface of epithelial cells. {"title":"Cartilage differentiation in micro-mass cultures of chicken limb buds.","authors":"C Hadházy,&nbsp;M B Lázló,&nbsp;K S Kostenszky","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In micro-mass cultures of stage 23-24 chicken embryos the expression of cartilage phenotype was density dependent and it always took place practically to the same extent at the density of 2 X 10(5) cells per culture. The cells formed aggregates on day 1 and cartilage nodules appeared on day 2. By day 3, 17% of the culture surface consisted of areas showing cartilage differentiation and these regions represented more than 50% on day 6. By day 14, the centre of th cultures contained a mass of cartilage. The progression of cartilage differentiation was indicated by the increasing amount of toluidine blue fixed by the cultures. By day 14 the DNA and protein contents increased 8.7 fold and 16.7 fold, respectively. Uronic acid and OH-proline were detected from the 2nd day. From day 3 to 14, the microgram uronic acid and microgram OH-proline per microgram DNA gradually increased 25 fold and 14 fold, respectively. A decreased density of the inoculum reduced the probability of cartilage formation and it failed to occur at the density of 1.25 X 10(4) cells per culture. Stage 19-20 limb cultures with identical density had the same morphological characteristics. Aggregates appeared but nodules failed to do so in cell cultures of stage 28 limb bud soft tissue region. Enzyme activity was higher in the preexisting, differentiated laminar bone tissue of young animals than in that of old animals, and higher in newly formed woven bone than in the preexisting lamellar one and higher in chondroid tissue and in epiphyseal cartilage than in the woven bone and chondroid tissue. Degeneration of numerous nerve processes could be observed after capsaicin treatment. Some of these nerve fibres were found in pre- and in postsynaptic situation. A few cell somata were also severely damaged after capsaicin treatment. No such degeneration was observed in the control animals. In some dogs the myocardium was adapted to ischaemia by progressive coronary occlusion of 1-5 min followed by restoration of circulation during 5 min. In other dogs adaption was followed by 20 to 35 min ischaemia. The animals were sacrificed immediately or after 2-10 days. Transient ischaemia produced less severe alterations then abrupt coronary obstruction. Adaptation followed by 20 and 35 min ischaemia induced foci that undergo cytolysis and scarring of maximum intensity on the 8th day. This response was elicited by all the three treatments but was assessed as mild in \"C\", moderate in \"MPA\" and severe in \"CA\" groups. On the basis of present work it is concluded that inhibition of spermatogenesis occurred even with low dose schedule of the two antiandrogens and MPA. In the normal state the adventitia of the bladder base contained simple sensory corpuscles, Pacinian corpuscles and free nerve endings. In the bladder wall the afferent nerve endings appeared to be free nerve endings without terminal modification. After extirpation of spinal ganglia, some of the nerve fibres degenerated and were located mostly in the trigone and the ventral neck regions of the bladder. The yellow fluorescent fibres were found in the circular muscle layer and the green ones in the longitudinal layer and between the two muscle layers. Strongly fluorescent enterochromaffin cells were observed in all parts of the alimentary tube in the epithelium of the mucosa. Yellow fluorescent nerve fibres and enterochromaffin cells showed the peak emission at 520-525 nm, which corresponds to that of serotonin fluorophore in a model system. Green fluorescent nerve fibres had their emission maximum at 460-470 nm, which corresponds to that of dopamine and noradrenaline. The HBV surface and core components were found to be more resistant to proteolytic enzymes and phospholipase than the other cellular components, and only combined digestion was effective. With preferential RNP and specific DNP staining the accumulation of granules 10-20 nm and 20-40 nm in diameter could be observed in the liver cell nuclei. The cytochemical-morphological polymorphism of the latter was well detectable. Part of the granules corresponded to accumulated perichromatin granules.
