Additionally, the study aimed to determine the factors that influence these views. The study also examined the barriers and encouragements, according to physicians, in the use of compounded medicines. The original questionnaire was developed specifically for this study. Included sample consisted of Polish-speaking physicians whose main specialization was gynaecology, gastroenterology, dermatology, family medicine, internal medicine, otorhinolaryngology, paediatric otorhinolaryngology, paediatrics. Answers were gathered using computer-assisted telephone interview. The majority of surveyed physicians believed that every Polish patient receives treatment tailored to their personal needs (68,7%) and agreed that compounded medicines are a way to facilitate individualization of pharmacotherapy (79,3%). Frequency of prescribing of magistral formulations, as well as views on quality of compounded medicines, ease of prescribing, and filling a prescription vary between specializations and places of practice. The problem that negatively affects the practice of the largest group of respondents was complicated and frequently changing legal regulations (63.3%), while the least frequently indicated problems were concerns about the quality of preparation (16.7%). Despite its numerous important physiological functions, the practical application of CoQ10 is limited due to its high molecular weight, poor water solubility, photolability, chemical instability, and low bioavailability. To overcome these challenges, we developed a solid dispersion formulation of protamine loaded CoQ10 (CoQ10/Pr) using an efficient mechanochemical technology without solvents. Protamine was used as carrier and poloxamer 188 as solubilizer, while butylated hydroxytoluene (BHT) and micro powder silica gel were utilized as stabilizers and lubricants. The optimal ball milling condition to prepare CoQ10/Pr was at 100 r/min for 4 hours. The treated CoQ10/Pr has exhibited rapid-release properties, with significantly enhanced water solubility by 392,000 times, and bioavailability by 0.68 times. Furthermore, the treated CoQ10/Pr became resistant to moisture, high temperature and strong light, which makes it suitable for long-term storage. The molecular docking suggested that the solubilization mechanism of CoQ10 was closely associated with the encapsulation and complexation of protamine, as well as the formation of an intermolecular complex through hydrogen bond between CoQ10 and arginine residues on protamine. In summary, the CoQ10 solid dispersion formulation enjoys advantages such as excellent water solubility, stability, bioavailability, and low cost. This formulation also meets the stringent quality requirements of products in the food, cosmetic, and health industries. To date, a simultaneous HPLC method for active sub-stances and impurities of this product has not been reported. Acetonitrile and buffer (Octane-1-Sulfonic Acid Sodium Salt and Potassium Dihydrogen Phosphate) were used as a mobile phase. The separation was achieved using Phenomenex Phenyl-hexyl (150 cm × 4.6 mm, 5 µm) column at 1 mL/min flow rate in gradient elution mode. The chromatograms were monitored at 240 nm. The validation parameters such as linearity, limit of quantification, limit of detection, accuracy, precision, robustness and specificity were investigated for the nasal spray. Preparations recommended for daily care of the dry skin are emollients. Emollient preparations demonstrate a moisturizing effect, they soften and smoothen the skin, keep water in the epidermis, supplement lipids of skin upper layers and protect from external harmful agents. The purpose of this study was to develop and characterize emollients containing natural emulsifier Olivem 1000 as topical delivery systems for urea or sodium hialuronate. Physicochemical and mechanical properties of designed formulations were evaluated. Moreover, ex vivo adhesion study on the hairless mice skin as a model of adhesive layer was carried out and in vivo preliminary assessment of transepidermal water loss (TEWL) was performed. Prepared emollient formulations were characterized by smooth consistency, they were easily spreadable on the skin surface and showed no phase separation. Formulations containing xanthan gum as a thickening agent were characterized by higher viscosity, hardness, cohesiveness and adhesiveness values. Prepared emollients showed thixotropic properties and they were classified as non-Newtonian shear thinning systems. All prepared emollients demonstrated bioadhesive properties. The analysis of TEWL showed that the greatest decrease in the TEWL value was noted in the case of emollient containing sodium hyaluronate which was applied to the person with the skin described as “the skin in a very poor condition”. Expectations also concerned the effectiveness of vaccines in inducing a strong immune response against the SARS-CoV-2 virus and the length of post-vaccination immunity. This topic seems to be especially important in the case of immunocompromised people such as those living with HIV (PLWH). The aim of the study was to examine the concentration of anti-SARS-CoV-2 IgG antibodies against the S1 subunit of the virus’spike protein as a marker of humoral response and the concentration of interferon-gamma (IFN-γ) as an indicator of cellular response in PLWH undergoing combination antiretroviral therapy (cART) and uninfected people (control group) several months after the COVID-19 booster. Concentrations of selected enzymes from the group of sirtuins (SIRT1,-3,-6) that participate in the course of HIV infection were also examined. Concentrations of IgG and IFN-γ were statistically significantly lower in PLWH compared to the control group. Correlations between IgG and IFN-γ concentrations were found both in PLWH and in the control group. Only the concentration of SIRT-6 was significantly lower in PLWH compared to the control group. Vaccination for COVID-19 enabled good long-term humoral and cellular immunity in both HIV positive and uninfected people. The CD4/CD8 ratio and T CD8+ T lymphocytes count played a role in the long-term immune response to booster vaccination. Paśko","doi":"10.32383/appdr/176361","DOIUrl":"https://doi.org/10.32383/appdr/176361","url":null,"abstract":"The aim of this study was to evaluate the consumption frequency of the selected examples of goitrogenic foods in four different groups of people in Poland, who used traditional, vegetarian, vegan and pescovegetarian diets. The validated consumption frequency questionnaire of the examples of goitrogenic foods in four groups of people (n=365) on afore-mentioned diets, was performed in Poland in 2022. The statistical correspondence analysis model (CAM) was used to quantify and reveal potential relations between dietary habits and thyroid dysfunction, family history of thyroid disease, use of thyroid drugs (L-thyroxine, propylthiouracil, thiamazole), or iodine dietary supplements/drugs. Statistical analyses revealed an increased odds ratio more than 3, 8, and 6 times higher for tofu; plant-based meat; and soy beverages, respectively, by vegetarian’s vs traditional diet. Respondents with diagnosed hypothyroidism consumed soy, flax products, but avoided iodine-rich products. All respondents on traditional diet used iodine-rich products while 22% vegetarian and 23% vegan excluded them. CAM analysis did not show any relation between brassica vegetables and thyroid dysfunction. Despite the results obtained using food questionnaire were not combined with biochemicals data, some tendencies in food behavior among Polish vegans, vegetarians, and pescovegetarians appeared. Apart from some interesting correlations and tendencies, revealed in our study, further in-depth research, combined with biochemical analysis, will allow to better understand, and explain these observations.
