Lv","doi":"10.3724/SP.J.1008.2015.01352","DOIUrl":"https://doi.org/10.3724/SP.J.1008.2015.01352","url":null,"abstract":"Objective To determine the contents of seven main flavonoids in Epimediumof different places of origin by high performance liquid chromatography(HPLC).Methods The HPLC condition was as follows:SHISEIDO MG-C18column(3.0mm×100 mm,3.0μm);the mobile phase consisted of acetonitrile and water containing 0.1% formic acid was used as gradient elute;the gradient of acetonitrile was 25%(0-10min),25%-40%(10-12min),40%-45%(12-22min),45%-75%(22-25min)and 75%(25-30 min);flow rate was 0.6 mL/min;UV detection wavelength was set at 270 nm;column temperature was 25℃;and injection volume was 5μL.The sample was ultrasonically extracted with 70% ethanol solution.Results The seven flavonoids,including epimedin A,epimedin B,epimedin C,icariin,icariside Ⅰ,icariside Ⅱ and anhydroicaritin were separated at baseline within 30 min.The standard curves had good linearity(r=0.999 9),with the RSD%of intraday and interday precision being both less than 2.0%,the recovery being 98%-102%,and the RSD% of stability and repeatability also being less than 2.0%.The results showed that the method could meet the requirements of method validation.The contents of seven flavonoids in reference crude meterial and Herba Epimedii from Liaoning,Gansu and Hubei province were determined.Conclusion The present method is rapid and simple,and can be used for quality control of Epimedium,which may lay a foundation for studying the pharmacokinetics and tissue distribution of the flavonoids of Epimedium.","PeriodicalId":6893,"journal":{"name":"Academic Journal of Second Military Medical University","volume":"36 1","pages":"1352"},"PeriodicalIF":0.0,"publicationDate":"2015-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"69859890","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

{"title":"Modified microneedle roller enhancing percutaneous absorption of triamcinolone acetonide by human hyperplastic scar skin","authors":"Ya Li, Juan-juan Zhang, Li-Chao Zhang, Quan‐gang Zhu, Hai-jin Yu, Zhijuan Wu","doi":"10.3724/SP.J.1008.2015.00058","DOIUrl":"https://doi.org/10.3724/SP.J.1008.2015.00058","url":null,"abstract":"Objective To evaluate the role of modified microneedle roller in promoting permeation of triamcinolone acetonide across human hyperplastic scar skin.Methods The morphology of human hyperplastic scar skin was visualized by methylene blue staining and H-E staining after the insertion by microneedles of different lengths.The content of triamcinolone acetonide was determined by high performance liquid chromatography(HPLC).The in vivo absorption characteristics across human hyperplastic scar skin were investigated by elution method and the homogenization method at 1hand 12 hafter drug application.Skin irritation was assessed by transepidermal water-loss(TEWL)measurement and laser Doppler flowmetry.Results The preferred microneedle length of 500μm effectively penetrated through the stratum corneum,and the distribution of the microchannels was homogeneous.It was found that 27.42% and 60.64% of the drug administered entered the skin at 1h and 12 h,respectively.The drug accumulation in the skin at 12 hdecreased to 45.98% of that at 1hafter intracutaneous injection,while the accumulation increased to 2.73-fold and 4.18-fold in the microneedle group and direct application of triamcinolone acetonide cream group,respectively,with the content of microneedle group being 3.56-fold that of the direct cream application group(P0.05).Triamcinolone acetonide distribution was not homogeneous at 12 hafter intracutaneous injection,with the level being(4.83±5.51)μg,while the distribution in the microneedle group was homogenous,with the level being(0.93±0.14)μg.TEWL results showed no significant difference in skin irritation between microneedle and intracutaneous injection group;however,laser Doppler flowmetry showed that the skin irritation of intracutaneous injection group was 8.40-fold that of microneedle treatment group.Conclusion Modified microneedle roller treatment followed by cream application,compared with intracutaneous injection,can yield more homogeneous distribution and less skin irritation in human hyperplastic scar skin,but its efficacy for scar treatment remains to be further verified.","PeriodicalId":6893,"journal":{"name":"海军军医大学学报","volume":"36 1","pages":"58"},"PeriodicalIF":0.0,"publicationDate":"2015-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"69842542","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

{"title":"Effect of silk fibroin on degradation and in vivo biocompatibility of poly (L-lactic-co-ε-caprolactone) electronspun nanofibrous scaffolds","authors":"Zixuan Cheng, Cheng-wei Yang, Li Liu, Yang Yang, Guoying Deng, Xiao-Jian Ye","doi":"10.3724/SP.J.1008.2015.00500","DOIUrl":"https://doi.org/10.3724/SP.J.1008.2015.00500","url":null,"abstract":"Objective To investigate the effect of silk fibroin(SF) on degradation and biocompatibility of poly(L-lactic acidco-e-caprolactone)(P[LLA-CL]) in vivo.Methods The scaffolds of P(LLA-CL)(w / w = 1 ∶ 1) blended with 25% of SF(SF / P[LLA-CL]) and P(LLA-CL) were prepared by electrospinning.Both kinds of scaffolds were subcutaneously implanted in 45 6-monthold rats for up to 6 months to evaluate their degradation and biocompatibility characteristics.Results Pathological sections showed P(LLA-CL) scaffold become swollen and began to separate into different layers after 3 months,and then become broken after 6 months;while SF / P(LLA-CL) scaffold largely maintained its structure after 6 months.Immunohistochemical staining showed a large number of macrophages on the surface and in P(LLA-CL) scaffolds 1 month after implantation,and they could still be found 3 months after implantation,accompanied by foreign body giant cells; while no obvious macrophages or foreign body giant cells were found in SF / P(LLA-CL) scaffolds at different time points.Examination of inflammatory gene expression showed that TNF-α and IL-10 expression in P(LLA-CL) scaffolds was significantly higher than that in SF / P(LLA-CL) scaffolds 1 week after implantation(P 0.05),the same was also true for TNF-α,IL-1β and IL-10 expression 1 month after implantation(P 0.05),for TNF-α and IL-10 expression 2 months after implantation(P 0.05),for TGF-β expression 3 months after implantation(P 0.05),and for IL-1β and TGF-β expression 6months after implantation(P 0.05).Conclusion SF incorporation can delay degradation,reduce inflammation,and improve the biocompatibility of P(LLA-CL) scaffolds,which may provide reference for scaffold design in tissue engineering.","PeriodicalId":6893,"journal":{"name":"海军军医大学学报","volume":"36 1","pages":"500"},"PeriodicalIF":0.0,"publicationDate":"2015-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"69847742","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
